Model Experiments for Peptide Synthesis. - Kinetic Investigations of the Reactivity of Activated Carboxyl FunctionsModel experiments for peptide synthesis were carried out with N-methylbenzylamine (2a) and N-methyl-N-(trimethylsily1)benzylamine (2b) as nucleophiles and activated Z-glycine derivates as electrophiles. 2-Glycine was activated as the imidazolide l h as well as the ester of 4-chlorophenol la, 4-chlorothiophenol 1b, 4-nitrophenol lf, 2,4,6-trichlorophenol lc, 2,4,5-trichlorophenol 1d, pentachlorophenol 1e and N-hydroxysuccinimide 1g. The kinetic investigations were carried out by 1H-NMR spectroscopy in chloroform at 28°C. The Z-glycine ester of N-hydroxysuccinimide and of 4-chlorophenol show the greatest difference in reactivity; their rate constants differ by a factor of about 5000. - Only the reaction of 2-glycine imidazolide with N-methylbenzylamine shows exactly second order kinetics. The reasons for the deviation from this reaction order and a modified rate law are considered. The free energy of activation (AG:J for rotation of 2-glycine-N-methylbenzylamide has been determined.

Modellversuche zur Peptidsynthese wurden mit N-Methylbenzylamin (2a) sowie N-Methyl-N-(trimethylsilyl)benzylamin (2b) als nucleophile Komponente und mit aktivierten Z-Glycinderivaten durchgefuhrt. Das Z-Glycin wurde als Imidazolid 1 h, sowie als Ester des 4-Chlor-phenols la, 4-Chlorthiophenols 1b, 4-Nitrophenols If, 2,4,5-Trichlorphenos 1d, 2,4,6-Trichlorphenols 1c, Pentachlorphenols 1e und N-Hydroxysuccinimids 1g aktiviert. Die in Chloroform bei 28°C durchgefuhrten Versuche wurden anhand von 1H-NMR-Spektren kinetisch verfolgt und die Geschwindigkeitskonstanten ermittelt. Den größten Reaktivitätsunterschied zeigten der N-Hydroxysuccinimidester und der 4-Chlorphenylester, deren Geschwindigkeitskonstanten sich um den Faktor 5000 unterscheiden. Nur bei der Umsetzung von Z-Glycinimidazolid mit N-Methylbenzylamin wird exakt eine Reaktion zweiter Ordnung gefunden. Die Ursachen für Abweichungen von dieser Reaktionsordnung bei allen anderen Reaktionen werden diskutiert; ein modifiziertes Geschwindigkeitsgesetz wird erörtert. Von Z-Glycin-N-methylbenzylamid wurde die freie Aktivierungsenergie (AG18) der Rotation gemessen.
