Notes: Summary A complement fixation kinetics test is described for dengue viruses employing antisera obtained from a variety of sources using the standard NCDC micro CF test with parameters that included various time periods of antigen-antibody-C' interaction and various concentrations of antigen. Antibody to the homotypic dengue serotype is detected earliest during the course of the kinetics. A progressive increase in homologous titers occurred followed thereafter by the detection of cross-reacting CF antibodies. The initial time of antibody detection was dependent upon the antigen-binding capacity of the serum as well as the antigen concentration employed. Irrespective of the antigen concentration, homologous peak titers were achieved only after overnight incubation; however, they were concurrent with heterologous CF antibodies. Implementation of this procedure facilitated the differentiation of dengue viruses as to type and sometimes as to strain; thus permitting identification which was not possible by the conventional serological procedure.
