    Notes: Audio frequency electrical conductivity and relaxation studies have been carried out on Parel 58 elastomer and Parel 58 elastomer complexed with a variety of lithium salts. The measurements have been carried out in vacuum over the temperature range 5–380 K and at pressures up to 0.65 GPa over the temperature range 230–380 K. Both the electrical conductivity for the complexed material and the electrical relaxation time associated with the α relaxation in the uncomplexed material exhibit VTF or WLF behavior. From a VTF analysis for both the vacuum electrical relaxation time and electrical conductivity, Ea is found to be about 0.09 eV and T0 is found to be about 40 °C below the "central'' glass transition temperature. In addition, it is found that the activation volumes for the electrical relaxation time and the electrical conductivity are the same when compared relative to T0. These results imply that the mechanism controlling ionic conductivity is the same as that for the α relaxation, namely large-scale segmental motions of the polymer chain.

    Notes: The phyllosphere microbial populations inhabiting the needles of three conifer species, Scots pine (Pinus sylvestris L.), Sitka spruce (Picea sitchensis L.) and Norway spruce (Picea abies (L.) Karst.), exposed to SO2 and O3, in an open-air fumigation experiment were analysed over a 3 year period using serial dilution after washing, direct plating and a fluorescein diacetate (FDA) enzyme assay. Total fungal populations ranged from 102 to 105 colonyforming units (CPU) g−1 fresh weight of needles. The dominant fungi isolated from needles varied with tree species and isolation technique; Aureobasidium pullulans (de Bary) Arnaud was most common on Scots pine and Norway spruce and white yeasts on Sitka spruce using the dilution plating method. However, direct plating of needle segments onto culture media indicated that Sclerophoma pythiophila (Corda) Hohnel was dominant on Scots pine and A. pullulans on Sitka and Norway spruce. Green needles of Sitka spruce were found to be endophytically colonized by Rhizosphaera kalkhoffii Bubak, but seldom by Lophodermium piceae (Fuckel) Hohn during extensive sampling in 1990. Statistical analyses revealed significant differences (P〈0.05) between plots in the 3 year mean of the total fungal populations or the fungal biomass (FDA assay) on all three tree species. Differences between plots were also observed for a number of dominant component species. Data were also analysed for treatment effects. A significant effect of SO2 treatment was observed on the total fungal populations on Sitka spruce (P〈0.05) which were reduced markedly by the low-SO2 treatment, while the O3 treatment caused a significant increase in total fungal numbers on Scots pine (P〈0.05). The FDA activity on needles of both Scots pine and Sitka spruce was noticeably higher in the 03-only treatment plot, but the overall O3 effect was not significant. Treatment effects were also detected on the occurrence of component species. The serial dilution method revealed an SO2 effect (P〈0.05) of a reduction in the occurrence of pink yeasts on Sitka spruce and an O3 effect (P〈0.05) of an increase in the occurrence of S. pythiophila on Sitka spruce (P〈0.01) but a decrease of Epicoccum nigrum Link and Cladosporium spp. on Scots pine. The direct-plating method revealed an SO2 effect of an increase in S. pythiophila on Norway spruce (P〈0.05). Ozone treatment caused a significant increase in the isolation of a black strain of A. pullulans on Norway spruce (P〈0.05). Endophytic colonization of Sitka spruce needles by R. kalkhoffii was found to be increased on two occasions by O3 exposure.

    Notes: Sperm motility variables from the milt of the common carp Cyprinus carpio were assessed using a computer-assisted sperm analysis (CASA) system across several months (March-August 1992) known to encompass the natural spawning period. Two-year-old pond-raised males obtained each month were primed with an exogeneous hormone injection of carp pituitary extract and stripped of milt 18–24h later. The milt was diluted, activated and videotaped using a high-speed (200 Hz) videocamera and recorder. Videotaped samples were subsequently analysed using the CellTrak/S CASA system (Motion Analysis Corp.) for percent motility, curvilinear and straight-line velocity. In addition to assessing changes in motility parameters across several months, a comparison was made between two predilution/ activation media combinations (homologous seminal plasma/NaCl+HEPES v. 2-h incubation in 200 mM KCl+Tris/Tris). The percentage of motile cells assessed immediately after activation was significantly greater in the summer months (May, June, July) when compared to a spring sampling point (March); when assessed 1 min after activation, cells prediluted in seminal plasma maintained a higher percentage of motility than those prediluted in KC1. Curvilinear and straight-line velocities exhibited a slight seasonal trend; variations in response to the predilution treatments were observed with these measurements. Sperm count gradually increased through April and May (9–63 × 109 to 2–38 × lO10ml– 1 milt), declined in June and July (to 1–83 × lO10ml– 1 milt), and was followed by a steep increase in August (2–74 × 1010 ml– 1 milt). Mean seminal plasma osmolality remained relatively constant (250–265 mOsmol kg – 1) throughout the sampling period.

