    Notizen: Spermatozoa of the hydroid Pennaria tiarella were examined with the electron microscope. The anterior region is characterized by the presence of 30-40 membrane-bounded vesicles which lie anterior to the nucleus. These vesicles are apparently derived from the Golgi apparatus.The nucleus is conical in shape with a protrusion at the anterior end. Posteriorly it is indented by four radially arranged mitochondria. Lying within the fossa formed by the mitochondria are proximal and distal (filament forming) centrioles. The distal centriole is characterized by nine centriole satellite projections which emanate from its matrix. The tubules of the distal centriole are continuous with the alpha filaments of the tail.The tails are typical 9 + 2 flagella with 9 peripheral doublet (or alpha) filaments surrounding two central (or beta) filaments.

    Notizen: The interstitial cells of Pennaria tiarella differentiate exclusively from the central endoderm of the planula. Shortly after their appearance, most of the interstitial cells become cnidoblasts. Subsequently, as the larva transforms into a polyp, both cnidoblasts and interstitial cells migrate from the endoderm, through endoblast and mesoglea, into the ectoderm. It is suggested that some interstitial cells remain in the endoderm and differentiate into the gland and mucous cells of the polyp gastroderm.

    Notizen: The ultrastructure of the centriolar satellite complex in the spermatozoon of the hydroid, Pennaria, is presented. The complex consists of nine spoke-like arms which emanate from the distal centriole and of nine wedgeshaped connectives which extend into the flagellum joining each of the alpha doublets and the flagellar plasmalemma. Based upon these observations a new model for the structure of the centriolar satellite complex is proposed. In addition, similar centriolar satellite complexes are reported in the spermatozoa of two echinoderms, Ctenodiscus crispatus and Thyone briareus, in support of the proposed model.

    Notizen: The presence of striated subumbrellar musculature in hydromedusae can be related to the development in hydrozoans of a free-swimming life stage. The detailed ultrastructure of the striated subumbrellar musculature of the anthomedusan, Pennaria tiarella is presented. The striated musculature of Pennaria resembles vertebrate striated muscle in filament arrangement (L2 lattice pattern) and M line structure. The striation pattern, out-of-register myofilaments, filament structure as determined by rotational symmetry, Z line structure, types of intercellular junctions, and sarcoplasmic reticulum are more similar to structures found in other invertebrate striated muscle.

    Notizen: Morphological changes in the mitotic cytoskeleton (MC) that occurred through the course of the first cleavage of Strongylocentrotus droebachiensis, cultured at 8°C or at 0°C, temperatures within the natural range for this species, have been investigated. Electron microscopy of MCs isolated from zygotes has revealed that they consisted largely of microtubules (mts). Thus, the morphology of these MCs is derived from the arrangement of the mts which form them. During anaphase, astral rays elongated while kinetochore fibers shortened. Asters enlarged during anaphase as a result of two events: astral ray lengthening and centrosphere enlargement. At the end of anaphase, asters of 8°C MCs filled the entire cell volume. The pattern of changes that occurred in 8°C mitotic apparatuses (MAs) also occurred in 0°C MCs. The observation of asters in 0°C MCs is contradictory to that of Stephens ('72b), who reported that 0°C MCs in this species were anastral. However, in 0°C metaphase MCs, the astral rays and spindle fibers were not as long as those in 8°C MCs. Also in 0°C MCs, the centrosphere was largely filled with dense material, whereas the centrosphere in 8°C MCs was larger and contained little dense material. Asters of 0°C MCs did not attain a large enough size to fill the egg volume completely, as did asters of 8°C MCs.

    Notizen: Several methods were utilized to visualize the structure and orientation of the blastocoelic extracellular matrix (ECM) in Strongylocentrotus purpuratus embryos at the mesenchyme blastula stage. Rapid freezing in liquid propane cooled to LN2 temperatures followed by freeze substitution was used to preserve the ECM without shrinkage due to dehydration. Scanning, transmission, and light microscopy were employed to elucidate the ECMs' structure. The blastocoelic ECM consisted of parallel fibrillar sheets that were interconnected by finer filaments and oriented along the animal-vegetal axis. The ECM completely filled the blastocoelic cavity as viewed by scanning electron microscopy. The basal lamina could be distinguished from the blastocoelic ECM as a thin coat on the plasma membrane of epithelial cells; the ECM was in contact with this coat. In contrast, the blastocoelic ECM attached directly to the plasma membrane of primary mesenchyme cells (PMC) which did not possess a basal lamina. The blastocoelic ECM was isolated as an intact “bag” and probed in a hydrated state with Con A and alcian blue. Confocal microscopy confirmed that the entire blastocoel was filled with a fibrillar ECM. These approaches offer advantages for future studies of the ECMs of sea urchin embryos and their roles in gastrulation. © 1992 Wiley-Liss, Inc.

    Notizen: A laser scanning confocal microscope has been used to investigate the development of the sea urchin embryo. The samples were fixed in Carnoy's solution at various developmental stages, stained for DNA with the Feulgen reaction, and optically sectioned with a BioRad MRC-500 confocal microscope. Computer-generated stereographic projection images and a three-dimensional contour tracing and reconstruction system were employed to investigate the cleavage pattern during the 6th cleavage division. Cell division is found to be asynchronous during the 6th cleavage, with macromere derivatives completing division first, followed by mesomeres, and finally by the outer quartet of micromeres (which begins division only after macromeres and mesomeres have completed their respective divisions). Sixth cleavage produces an embryo comprising 60 cells. Asynchronous division was also observed within individual tiers of blastomeres. Variations in the orientations of cell division axes within individual tiers of cells were also observed. The utility of computer-graphics reconstruction techniques for both quantitative and qualitative developmental analysis are discussed.
