    Notes: The 6Fr Hydrolyser thrombectomy catheter for acute myocardial infarction (AM) successfully removed a massive thrombus in the coronary artery in three patients. The 6Fr Hydrolyser catheter could be advanced into the right coronary artery with bare-wire methods via a 6Fr sheath at the radial artery. This approach suggests that the device can be an alternative method for thrombolysis in selected AMI patients with massive thrombus in the coronary artery.

    Notes: The Hong Mang Mai wheat cultivar is tolerant to deep-sowing conditions because it has an elongated first internode that is sensitive to gibberellin (GA3). The cells in the GA-treated first internodes were approximately 4.2 mm long, twice as long as the untreated Hong Mang Mai first internode cells. The elongation of the first internode of Hong Mang Mai, particularly when treated with GA3, was accompanied by remarkable spiral growth. In contrast, the first internodes of the GA-insensitive cultivar Norin 10 did not exhibit GA3-induced elongation or spiral growth. The walls of the first internode cells of GA3-treated Hong Mang Mai seedlings showed increased extensibility and higher (1→3), (1→4)-β-d-glucanase activity, autolysis and glucan contents than the cell walls of untreated Hong Mang Mai first internodes. The changes in the cell wall extensibility due to GA3 treatment correlated strongly with the GA3-induced changes in cell wall glucan content, autolysis, and glucanase activity. GA3-treated Hong Mang Mai seedlings showed elevated expression of Glucanase EI gene in the first internode compared to GA3-treated Norin 10. Thus, GA aids first internode elongation in Hong Mang Mai by enhancing glucan turnover and thus increasing cell wall loosening. The spiral growth of the first internode also helps the plant elongate against soil resistance, thereby promoting the deep-sowing tolerance of this cultivar.

    Notes: The main mechanism causing catabolite repression in Escherichia coli is the dephosphorylation of enzyme IIAGlc, one of the enzymes of the phosphoenolpyruvate:carbohydrate phosphotransferase system (PTS). The PTS is involved in the uptake of a large number of carbohydrates that are phosphorylated during transport, phosphoenolpyruvate (PEP) being the phosphoryl donor. Dephosphorylation of enzyme IIAGlc causes inhibition of uptake of a number of non-PTS carbon sources, a process called inducer exclusion. In this paper, we show that dephosphorylation of enzyme IIAGlc is not only caused by the transport of PTS carbohydrates, as has always been thought, and that an additional mechanism causing dephosphorylation exists. Direct monitoring of the phosphorylation state of enzyme IIAGlc also showed that many carbohydrates that are not transported by the PTS caused dephosphorylation during growth. In the case of glucose 6-phosphate, it was shown that transport and the first metabolic step are not involved in the dephosphorylation of enzyme IIAGlc, but that later steps in the glycolysis are essential. Evidence is provided that the [PEP]–[pyruvate] ratio, the driving force for the phosphorylation of the PTS proteins, determines the phosphorylation state of enzyme IIAGlc. The implications of these new findings for our view on catabolite repression and inducer exclusion are discussed.

    Notes: We have partially characterized root hydrotropism of an agravitropic pea mutant, ageotropum (from Pisum sativum L. cv. Weibull's Weitor), without interference of gravitropism. Lowering the atmospheric air humidity inhibited root elongation and caused root curvature toward the moisture-saturated substrate in ageotropum pea. Removal of root tips approximately 1.5 mm in length blocked the hydrotropic response. A computer-assisted image analysis showed that the hydrotropic curvature in the roots of ageotropum pea was chiefly due to a greater inhibition of elongation on the humid side than the dry side of the roots. Similarly, gravitropic curvature of Alaska pea roots resulted from inhibition of elongation on the lower side of the horizontally placed roots, while the upper side of the roots maintained a normal growth rate. Gravitropic bending of Alaska pea roots was apparent 30 min after stimulation, whereas differential growth as well as curvature in positive root hydrotropism of ageotropum pea became visible 4–5 h after the continuous hydrostimulation. Application of 2,3,5-triiodobenzoic acid or ethyleneglycol-bis-(β-aminoethylether)-N,N,N',N'-tetraacetic acid was inhibitory to both root hydrotropism of ageotropum pea and root gravitropism of Alaska pea. Some mutual response mechanism for both hydrotropism and gravitropism may exist in roots, although the stimulusperception mechanisms differ from one another.

    Notes: The amino acid sequences of the large polypeptides of thiamin-binding proteins (TBPs) from sesame (Sesamum indicum L.) seeds (STBP-I, -II and -III) were analyzed. The large polypeptides of STBP-I, -II and -III had the same amino acid sequences as did their small polypeptides. The peptide sequence information obtained from STBPs was used to synthesize DNA primers for amplification of the gene(s) encoding STBPs. A 200-bp fragment was amplified from cDNA synthesized from RNA from sesame seeds 4 weeks after flowering. The 200-bp fragment was used to clone full-length cDNA(s) encoding STBP(s) with RACE techniques. A 644-bp fragment was amplified, cloned and sequenced. The cDNA was a full-length clone encoding STBP(s). It contained an open reading frame, which defined a 143-residue polypeptide. The identified small and large polypeptide sequences of STBPs exactly matched the sequence encoded within the cDNA clone. These results indicated that the small and large polypeptides of STBPs were encoded on the mRNA as a single large proprotein precursor and that the final mature forms were generated by post-translational processing in the same manner as the other 2S albumins of plant seeds.

