Notizen: Differences in isozyme and RAPD-PCR polymorphisms amongst 33 isolates of Fusarium avenaceum were compared using native polyacrylamide gel electrophoresis and agarose gel electrophoresis. The isolates were collected from different regions of Finland. Amongst eight enzymes analysed clear isozyme polymorphism was detected in five enzymes which could be grouped into 20 different electrophoretic phenotypes and three main groups at the similarity level of 70% in unweighted pair group method with arithmetic average (UPGMA) analysis. RAPD-PCR analysis differentiated all F. avenaceum strains from each other. The phenotypes resulting from RAPD-PCR analysis were grouped into five main groups by UPGMA analysis at the similarity level of 55%. These main groups had several similarities with the main groups from isozyme analysis. RAPD-PCR patterns of 16 isolates of Fusarium graminearumF. culmorumF. equisetiF. oxysporum and F. redolens were also studied and strains from each Fusarium species formed individual groups in UPGMA and principal components analyses. Thus, the extent of isozyme and RAPD-PCR polymorphisms found in Fusarium strains potentially provides a method for identifying the fungi both at strain and species level.

Notizen: Abstract  The fatty acid composition of "fish wurst", a fermented salami-type sausage made of pork, lard and Baltic herring fillets (Clupea harengus var. membras) was investigated. Changes in the proportions of the 35 most abundant fatty acids were examined throughout the 1-month ripening period followed by a 4-month storage period. The fat composition of the product was stable (32–35%) and retained the characteristics of the main ingredients: oleic acid (37.4%, mean of three production batches) palmitic acid (23.7%) and linoleic acid (10.7%) from lard and fish, stearic acid (11.7%) mainly from lard, and palmitoleic acid (3.0%) and long-chain (C20–C24), polyunsaturated fatty acids (c.a. 6%) mainly from fish. During the 4-week ripening period a statistically significant increase (P≤0.05) was detected in the proportions of minor fatty acids only, i.e. eicosenoic acid (20 : 1n-9), eicosadienoic acid (20 : 2n-6), docosadienoic acid (22 : 2n-6) and docosatrienoic acid (22 : 3n-3). During the 4-month storage of the ripe sausage, the fatty acid composition stabilized. Only the proportion of stearic acid increased significantly during storage, from 11.7% to 12.5%.

Notizen: Abstract Different screening methods for selection of biological control agents (BCAs), for controlling soil and seed-borne diseases, are discussed. The shortcomings of laboratory methods focused on mechanism of action are discussed and we conclude that these methods should be used with caution if candidates with multifactorial or plant mediated mechanisms of control are to be obtained. In vitro screens may be useful for specific groups of microorganisms, thus, screens for antibiotics may be relevant for Streptomyces spp., and promising results have been obtained using soil plating or precolonized agar methods to screen for mycoparasitism and competitive saprophytic ability. Experience with screening in the Nordic programme ‘Biological control of seed borne diseases in cereals’ is summarized. Research in the four participating countries – Finland, Sweden, Norway and Denmark – followed the same paradigm: that of obtaining antagonists, well adapted to different Nordic environments, and developing them as effective BCAs. Potential antagonists were isolated from different sources and in planta screening methods were developed in order to optimize selection of antagonists effective against a range of seed borne pathogens. Screens in the laboratory or greenhouse were followed by screening in the field. The different screening procedures are compared and evaluated.

Notizen: Summary An ARC/CL coded wheat flour reference material (ARC/CL RM) with established recommended concentration values for 12 elements [1,2] was further characterized for P, K and Cr contents by means of an intercomparison study. Minor corrections were made to earlier recommended values in accordance with new results obtained. Fifteen laboratories using a total of 11 methods based on six entirely different analytical principles participated in the intercomparison study. The medians and standard deviations were calculated for the results of the intercomparison study. After rejecting as outliers values falling outside the median ± sd, the present recommended values were obtained by recalculating the median and 95% confidence limits. Recommended values were established for the major elements Ca, K, Mg, P, the minor elements Cu, Fe, Mn, Zn, and the trace elements Mo, Cr, Ni and Se. The relative standard deviations for the 95% confidence limits of the medians were lower than 10% for major and minor elements, 11% for Mo and 18% for Cr. All of the recommended concentration values fell into category A, having a high degree of confidence. The present RM, with its 16 recommended concentrations, is of value for use in nutritional, food composition and contaminant monitoring studies.

Notizen: Summary Lead and cadmium contents of berries and vegetables of both domestic and imported origin were studied in samples collected from the Finnish food industry (berries) or from large wholesale markets (vegetables) located in Southern and Central Finland. Contents of Pb and Cd were analysed by GFAAS after wet digestion using concentrated HNO3. The contents of heavy metals in berries and vegetables were either very low, at the same level or lower than reported earlier from Sweden, the Netherlands and the USA. The mean contents of Pb (μg/kg) and Cd (μg/kg) in berries and vegetables on the Finnish Market in 1987 were respectively: strawberry 11, 24; black currant 23, 2; red currant 27, 7; apple 2, 〈1; tomato 3, 2; cucumber 2, 〈1; carrot 8, 10; onion 4, 18; white cabbage 3, 3; cauliflower 6, 5 and lettuce 8, 13. The results of the present study are in agreement with those of a Finnish Market Basket study showing that berries and vegetables only slightly contribute to the total lead and cadmium intake in Finland.
