    Notes: Summary Proline production via a part of the arginine biosynthetic pathway was examined. About 20 mg/ml ofl-proline was produced by using arginine biosynthetic enzymes. Accordingly, three mutations of arginine biosynthesis, namely, derepression of arginine biosynthetic enzymes (assigned byargR2), feedback inhibition-resistant N-acetylglutamate synthase (assigned byargA2) and defectiveness in N-acetylornithine aminotransferase (assigned byargD −) were introduced by three transductional crosses into a proline-producing strain which produced about 55 mg/ml ofl-proline. The constructed strain produced 62 mg/ml ofl-proline, although about 10 mg/ml ofl-arginine and 1 mg/ml of N-acetylglutamate-γ-semialdehyde were produced as by-products.

    Notes: Summary Proline-producing strains of Serratia marcescens Sr41 were constructed by three rounds of mutagenesis. A strain SP103 which did not degrade l-proline carried the putA mutation leading to lack of proline oxidase. A 3,4-dehydroproline-resistant mutant SP105, derived from strain SP103, carried the dpr-1 mutation which resulted in desensitization of the feedback inhibition of glutamate kinase. Strain SP103 produced 5.5 mg of l-proline per ml of fermentation medium containing sucrose and urea. Growth inhibition by proline analogs was enhanced when succinate was used as a carbon source in the medium. A thiazolidine-4-carboxylate-resistant mutant SP126 derived from strain SP105 produced 20.5 mg of l-proline per ml of medium. The mutation carried by strain SP126 might be distant from dpr-1 and putA mutations on the chromosome. Pyrroline-5-carboxylate reductase was not repressed by proline in S. marcescens Sr41.

    Notes: A rotating flow mixing device having three directional inlets was developed for a high performance liquid chromatographic peroxyoxalate chemiluminescence detection system for the sensitive detection of fluorescent compounds separated with acidic mobile phases, such as those containing trifluoroacetic acid. The diameters of the three inlets were 0.3, 0.3 and 0.5 mm i.d., for an acidic column eluate, chemiluminogenic reagents (bis[4-nitro-2-(3,6,9-trioxadecyloxycarbonyl)phenyl] oxalate and hydrogen peroxide) and imidazole solution (for neutralization and as a catalyst), respectively. The three mixed solutions flowed out through an outlet (0.5 mm i.d.) at the centre of the top of the vessel. The reaction conditions to give the appropriate signal-to-noise ratio (S/N) were investigated using dipyridamole (an antiplatelet aggregation drug) as a typical fluorescent compound. The system was applied to the detection of dipyridamole and dansylated amino acids with a mobile phase containing trifluoroacetic acid. The detection limits for dipyridamole and each dansylated amino acid were 10 amol and sub-fmol (S/N=2), respectively.