    Notes: Computer-assisted semen analysis (CASA) technology was applied to the measurement of sperm motility parameters in the common carp Cyprinus carpio. Activated sperm were videotaped at 200 frames s−1 and analysed with the CellTrak/S CASA research system. The percentage of motile cells and both sperm head curvilinear velocity and straight-line velocity were measured following exposure of carp sperm to three predilution conditions and activation in media of differing ionic strengths and osmotic pressures. The highest percentage of motile sperm was obtained following predilution of sperm in seminal plasma and activation in Na-HEPES buffer pH 8.0. This level of motility was equalled after predilution in 200 mm KCl for 2 h. Straight-line velocities and curvilinear velocities of 130 μm s−1 and 210 μm s−1, respectively, were observed. Duration of motility was higher under seminal plasma predilution conditions (over 50% motile sperm at 55 s post-activation). The application provides a sound basis for the assessment of Sperm Characteristics in fish.

    Notes: Abstract. Earthworm acute toxicity, plant seed germination/root elongation (SG/RE) and plant genotoxicity bioassays were employed to evaluate the remediation of a lead-contaminated soil. The remediation involved removal of heavy metals by a soil washing/soil leaching treatment process. A portion of the soil after remediation was rinsed with water in order to simulate exposure to rainfall. The bioassay results showed that the soils before treatment (BT) and after treatment plus water rinsing (RT) were not toxic to earthworms in a 14-day exposure, while after treatment (AT) showed significant toxicity. The LC50 values for Eisenia fetida and Lumbricus terrestris were 44.04 and 28.83 (as % AT soil supplemented in artificial soil), respectively. The phytotoxicity data indicated that all three test soils significantly inhibited lettuce SG/RE in a dose-related manner, with AT being the most phytotoxic. In oats, RT had no effect on SG/RE and AT was more toxic than BT. For the two local site grass seeds tested (blue grama and sideoat grama), the AT soil was the most phytotoxic followed by BT and RT. In Allium cepa (common onion), BT and AT induced similar levels of genetic damage to root tip cells, whereas RT was not genotoxic. High salt levels generated during the remediation process appeared to be responsible for the increased toxicity of AT soil for both plants and earthworms. The rinsing of the AT soil with water effectively removed both acutely toxic and genotoxic components of the soil.

    Notes: Abstract Medicago sativa lines with a high incidence of regeneration were established as suspension cultures and used to select for NaCl tolerant lines. Attempts were then made to regenerate plants from these lines. Regeneration was severely depressed in NaCl tolerant calli and the only plants that were successfully regenerated were from one callus of M. sativa cv. Regen S which grew in 62.5 mM NaCl. Plants from this callus, and new calli derived from the recovered plants, have shown a tolerance to NaCl comparable to calli and plants from the initial seed stock rather than an improved level of tolerance.

    Notes: Abstract Molecular approaches are particularly useful for measuring genetic diversity and were applied to samples of central stonerollers obtained from sites along tributaries to the Great Miami River in Ohio. We used Random Amplified Polymorphic DNA (RAPD) analysis to assess the level of genetic diversity within and among these populations. RAPD analysis generates genetic profiles that were used to develop indices of genetic similarity. The RAPD method provides a cost effective means of generating an arbitrary sample of anonymous loci across the genome and generate a virtually unlimited set of loci for use in genetic analysis in the absence of specific sequence information. These attributes make RAPDs well suited for use in evaluating the diversity and assessing the potential vulnerability to exposure of populations across multiple spatial scales. The results demonstrate that a significant amount of structuring exists among populations analyzed to date and that a trend exists towards genetic diversity being an inverse function of site distance from the main stem as well as a being directly related to stream order. This indicates that populations farthest from main conduits or in lower order streams, and thereby most isolated, may be the most vulnerable populations to stressor exposure. It is hoped that information pertaining to genetic diversity, when integrated with other metrics of resource condition, will aid in making scientifically grounded decisions on resource management that enhance the probability of population survival and preserve natural evolutionary processes.
