Zusammenfassung Bereits 10 min nach 0,5 ml/kg KG CCl4 i.p. ist bei reifen ♂ Wistar-Ratten die Phenazonhydroxylierung und Aminophenazon-N-Demethylierung durch 9000 g-überstand in vitro um etwa 50% gehemmt. Nach 60–90 min werden wieder normale oder sogar übernormale Werte erreicht. Die Kodein-O-Demethylierung hingegen ist 10 min nach der CCl4-Applikation nicht beeinträchtigt, sinkt kontinuierlich ab und liegt nach 180 min deutlich unter den Normalwerten. Werden den Inkubationsansätzen mehr als 10fach höhere CCl4-Mengen zugesetzt, als maximal nach i.p. Applikation zu erwarten sind, ist eine Hemmung der Phenazon-Hydroxylierung, Aminophenazon-N-Demethylierung und Kodein-O-Demethylierung nicht sieher nachweisbar, während bei den CCl4-Konzentrationen in vitro, die der in vivo maximal erreichbaren CCl4-Konzentration in der Leber etwa entspricht, eher eine geringe Aktivitätszunahme gemessen wurde. Aus den Ergebnissen wird geschlossen, da\ die in vivo-Wirkung von CCl4 bei der angewandten Methode nicht auf in vitro-Effekte von CCl4 zurückzuführen sind.

Summary Already 10 minutes after the intraperitoneal application of 0.5 ml/kg CCl4 in mature Wistar male rats, phenazone hydroxylation and aminophenazone N-demethylation reactions in vitro using 9,000 g supernatant are inhibited by about 50 per cent. Normal or above normal values are again reached only after 60 to 90 minutes. In contrast, the 0-demethylation of codeine remains uneffected 10 minutes after CCl4 application, but it then decreases continuously and is definately below normal after 180 minutes. No significant inhibition of any of these reactions could be definitely demonstrated when CCl4 was added to the incubation mixture in an amount more than ten times that expected in the liver after intraperitoneal application. On the other hand, at CCl4 concentrations in vitro which approximated the maximum ones reached in the liver in vivo, a slight increase in the activity of these reactions was observed. It is concluded that the effects of CCl4 in vivo, given the methods used here, are not to be accounted for by the effects of CCl4 in vitro.