    Notes: We previously demonstrated that γ-glutamyl aminopeptidase (also called γ-glutamyl transpeptidase) (GGT) of Neisseria meningitidis is involved in the bacterial multiplication in cerebrospinal fluid. To further understand the function of meningococcal GGT, the biochemical properties were investigated in this study. The deduced amino acid sequence in N. meningitidis GGT was 37% identical to that of Escherichia coli GGT and that of Helicobacter pylori GGT, respectively, while a typical signal sequence was not found at the N-terminus of meningococcal GGT. Western blotting using rabbit antiserum against recombinant meningococcal GGT protein demonstrated that the meningococcal GGT is processed into two subunits in N. meningitidis at the conserved amino acid, threonine 427. The experiments on subcellular fractionation suggested that the majority of meningococcal GGT is associated with inner membrane facing to the cytoplasmic side. This cell localization might be unique for N. meningitidis compared to other GGTs.

    Notes: We have examined the graviresponding sites in the shoots of seedlings of rice (Oryzasativa L.) and their relation to the agravitropic growth of a‘lazy’line. The graviresponding sites of the seedling shoots of a japonica type of rice. cv. Kamenoo. shifted from the mesocotyl/coleoplile region to the leaf-shealh base when the shoots grew in the dark. A lazy line ot rice. lazy-Kamenoo. showed gravicurvature in the mesocotyl/ coleoptile region at the early stage of growth, hut eventually lost its graviresponse as the seedlings grew. The loss of graviresponsiveness of lazy-Kamenoo was attributed to a reduced response of the coleoptile and a diminished response of the leal-sheath base to gravity. Later. the leaf-sheath base of lazy-Kamenoo became gravitropically incompetent. causing agravitropic growth of the shoots. Thus, shoots of lazy-Kamenoo lose graviresponsiveness in an organ-dependent fashion.

    Notes: Mechanical perturbation (MP) applied unilaterally to cucumber (Cucumis sativus L.) hypocotyls induced thigmotropic curvature toward the stimulus. Gravitropic or phototropic curvature of the hypocotyl was inhibited by symmetrical application of MP to both sides of the hypocotyl. When both MP and IAA were unilaterally applied simultaneously to the same side, the hypocotyls always bent toward the MP stimulus, as in thigmotropism alone. Thus, the exogenous IAA did not control the direction of curvature. Aminoethoxyvinyl glycine (AVG) blocked thigmotropism as well as gravitropism and phototropism, but promoted IAA-induced curvature. MP-stimulated ethylene evolution peaked about 4 h after MP, followed by a peak of thigmotropic curvature. For all tropisms more ethylene evolved from the stimulated side than from the other side of the hypocotyls. MP-induced ethylene acting as a growth inhibitor, auxin-transport inhibitor, and/or modulator of tissue sensitivity to auxin, may be involved in thigmotropism and MP-induced inhibition of various tropisms. Ethylene produced as a result of MP was not affected by the removal of cotyledons. This MP-induced ethylene was additive to that of phototropically or gravitropically stimulated ethylene.

    Notes: An extracellular solution obtained from bean (Phaseolus vulgaris L. cv. Resistant Cherokee Wax) stems induced phytoalexin-like substance and ethylene production in a soybean [Glycine max (L.) Merr. cv. Wayne] cotyledon bioassay. The elicitor-like activity for phytoalexin formation and ethylene production was increased by mechanical perturbation of bean stems. Moreover, the application of extracted or known elicitors to bean plants mimicked the effect of mechanical perturbation (i.e., inhibition of stem elongation and enhancement of radial growth). The effects of extract when applied exogenously, on elicitor-like activity in the bioassay as well as stem thickening were decreased by aminoethoxyvinylglycine, an inhibitor of ethylene biosynthesis. These results suggest that elicitor-like substances which are formed in response to mechanical perturbation contribute to the thigmomorphogenesis.

    Notes: The shoots of a Japanese strain of morning glory (Pharbitis nil ) called ‘Shidare-asagao’ display agravitropic and weeping growth. It has been shown that this shoot agravitropism may be due to the defective differentiation of endodermal cells that contain statoliths. Roots of the weeping morning glory show normal responsiveness to gravity and the shoots are positively phototropic. Shoots of the morning glory cultivar Violet used as a wild type exhibited distinct circumnutation with circular movements that increase as the plants grow. In weeping morning glory, however, nutation was limited to slight back and forth or side to side movements. To determine whether endodermal cells participate in circumnutation through a function that is independent of their role in gravitropism, the nutational movements of various gravitropic mutants of Arabidopsis thaliana were compared. The inflorescences of wild-type Arabidopsis showed relatively large circular movements. Inflorescences of the pgm-1 mutant, which is defective in starch synthesis, showed reduced nutation. Even more seriously affected were the sgr1-1/scr-3 and sgr7-1/shr-2 mutants, which are defective in endodermal cell differentiation, and the auxin-resistant axr2-1 mutant showed no significant nutational movements at all. 1-N-naphthylphthalamic acid (NPA) could inhibit Violet circumnutation, supporting the notion that auxin participates in circumnutation. Thus, the gravitropic response is an essential component in plant shoot circumnutation. Endodermal cells are involved in such circumnutation possibly because of their role in inducing the gravitropic response.
