Zusammenfassung Bei der Untersuchung des Einflusses verschiedener Fixationsmethoden auf die Feinstrukturen der Crista ampullaris des Meerschweinchenlabyrinthes wurden folgende Methoden miteinander verglichen: 1. Intravasale Perfusionsfixation mit 3% igem Glutaraldehyd, Osmiumnachfixation (Millonig). 2. Intravasale Perfusionsfixation, Vorspülen mit Novocain und Liquemin, Fixation mit 3% igem Glutaraldehyd, Nachfixation Osmiumtetroxid (Millonig). 3. Intraampulläre Fixation mit 3 bzw. 6% igem Glutaraldehyd, Osmiumnachfixation (Millonig). 4. Immersionsflxation mit 3% igem Glutaraldehyd, Osmiumnachflxation(Millonig). Die besten Ergebnisse wurden an den Sinneszellen mit der intraampullären Perfusionsfixation mit Glutaraldehyd und Osmiumnachfixation gefunden, die schlechtesten bei der intravasalen Perfusionsfixation. Die Ergebnisse bei den dunklen Zellen waren uniformer. Bei allen angewendeten Methoden fanden sich regelmäßig Artefakte, die denen glichen, die nach Anwendung ototoxischer Substanzen auftraten. Nach diesen Untersuchungen empfiehlt es sich, bei Arbeiten an den dunklen Zellen primär und intraampullär Osmiumtetroxid zu verwenden. An den Sinneszellen war die Bildqualität bei Anwendung der intraampullären Perfusionsfixation mit Glutaraldehyd und Osmiumnaehfixation besser, als bei alleiniger Osmiumanwendung, wobei allerdings in geringerem Umfang Artefakte auftraten.

Summary The influences of various fixative-methods on the crista ampullaris of healthy guinea pigs have been studied. The following methods were used: 1. Vascular perfusion with 3% glutaraldehyde, postfixation with osmiumtetroxide (Millonig). 2. Vascular perfusion as under Nr. 1, prewash with Novocain and Liquemin. 3. Intraampullar fixation with 3% glutaraldehyde, postfixation with osmiumtetroxide (Millonig). 4. Intraampullar fixation with 6% glutaraldehyde, postfixation with osmiumtetroxide (Millonig). 5. Fixation by immersion with 3% glutaraldehyde, postfixation with osmiumtetroxide (Millonig). The intraampullar fixation was considered superior to the other methods with respect to the sensory cells. Remarkably strong signs of damage were found by using the vascular technique. The results in respect to the dark cells were more uniform. We always found signs of damage similar to those following application of ototoxic substances. With respect to the dark cells we therefore believe that it is necessary to use primarily the intraampullar fixation with osmiumtetroxide. On the other hand, better pictures of the sensory cells are obtained by using firstly the intraampullar fixation with glutaraldehyde and subsequently postfixation with osmiumtetroxide. Under these circumstances a small amount of artefacts were nevertheless to be found.
