    Notes: Summary A method is described by which hyperimmune anti-species globulin can be obtained in approximately 10.0 ml. quantities from the albino female mouse. The method utilizes an ascitic reaction produced by Freund's complete adjuvant to which Mycobacterium smegmatis has been added. The anti-globulin titre obtained after an immunization course of four inoculations of rabbit or human gamma globulin is usually about 1∶64. The method is trouble free and the anti-globulin is obtained in a clean form which only needs separation after clotting.

    Notes: Summary A simple procedure is described for the preparation of FITC coupled mouse anti-species globulin. The procedure is based upon sodium sulphate precipitation, followed by inwards dialysis of FITC into the purified mouse globulin. Passage through G-25 sephadex columns gave conjugates which were virtually free of non-specific staining properties. The results obtained from 3 batches each of anti-rabbit and anti-human globulin are described in terms of specific and non-specific staining titres for use in the identification of bacteria and viruses in diagnostic material.

    Notes: Summary An investigation of some of the characteristics of the VERO cell line (Cercopithecus aethiops kidney) is reported, in which the suitability of the cells for use in routine diagnostic virology was examined. VERO cells will: 1. grow to monolayers as rapidly as other heteroploid cell lines, but will maintain as usable monolayers in conventional maintenance medium for a significantly longer time; 2. grow a selection of ten representative viruses to a higher titre than will two other heteroploid lines or two cell strains; 3. support the growth of a very wide range of viruses with development in most instances of a characteristic cytopathic effect. VERO cells are recommended as possible substitutes for primary monkey kidney cell cultures, and are superior in overall performance to primary human amnion, HEp-2 or HeLa cells for diagnostic use in virology.

    Notes: Summary The preparation of an antiserum to a purified suspension of adenovirus type 5 hexons is described. The antiserum reacts by complement fixation tests with type 5 and with many other adenoviruses. The anti-hexon serum can be used for the immunofluorescent detection of the site of hexon synthesis within tissue culture cells infected with many different adenoviruses, and therefore allows diagnostic detection of the adenovirus group antigen. In experimentally infected cultures, “group” identification could be made within 24 hours of infection when many different adenoviruses were tested.

    Notes: Summary A series of related benzo(b)thiophene derivatives was found to exhibit a high degree of anti-influenza activityin vitro. Representatives of the most active sidechain groups were chosen for a more detailed study of their mode of action. Timed treatment experiments indicated that two distinct types of activity were evident in this group of compounds. One occurred early in the virus growth cycle at a time similar to inhibition by amantadine HCL. The second inhibited some metabolic event after viral uncoating but before the earliest time at which nuclear antigen inhibition by p-fluorophenylalanine could be demonstrated.

    Notes: Summary A series of related benzo(b)thiophene derivatives was found to exhibit a high degree of anti-vaccinia activityin vitro. Representatives containing the four most active sidechain groups were chosen for a more detailed mode of action study. Timed experiments indicated that each compound inhibited vaccinia virus at a different point in its replication cycle. The guanidino and pyrrolidino derivatives were acting early in the growth cycle while the alkyl compound acted 2–3 hours later. The action of the morpholino representative corresponded closely to that of 5-iodo-2′-deoxyuridine.

    Notes: Summary A comparison has been made between the indirect fluorescent antibody technique and conventional tissue culture techniques for the identification of pathogenic viruses in cerebro-spinal fluid specimens obtained from patients suffering from aseptic meningitis. The viruses identified from 20 patients included Coxsackie B3, B5, ECH06, Mumps and Herpes simplex. The infecting virus was correctly identified with rapidity in 100% of cases by the immunofluorescent technique, whereas only 70% were isolated in tissue culture. The reasons for the disparity are discussed — and the findings are compared with serological findings from the same cases.

    Notes: Summary A method is described for the construction of five intersecting pools of guineapig antiserum to be used in the immunofluorescent identification of eight common respiratory viruses. The component antisera are highly purified and produce no non-specific fluorescence at working titre before or after incorporation into pools. The antiserum pools are fully stable at 4°C for up to 2 weeks, and for at least 3 months when stored at −20° C. Due to their intersecting nature, a direct “read-out” of the type of virus present is possible. The pool system also offers the advantage of built-in negative controls for each specimen examined.

    Notes: Summary A large group of related benzo(b)thiophene derivatives was screened for antiviral activity in a simple cell culture system. Three viruses were used; influenza A2, vaccinia and herpes simplex type 1.51 per cent of the compounds inhibited at least one virus by ≥75 per cent, while 25 per cent were inhibitory by ≥90 per cent. This high degree of antiviral activity is in agreement with the range of biological properties possessed by benzo(b)thiophene derivatives as a whole.
