    Notes: Summary Hepatocytes isolated from liver tissue taken by biopsy from 18 patients with hepatic or extrahepatic diseases displayed a weight class-organization similar to that of other animal species. In most cases the cell classes had a period of 108 pg varying in number from 4 to 14; cell weight range was 96–432 pg as a minimum (4 classes) and 108–1536 pg as a maximum (14 classes). In 5 cases cell classes showed a period of 120 pg resulting 7–9 in number; cell weight range was 216–960 pg (7 classes) or 216–1200 pg (9 classes). No correlation was found between sex, age, liver histopathology, disease of the patients and the various parameters measured.

    Notes: Summary The amount of DNA per nucleus has been determined in the rat hepatocytes during foetal and neonatal life, with three different techniques (Feulgen photometry, microinterferometry and biochemical method). The results show that the DNA content per diploid nucleus in the hepatocytes of 18-day-old foetuses is 35–40% less than that found in the diploid nuclei of six-day-old and adult animals. The largest increase in the amount of DNA per nucleus occurs in the first three days of life. The number of chromosomes is constant at all ages investigated.

    Description / Table of Contents: Zusammenfassung Gegenstand der vorliegenden Untersuchungen ist die Wirkung von Silymarin auf mengenmäßige Veränderungen des Gesamtfeststoffgehalts der Leberzellen der Ratte sowie auf Veränderungen der Klassenstrukturie rung dieser Zellen bei akuter Vergiftungen mit Phalloidin und α-Amanitin. Phalloidin führt 3 Std nach Verabreichung von ca. 2/5 der LD50 zu merklichen Veränderungen in der Klassenstruktur der Hepatozyten, wobei es zu einer Verlagerung eines großen Teils der Leberzellen in die Intervalle zwischen den einzelnen Klassen kommt. Das Trockengewicht der Leberzellen erfährt eine geringfügige Steigerung. α-Amanitin führt bei einer Dosierung von 1/4 bzw. 1/2 der LD50 nach 3 Std zu einer Herabsetzung der Zahl der Hepatozytenklassen; diese Veränderungen kommen durch Beseitigung der schweren Leberzellen, durch eine Verlagerung der Zellen innerhalb der einzelnen Klassen, durch das Auftreten sehr leichter Zellen und durch die Herabsetzung des Trockengewichts der Hepatozyten um ca. 25% zustande. Bei Verabreichung von Silymarin 30 min vor der Intoxikation kommen die durch Phalloidin und α-Amanitin in einer Dosierung von 1/4 der LD50 herbeige führten Veränderungen nicht zur Geltung. Die Wirkung von α-Amanitin in einer Dosierung von 1/2 der LD50 tritt nur mehr stark abgeschwächt in Erscheinung. Der Effekt von α-Amanitin und Phalloidin sowie die Schutzwirkung von Silymarin auf den Gesamtfeststoffgehalt des Protoplasmas der Leberzellen und auf ihre Klassenstrukturierung werden unter Berücksichtigung unserer Kenntnisse über den Wirkungsmechanismus dieser Substanzen diskutiert.
    Notes: Abstract The effects of silymarin on the total dry mass and class pattern of rat hepatocytes have been studied during acute poisoning by phalloidin and α-amanitine. Phalloidin (2/5 of the LD50) after 3 h causes a marked change in the hepatocyte class pattern due to a displacement of a high percentage of cells in the intervals among classes, while the cell dry mass increases slightly. α-Amanitine (1/4 or 1/2 of the LD50) after 3 h causes a decrease in the number of classes of hepatocytes due to a disappearance of the heavier ones, a displacement of cells in the intervals among classes, an appearance of very light cells, and a decrease by about 25% in the mean dry mass of the hepatocytes. Silymarin, administered 30 min before poisoning, prevents all the changes due to 2/5 of the LD50 of phalloidin and to 1/4 of the LD50 of α-amanitine, and strongly reduces the effects of 1/2 of the LD50 of ⇌-amanitine. The effects of α-amanitine and phalloidin and the protective action of silymarin on the dry mass and class pattern of hepatocytes are discussed.

    Notes: Abstract To test the activity of P-170 glycoprotein in isolated hepatocytes, a method has been developed employing the fluorescent dye rhodamine 123 (R-123). The uptake of R-123 by both freshly isolated and 4-hr-plated hepatocytes depends on dye concentration, time of incubation, and cell number. The efflux of R-123 from cells is inhibited by sodium azide and by verapamil. In standard conditions the efficiency of efflux of R-123 from cells correlates with the relative amount of immunoblottable glycoprotein. The method has been applied to detection of P-170 activity in hepatocytes from animals of different ages as well as from carcinogen-treated animals. The proposed assay appears a simple and adequate tool for the functional assessment of multidrug transporter in liver.

    Notes: Abstract The amount and activity of the multi-drug transport P-glycoprotein (Pgp) have been measured in cultured hepatocytes derived from different rat strains. A marked increase in Pgp, as revealed by Western blotting, occurred 48 h after seeding in hepatocytes from Sprague-Dawley, Wistar and Fischer 344 rats, the last showing the highest value. The addition of dexamethasone (DEX) to culture medium delayed Pgp overexpression in all the strains, proportionally to the protein amount in the absence of hormone. The R-123 functional test for Pgp showed that Fischer 344 hepatocytes had the lowest ability to extrude the fluorescent dye as compared with Sprague-Dawley or Wistar rats. These results suggest that the Fischer 344 rat is more prone than other strains to culture stressing conditions, leading to an overexpression of Pgp that is not necessarily functional.
