    Notes: Summary. The use of prostaglandin synthetase inhibitors (indomethacin and ibuprofen) for 〉72 h in women with preterm labour (n=67) was significantly (P〈0.001) associated with ultrasoundrecorded oligohydramnios compared with the use of ritodrine or magnesium sulphate (control group, n=67). Oligohydramnios developed in 26 of 37 women (70%) treated with indomethacin and 8 of 30 women treated with ibuprofen (27%) (P〈0.01). Oligohydramnios developed in only two control subjects and one of these pregnancies was associated with intrauterine growth retardation. All 34 cases of oligohydramnios in the treatment group resolved after stopping medication, whereas the two in the control group did not resolve. No instances of renal failure, premature closure of the ductus arteriosus, pulmonary hypertension, or bleeding disorders were noted in any of the treated or control infants.

    Notes: Abstract  Constitutive, high-level expression of the potentially therapeutic WAF-1/CIP-1/SDI-1 gene is incompatible with cell growth. A promoter conversion retroviral vector carrying the WAF-1/CIP-1/SDI-1 gene under the transcriptional control of the glucocorticoid inducible promoter of mouse mammary tumor virus was used to infect human bladder carcinoma or feline kidney cells. Reduced cell growth due to a greater proportion of cells being in the G0/G1 phase of the cell cycle was observed when WAF-1/CIP-1/SDI-1 expression was activated by addition of glucocorticoid hormone. This system demonstrates the potential long-term therapeutic use of WAF-1/CIP-1/SDI-1 delivered by retroviral vectors for inhibiting the growth of rapidly proliferating cells. Moreover, the conditional expression of genes such as WAF-1/CIP-1/SDI-1 from such retroviral vectors may facilitate analysis of their function.

    Notes: Abstract Maërl, comprised of shallow, subtidal deposits of calcareous red algae belonging to the family Corallinaceae, is used in agriculture, primarily to increase soil pH. Its use has been strongly criticised because of its high price compared to limestone. The chemical and physical characteristics of maërl and limestone are compared to determine whether these indicate if any benefit is to be gained with the use of the former. Analysis by inductively coupled plasma emission spectrophotometry shows that the proportion of magnesium in maërl is about ten times higher than that in the limestone samples tested. The levels of iron, boron and especially strontium are noticeably higher in the calcified seaweed than in the limestone, although the manganese contents are lower. Scanning electron microscopy shows that the surface characteristics of maërl and limestone are similar but, in section, maërl is considerably more porous because of its cellular structure. Atomic force microscopy revealed minor differences in fine structure between the two. The differences between maërl and limestone would not appear to compensate for the considerably higher costs involved with the utilization of the former material.

    Notes: Abstract The fusion of normal human fibroblasts with SV40-transformed human fibroblasts resulted in hybrid clones, 85% of which exhibited a finite in vitro life-span. Foci of rapidly dividing cells appeared in 15% of the hybrid clones. The cells within these foci repopulated the culture and could then be subcultured through more than 100 population doublings. One or two foci of dividing cells occurred per culture of 105 or more cells. The change to an indefinite life-span was, therefore, a rare event. All hybrid clones, including those that exhibited a finite in vitro life-span, expressed viral T antigen. Thus, even though viral DNA was present and being expressed in all hybrid clones, the senescent phenotype was dominant in these hybrids.

    Notes: Abstract A biochemical selection system was used to isolate hybrids after fusion of human diploid fibroblast clones of varying proliferative potential. The distribution of proliferative potentials of the hybrids resembled that of the parent with smaller proliferative potential. Therefore, the phenotype of limited division was dominant in hybrids. This dominance was associated with cells that could achieve seven or less divisions present in the parent populations. Terminally nondividing cells present in clones near the end of their proliferative potential differed from those present in clones that had ceased division (senescent), in that a higher percentage could be induced to divide at least once following fusion with a cell capable of division.

    Notes: In the past, it has been difficult to grow human diploid fibroblast cells at clonal densities. Newly devised cell culture media and rigorously controlled environmental conditions have greatly increased the ease with which such cells can be cloned. The present work was undertaken to determine whether, under appropriate conditions, diploid fibroblast cells from human embryonic lung, grow as well at clonal densities as in mass culture. The parameters studied were: (1) population doubling time, (2) in vitro proliferative capacity, (3) attachment, (4) percentage of non-dividing cells. In all cases essentially the same results were obtained for cultures at clonal densities and mass cultures. These results indicate that the behavior of these types of cells in clonal culture can be used to infer the behavior of individual cells and clones within a mass culture.

    Notes: The effect of culture conditions on calf dorsal aorta endothelial cells was studied. Population doubling time varied as a function of the cell seeding density, growth medium, serum supplement, and concentration of fibroblast growth factor (FGF). The shortest population doubling time was found for cells (population doubling level 0-30) grown in Eagle's Minimal Essential Medium (MEM) supplemented with 10% fetal bovine serum (FBS) and 100 ng/ml FGF. The stimulatory effect of FGF on bovine endothelial cell proliferation was dependent on cell inoculation density. FGF significantly increased cell division rate at cell inocula less than 1 ± 104 cells/cm2 but not at higher densities. The population doubling time and cell size increased as the mass culture population doubling level increased. The replicative lifespans of bovine endothelial cells grown in medium supplemented with 20% FBS were 10-15% greater than parallel cultures supplemented with 10% FBS. Cultures grown in medium supplemented with 10% FBS and 50 ng/ml FGF showed a 50% increase in replicative lifespan compared to cultures grown in medium supplemented with 10% FBS alone. When FGF was used the increase in the number of doublings was a function of the length of time the cells were grown in the presence of FGF. This report extends comparable observations on the in vitro aging of human diploid fibroblasts to bovine endothelial cells.

    Notes: Angiotensin-converting enzyme (EC 3.4.15.1) is a carboxyterminal dipeptidyl peptidase. The enzyme catalyzes the conversion of the decapeptide angiotensin I to the octapeptide angiotensin II. In addition, the enzyme catabolizes bradykinin. Because of these actions, the enzyme is of pivotal importance in blood pressure homeostasis. Numerous investigators have demonstrated the presence of the enzyme in association with endothelial cells but relatively little is known concerning the factors controlling the expression of enzyme activity by endothelial cells in culture. We have demonstrated that endothelial cells in culture do not express significant amounts of enzyme activity until several days after growth ceases due to high cell density. This is important because it demonstrates a change in function with stage of growth in culture and a possible difference in functional capabilities between nondividing endothelial cells and cells that are dividing in response to injury. Since density-dependent expression of differentiated traits does not appear to be unique to endothelial cells an understanding of the mechanisms underlying this phenomenon may provide a general explanation for the expression of differentiated traits by cultured cells.
