    Description / Table of Contents: Zusammenfassung Es wird ein Verfahren vorgestellt, das die Aufnahme von spektralen Absorptionskurven histochemischer Farbreaktionen mittels prozeßgesteuerter Mikrospektralphotometer ermöglicht, die nach dem Einstrahlprinzip aufgebaut sind. Die Vorteile gegenüber der konventionellen Methode sind: 1. Zeitersparnis um den Faktor 50–100 durch automatisierten Meßablauf und Rechenhilfe. 2. Hohes Auflösungsvermögen auf Grund großer Meßpunktdichte. 3. Universellere Anwendbarkeit als die Spektroskopie mit Zweistrahlphotometern.
    Notes: Summary A method is described, which enable one cytophotometrically to draw exactly the spectral curves of histochemical reactions using one beam photometers and computer processing. There exist advantages compared with conventional methods: 1. Gain of time (50 to 100 times) according to automation in measurement and calculations. 2. High resolving power according to closely packed readings. 3. Great versitility compared with spectroscopy using two beam photometers.

    Notes: Summary Flow through microfluorometry of phenantridium and fluoresceinisothiocyanate stained HeLa cells showed the absence of extranuclear RNA related fluorescence after prolonged hydrolysis of cells in ethanolic barium hydroxide. DNA distribution were as good as in RNAse treated samples. Distribution pattern of cellular protein/DNA ratios yielded greater resolution power than those after RNAse digestion. The cellular protein content has not been affected. The ethanolic hydrolysis has the advantage to be of less expense and to allow cell conservations for further measurements with other well preserving fixatives. Thus, application of glutaraldehyde has been proved to be valuable.

    Notes: Summary A dual laser beam excitation device for flow analysis of biological particles has been developed. The aid of this arrangement is to increase the range of fluorescent agents employed so far in quantitative and qualitative cytochemistry. Combining an argon ion and a helium-cadmium laser two color fluorescence measurements were performed employing propidium iodide as a DNA stain and fluorescamine which stains total protein in fixed cells. Energy transfer processes between the antibiotic and DNA specific dye mithramycin and propidium iodide both being bound to nuclear chromatin were analyzed. Utilization of energy transfer processes is generally discussed as a mean to extract information about the structure and conformation of nuclear chromatin in situ. The application of a crypton ion laser with three lines near 400 nm and a single line at 350 nm having a light output in each range of nearly one Watt gives the opportunity of utilizing DNA fluorochromes which have an excitation maximum in the deep blue region. DNA spectra are shown employing mithramycin, the benzimidazol derivative 33258 (Hoechst) and the indol compound DAPI which has a high DNA specifity combined with a great stability under UV illumination. By separating two focussed laser beams at their intereecting points with the liquid sample stream the trajectory of each flowing cell crosses the beams sequentially, which causes a solitary dual excitation of each cell. The advantages of a solitary excitation device compared with a simultaneous one is discussed.

    Description / Table of Contents: Zusammenfassung Durch Mikrodensitometrie und Mikröfluorometrie wird der DNA-Gehalt isolierter, feulgengefÄrbter Kerne aus etiolierten Hypocotylen vonSinapis alba bestimmt. Im Laufe des Streckungswachstums der Hypocotyle von 40–64 Stunden nehmen die endopolyploiden Zellen zu. Gleichzeitig steigt der DNA-Gehalt der Chromozentren stÄrker an, so da\ man eine überreplikation der DNA in den heterochromatischen Chromosomenabschnitten annehmen kann. Die biochemischen und die cytophotometrischen Bestimmungen der DNA-Menge stimmen gut überein. Eine Bakterienkontamination als Ursache für die biochemisch gemessene DNA-Zunahme kann auf Grund der direkten Kernmessungen ausgeschlossen werden.
    Notes: Summary The relative DNA content (arbitrary units) per nucleus was determined in isolated nuclei of the etiolated hypocotyl ofSinapis alba by Feulgen microdensitometry and microfluorometry. An increase of endopolyploidy during elongation growth was detected. With increasing hypocotyl age (40 to 64 hours) the DNA concentration in the chromocenters increases. A hyperreplication of the DNA in the polyploid nuclei can be assumed. Biochemical determinations of DNA and the relative DNA amounts per nucleus determined by cytophotometry agree closely. Bacterial contamination has been excluded to account for the biochemically determined DNA increase.

    Notes: Abstract The influence of injecting 38-year-old Larix occidentalis with gibberellin A4/7 (GA4/7) on seed and pollen cone production was studied in western Montana, U.S.A. Two natural stands thinned to several spacings in 1961 were chosen. The two widest spacings were used. In 1991, 60 trees selected for study at each site were randomly divided into two groups of similar DBH, half as controls and half injected in June 1991 with a solution of GA4/7 in ethyl alcohol. The volume injected was adjusted to the DBH of each tree (60 mg per 5 cm diameter). In June 1994, half the trees treated with GA4/7 in 1991 and half the untreated trees were injected with GA4/7 in the same manner as before. In 1992 and 1995, seed cone production increased on GA4/7-treated trees compared to the controls. Pollen cone production was significantly increased by GA4/7 treatment but only at one site. Delayed effects of GA4/7 on seed cone production was not evident in subsequent years after treatment. Foliar and shoot damage on treated trees was attributed to GA4/7 treatment but most trees recovered completely the following year. Finally, GA4/7 treatment did not affect cone length, potential seed per cone or filled seed per cone.

    Notes: Abstract The effects of top pruning on subsequent seed and pollen cone production in lodgepole pine (Pinus contorta Dougl.) grafted clones in two seed orchards were evaluated. In the older orchard (14 years old), three pruning regimes were applied: an untreated control, a moderate pruning where 25% of the crown was removed and a severe pruning where 40% of the crown was removed. In the younger orchard (ten years old), only a control pruning and a severe pruning (50% of crown removed) were applied. Five-year seed cone production was lower in treated ramets compared to controls in both orchards. However, when cumulative seed cone yields were adjusted for reduced tree heights after pruning, differences were no longer significant in the older orchard. Strong clonal variation was observed in seed cone yields in both orchards. Pollen cone development was less variable among treatments in the older orchard. In the younger orchard, controls consistently produced heavier pollen crops. Pruning recommendations for similar orchards are made and results discussed for reducing the impacts of losses in seed cone yields.

    Notes: Flow system technology enables the biological and medical experimenter to investigate the distribution spectra of various cellular characteristics separately or in parameter combination on the basis of ultrarapid single cell measurements. A typical rate of recognition is about 1000 to 5000 cells per second and the precision of measurements and their statistical relevance has been previously unobtainable. According to the approach of the multiparameter analysis and high data rate, computer assistance in flow system technology is given primary consideration. In this study three different kinds of software controlled modes in data acquisition are demonstrated: normal acquisition and linear accumulation of single parameters, spectra accumulation of two correlated parameters of each single cell and documentation as linear, two- or three-dimensional distribution pattern, and linear accumulation of two correlated parameters simultaneously with their actual signal-to-signal ratio. A first attempt to analyze distribution spectra was the application of the entropy of the structure routinely used in cybernetics. This function seems to be a measure for determining the degree of synchrony in an appropriate pretreated cell population. A special mathematical strategy has been applied to the linear spectra of cellular DNA content, whose advantage is the quantitative extraction of the fractions concerning the various phases of the life cycle cells. The validity of this special curve fitting procedure has been proven on various experimental cell populations.
