    Notes: Abstract: The six neurofilament proteins (NFPs) in the goldfish Mauthner axon (M-axon) have molecular sizes of 235, 145, 123, 105, 80, and 60 kDa. To determine if NFPs in the M-axon are phosphorylated, isolated Mauthner axoplasm (M-axoplasm) and a neurofilament-enriched extract (NFE) prepared from M-axoplasm were incubated with 32P, which resulted in the radiolabeling of NFPs as determined by their detection on autoradiograms. Kinase inhibitors directed against cyclic AMP-dependent kinases (PKAs) or cofactor-independent kinases significantly reduced the in vitro phosphorylation of NFPs in NFE, whereas inhibitors directed against protein kinase C did not significantly reduce the in vitro phosphorylation of NFPs in NFE. Experiments using two kinase inhibitors directed against different kinases significantly reduced the in vitro phosphorylation of NFPs in NFE to a greater extent than the reduction produced using any single kinase inhibitor. These data suggest that NFPs in the M-axon are phosphorylated and that the in vitro (and perhaps the in vivo) phosphorylation of NFPs is mediated by PKA and/or cofactor-independent kinases that copurify with NFPs.

    Notes: In situ x-ray photoelectron spectroscopy (XPS), etch rate measurements, and optical emission spectroscopy have been used to examine the etching characteristics of tungsten in CF4/O2 reactive ion etching plasmas. It is found that the etch rate maximum of tungsten occurs at a proportion of oxygen in excess of that required to produce the maximum gas phase fluorine atom concentration, and this cannot be explained by using an etch mechanism model similar to that developed for silicon. XPS results have been used to identify tungsten oxyfluoride (WOF4) on the etched surface, and a model for tungsten etching is proposed that involves tungsten oxyfluoride as an important etch product.

    Notes: Reactive ion etching of Si and SiO2 in SF6 plasmas in which the samples are mounted on a liquid-nitrogen-cooled electrode has been studied. At this temperature SF6 condenses on the electrode surface, but it is possible to maintain a plasma. Si etch anisotropy has been demonstrated at low temperature, in agreement with previous studies. Mass spectrometry and optical emission spectroscopy indicate that fluorine is the dominant species in the plasma because SF6 and SFx species are removed from the gas phase by condensation.

    Notes: We have studied reactive ion etching of Si1−xGex alloys with x≤0.15 and Ge in HBr plasmas. The etch rate of SiGe increases monotonically with the Ge content of the alloy and for a Si85Ge15 alloy was (approximately-equal-to)50% greater than for Si. Etch profiles are identical to those formed in singe-crystal Si. X-ray photoelectron spectroscopy studies shown that the surface of the etched SiGe alloys are depleted in Ge and consist of about one monolayer of brominated Si over the alloy.

    Notes: Uniform arrays of approximately 57 nm diam free-standing quantum dots have been produced from GaAs/AlGaAs single quantum well material by electron-beam lithography and low damage electron cyclotron resonance plasma etching. Low-temperature (5 K) photoluminescence and photoluminescence excitation spectroscopy were used to characterize the material before and after processing into quantum dots. Clear free-exciton features of linewidth comparable to that obtained from the unprocessed material have been observed in the excitation spectra of the quantum dots. As expected for this size of dot, no significant shift in the wavelength of the luminescence was observed, however, there is an apparent enhancement of the external luminescence efficiency when the geometric fill factor is taken into account. The results also show that luminescence efficiency measurements seeking to identify and elucidate intrinsic 0D effects (i.e., those due to quantum confinement in the active region) should be performed with photoexcitation directly into the active region energy states. © 1995 American Institute of Physics.

    Notes: Serotonin 5-HT2C receptors (5-HT2CRs) are almost exclusively expressed in the CNS, and implicated in disorders such as obesity, depression, and schizophrenia. The present study investigated the mechanisms governing the coupling of the 5-HT2CR to the extracellular signal-regulated kinases (ERKs) 1/2, using a Chinese hamster ovary (CHO) cell line stably expressing the receptor at levels comparable to those found in the brain. Using the non-RNA-edited isoform of the 5-HT2CR, constitutive ERK1/2 phosphorylation was observed and found to be modulated by full, partial and inverse agonists. Interestingly, agonist-directed trafficking of receptor stimulus was also observed when comparing effects on phosphoinositide accumulation and intracellular Ca2+ elevation to ERK1/2 phosphorylation, whereby the agonists, [±]-2,5-dimethoxy-4-iodoamphetamine (DOI) and quipazine, showed reversal of efficacy between the phosphoinositide/Ca2+ pathways, on the one hand, and the ERK1/2 pathway on the other. Subsequent molecular characterization found that 5-HT-stimulated ERK1/2 phosphorylation in this cellular background requires phospholipase D, protein kinase C, and activation of the Raf/MEK/ERK module, but is independent of both receptor- and non-receptor tyrosine kinases, phospholipase C, phosphoinositide 3-kinase, and endocytosis. Our findings underscore the potential for exploiting pathway-selective receptor states in the differential modulation of signaling pathways that play prominent roles in normal and abnormal neuronal signaling.

    Notes: The seasonal distributions of humpback and blue whales (Megaptera novaeangliae and Balaenoptera musculus, respectively) in the North Atlantic Ocean are not fully understood. Although humpbacks have been studied intensively in nearshore or coastal feeding and breeding areas, their migratory movements between these areas have been largely inferred. Blue whales have only been studied intensively along the north shore of the Gulf of St. Lawrence, and their seasonal occurrence and movements elsewhere in the North Atlantic are poorly known. We investigated the historical seasonal distributions of these two species using sighting and catch data extracted from American 18th and 19th century whaling logbooks. These data suggest that humpback whales migrated seasonally from low-latitude calving/ breeding grounds over a protracted period, and that some of them traveled far offshore rather than following coastal routes. Also, at least some humpbacks apparently fed early in the summer west of the Mid-Atlantic Ridge, well south of their known present-day feeding grounds. In assessing the present status of the North Atlantic humpback population, it will be important to determine whether such offshore feeding does in fact occur. Blue whales were present across the southern half of the North Atlantic during the autumn and winter months, and farther north in spring and summer, but we had too few data points to support inferences about these whales' migratory timing and routes.

    Notes: Accurate identification of humpback whales from photographic identification data depends on the quality of the photographs and the distinctiveness of the flukes. Criteria for evaluating photographic quality and individual distinctiveness were developed involving judgments about overall quality or distinctiveness and about specific aspects of each. These criteria were tested for the level of agreement among judges. The distinctiveness scheme was tested for the independence of distinctiveness judgments and photographic quality. Our results show that judges could agree when evaluating specific and overall aspects of photographic quality and individual distinctiveness. The level of agreement varied for different pairs of judges, and less adept judges were identified. Ability to agree on evaluations of photographic quality was independent of the experience of the judges. Overall photographic quality and overall distinctiveness were successfully predicted from more specific variables, but the agreement between judges for these was not significantly greater than the agreement for the overall measures judged directly. There was no correlation between individual distinctiveness and photographic quality for four of the five judges, but the power of this rest may be low. Analyses of photographic identification data frequently require evaluations of photographic quality and individual distinctiveness. To obtain reliable results from such analyses, evaluation schemes and judges should be tested to ensure reliable and consistent evaluations.
