    Notes: Abstract The diameter of nucleoprotein fibres of metaphase chromosomes is sensitive to salt concentrations. Treatment of human lymphocyte cells in metaphase with a hypotonic medium and spreading them on a water surface causes swelling of the chromosome fibres from 150–180 Å to 230–250 Å. Treatment of the chromosomes with 0.15–0.45 M NaCl, a concentration at which histones are not yet removed from the nucleoprotein complexes, apparently does not affect the chromosome structure. Treatment with NaCl solutions between 0.6 M and 2.0 M NaCl leads to a progressive extraction of the chromosomal proteins and decreases the diameter of the chromosome fibres to 80–100 Å and less.

    Description / Table of Contents: Summary Solutions of high molecular calf-thymus-DNA were mixed with dimethyl formamide solutions of 12 cancerogenic inactive and active aromatic hydrocarbons and amines in order to study the reciprocal effect with DNA. The solutions were then dialyzed, extracted with organic solvents, and centrifuged. From the UV spectra of the solutions the following were determined: (a) the amount of “bound” compounds from the bands of aromatics above 300 mμ; (b) the extinction at 260 mμ as a measure of the DNA present in the solution. By following the extinction with increases in temperature the hyperchrome effect and the “melting point” of DNA were determined. Relationships between the data obtained and the cancerogenic activity of the compounds were not found. In addition, pyridino-[2,3∶3,4]-phenoxazon-(2) and the follicle hormone estradiol-(3,17 β) were without effect. The reciprocal effect of DNA with the mutagens, proflavin and acridine orange, was confirmed. A distinct increase of the “melting point” of DNA by means of N-phthalyl-glutaminic acid-imide was striking.
    Notes: Zusammenfassung Zur Untersuchung der Wechselwirkung mit DNA wurden Lösungen von hochmolekularer Kalbsthymus-DNA mit Dimethylformamid-Lösungen von zwölf krebserzeugend unwirksamen und wirksamen aromatischen Kohlenwasserstoffen und Aminen versetzt. Anschließend wurden die Lösungen dialysiert, mit organischen Lösungsmitteln extrahiert und zentrifugiert. Aus den UV-Spektren der Lösungen wurden bestimmt: a) aus den Banden der Aromaten oberhalb 300 mμ die Menge an “gebundener” Verbindung, b) die Extinktion bei 260 mμ als Maß für die in Lösung vorhandene DNA und durch Verfolgung der Extinktion bei Temperaturerhöhung der hyperchrome Effekt und der “Schmelzpunkt” der DNA. Beziehungen zwischen den erhaltenen Daten und der krebserzeugenden Wirksamkeit der Verbindungen wurden nicht gefunden. Ohne Effekt waren ferner Pyridino-[2,3:3,4]-phenoxazon-(2) und das Follikelhormon Östradiol(3,17 β). Bestätigt wurde die Wechselwirkung mit DNA für die Mutagene Proflavin und Acridinorange. Auffallend ist eine deutliche Erhöhung des “Schmelzpunktes” der DNA durch N-Phthalyl-glutaminsäure-imid.

    Notes: Zum Studium des 5.6-Dihydro-1.4-oxazinon-(2)-Systems werden 3-Phenylderivate durch Kondensation von 1.2-Aminoalkoholen mit Phenylglyoxylsäureester dargestellt. Im Falle des 1-Amino-2-hydroxy-2-methyl-pentans, das eine tertiäre Hydroxylgruppe trägt, führt die Umsetzung zum Oxazolidinderivat. Die als Nebenprodukte bei den Kondensationen auftretenden Phenylglyoxylsäure-[2-hydroxy-alkylamide] liegen, wie sich spektroskopisch zeigen läßt, im Gleichgewicht mit cyclischen Halbacetalstrukturen vor.

    Notes: Durch katalytische Hydrierung der 3-Phenyl-5.6-dihydro-1.4-oxazinone-(2) (1) werden N-unsubstituierte Morpholone-(2) (II) zugänglich. An einem Beispiel wird deren Tendenz zur 2.5-Dioxo-piperazin-Bildung nachgewiesen. Reduktion der Dihydro-oxazinone mit Lithiumaluminiumhydrid führt in hohen Ausbeuten zu Dialkanolaminen IV. Die Reduktion der Phenylglyoxylsäure-[2-hydroxyalkylamide] (VI) liefert isomere Dialkanolamine VII. Durch Wasserabspaltung lassen sich die Dialkanolamine in substituierte Morpholine überführen.

    Notes: The toxic metabolite of the basidiomycete Gloeophyllum abietinum, oosponol (6b), and the structural analogues (Figure 1) were synthesised in order to investigate which partial structures of the molecules are responsible for their biological activities. Different organisms were employed to test the antibiotic activities of the analogues. From the results obtained with the synthetic analogues of oosponol (6b), it became evident that the toxicity of this fungal metabolite can be attributed to a vinylogous acid anhydride structure, which in nature is produced by dehydrogenation of the nontoxic precursor oospoglycol (7).
