    Notes: Summary The pathogenesis of disturbances of the initiation and conduction of the cardiac impulse in cardiac amyloidosis has been a matter of controversy. In this study, we have analyzed the histopathology of the sinoatrial node and the atrial myocardium in nine cases of familial amyloidosis with polyneuropathy. Our results indicate that in this disease, amyloid infiltration of the sinoatrial node and the atrial musculature per se may account for the electrophysiological disturbances of these regions.

    Notes: Summary The expression and distribution of cytokeratins and vimentin in fifteen malignant salivary neoplasms were examined by immunocytochemical techniques using, five monoclonal antibodies (mAbs) against different epitopes of Cytokeratins (CKs) (mAbs PKK1, PKK2, and PKK3, identifying CKs 8, 18 and 19, CKs 7, 17 and 19, and CK 18, respectively) and Vimentin (mAbs V9 and V24). Antibody PKK1 gave strong reactions in all neoplasms showing the similarity of these tumours to other digestive system adenocarcinomas. Three general staining patterns of the neoplasms were recognized with respect to the reactivity of mAbs PKK2, PKK3, and V9. Mucoepidermoid cancer, salivary duct carcinoma and a clear cell carcinoma had a higher relative content of CKs 7, 17 and 19 than of CK 18. Adenoid cystic carcinoma showed the same CK pattern but in the periphery of the tumour cords vimentin was readily detected. In two acinic cell carcinomas, the relative content of CK 18 was higher than that of CKs 7, 17 and 19. Furthermore vimentin was expressed in the tumour cells. However, one mucoepidermoid carcinoma showed vimentin expression and two acinic cell carcinomas were vimentin negative and more reative for PKK2 than PKK3. Pecularities in CK expression were seen: squamous areas of mucoepidermoid carcinomas were stained by mAb PKK3 although CK 18 is not present in normal squamous epithelia or in squamous cell carcinomas of tongue and skin. In conclusion, the different salivary neoplasms can be distinguished on basis of IFP content. Such a differentiation fits with current theories of histogenesis, i.e. vimentin is seen in tumours presumed to arise from intercalated duct reserve cells, whilst the vimentin negative neoplasms would be expected to arise in excretory duct reserve cells.

    Notes: Summary A comparative ultrastructural study of bovine Purkinje fibres and ordinary myocytes during fetal development has been undertaken. Differences between the two cell types with respect to the intercalated disc, amount of myofibrils, arrangement of mitochondria, amount of glycogen and formation of T-tubules became apparent gradually. In all stages studied an abundance of intermediate filaments was typical for the Purkinje fibres. Myofibrillar M-bands developed at an earlier stage in Purkinje fibres than in ordinary myocytes. Myofilament-polyribosome complexes typical of adult cow Purkinje fibres were not observed in the fetal hearts. Only in late fetal stages were leptofibrils observed in both cell types. We conclude that in the bovine heart Purkinje fibres develop along a different pathway from ordinary myocytes.

    Notes: Summary In the bovine fetal heart, subendocardial bundles of cells could be distinguished from the main myocardial mass. Their morphological characteristics suggest that they represent bundles of Purkinje fibres. An intense fluorescence after incubation in antisera against the intermediate filament protein skeletin also supports this suggestion. Further, the bundles exhibited different histochemical reactions from the main myocardial mass. During development the histochemical pattern changed. Bundle cells in mitosis were observed. With increasing fetal age, binucleate cells were seen progressively more frequently. Our observations indicate that the Purkinje fibres differentiate along a line separate from the ordinary myocardial cells and that they acquire their adult characteristics gradually.

    Notes: Summary Adult and developing salivary glands were investigated using five monoclonal antibodies against cytokeratins (CKs) and vimentin. Acinar cells displayed mainly CK 18 whereas CKs 7, 17 and 19 were only detected in duct and myoepithelial cells. All epithelial and myoepithelial cells were unreactive for one vimentin antibody (Vim 9) whereas with the other (Vim 24), myoepithelial cells and basal cells of excretory ducts were stained. Fetal cells showed the CK pattern of duct cells. At gestational week 18, a reaction for both vimentin antibodies could be found in basal cells of terminal tubules. Although vim 9 reactivity has been shown for a number of salivary neoplasms, it has not been detected in any adult epithelial salivary tissue. The finding of this reactivity in the fetal gland indicates that the expression of this intermediate filament protein in certain salivary neoplasms may be a sign of dedifferentiation resulting in the expression of a filament pattern found in an earlier stage of gland development.

    Notes: Abstract Two female siblings with the fetal akinesia deformation sequence (FADS) are described. Both showed facial anomalies, arthrogypotic extremities, hypoplastic lungs, and fetal growth retardation. The central nervous system of the second sibling, including the spinal cord, was normal. The skeletal muscle was studied by immunohistochemistry for the expression of several myosin heavy chain isoforms, M-band proteins and intermediate filament proteins. The skeletal muscle was immature and atypical muscle spindles containing up to 31 intrafusal fibers were found. These findings suggest that a lethal FADS phenotype may involve a maturation defect of the skeletal muscle, and, in this family, may be inherited in a recessive fashion.

    Notes: Abstract  The distribution of intermediate filament proteins in adult human blood vessels and in human fetal elastic arteries is relatively well-known. However, the distribution of these proteins in the course from neonate to adult has not been established. In this investigation, human postnatal arteries were studied with immunohistochemistry, using antibodies targeted on the intermediate filament proteins desmin, vimentin and cytokeratins 8, 18 and 19. Vimentin was present in most smooth muscle cells in all vessels and at all ages. The proportions of desmin-expressing cells increased in the elastic arteries during the first year of life and was higher in the pulmonary trunk than in the aorta. In the muscular arteries, the proportion of desmin-labelled cells increased in the coronary and the deep femoral arteries, but remained constant in the renal and the cerebral arteries. Cytokeratins were detected in the pulmonary trunk earlier than in the aorta. Cytokeratins were present throughout the wall of the ductus arteriosus, but desmin was present only in some cells. Thus, there are postnatal changes in the distribution of intermediate filament proteins in the elastic arteries and in some muscular arteries, whereas the intermediate filament pattern remains unchanged in other muscular arteries.

    Notes: Abstract  The aim of this investigation was to determine whether 10 weeks of three different types of training can alter the myosin heavy chain (MyHC) composition of the trapezius muscle. Twenty-one women were randomly assigned to three training groups that performed strength (n=9), endurance (n=7) or coordination training (n=5). Pre and post biopsies were taken from the upper part of the descending trapezius muscle and were analysed for MyHC isoform content using 5% gel electrophoresis. In addition, we have studied the expression of embryonic and neonatal MyHCs using double-immunofluorescence staining. In the strength-trained group, there was a significant increase in the amount of MyHC IIA and a significant decrease in the amount of MyHC IIB and MyHC I. In the endurance group, there was a significant decrease in the amount of MyHC IIB. MyHC composition in the coordination group was not altered. Following the training period, myotubes and individual small-sized muscle fibres were observed in the strength and endurance trained groups. These structures were stained with the markers for early myogenesis (MyHC embryonic and neonatal). These data suggest that specific shifts in MyHC isoforms occur in the trapezius muscle following strength and endurance training. The presence of small-sized muscle fibres expressing the developmental isoforms of MyHC suggests that strength and endurance training induced the formation of new muscle fibres.
