    Notes: Both restriction fragment length polymorphism (RFLP) and amplified fragment length polymorphism (AFLP) analyses were employed to map a new source of resistance to powdery mildew in mungbean. Disease scores of an F2 population derived from the cross between a moderately resistant breeding line VC1210A and a susceptible wild relative (Vigna radiata var. sublobata, accession TC1966) showed a continuous distribution and was treated as a quantitative trait. Although no significant quantitative trait loci (QTL) that can explain the variation was detected by QTL analysis based on the reconstructed RFLP linkage map, new marker loci associated with resistance were discovered by AFLP analysis. The RFLP loci detected by two of the cloned AFLP bands are associated with resistance and constitute a new linkage group. A major resistance quantitative trait locus was found on this linkage group that accounted for 64.9% of the variation in resistance to powdery mildew. One of the probes developed in this study has the potential to assist in breeding for powdery mildew resistance in mungbean.

    Notes: A technique has been developed for determining decreases in the availability of individual essential amino acids in a food as a result of processing, cooking or other treatments. Diets containing casein and free L-amino acids were used to establish critical dietary amino acid levels. These critical levels were such that reductions of 20% in the level of any of nine essential amino acids significantly reduced protein utilization as estimated by rat bioassays. When these critical levels were used in evaluating amino acid availability in unheated and heated lactalbumin, each of the nine essential amino acids was found to be affected by the heat treatment.

    Notes: Abstract The fatigue behaviour of alumina, zirconia-toughened alumina (ZTA) and tetragonal zirconia (TZP) have been investigated using three different techniques. Direct push-pull testing has been used to generate both static and cyclic fatigue data. The results clearly show that all the materials studied are susceptible to both static and cyclic fatigue, and that the times to failure under cyclic loading are considerably shorter than under static loads. The fatigue failure origins have been identified and the influence of surface condition on fatigue life has been assessed. The slow propagation of cracks subject to cyclic tensile and compressive loads has been studied using compact tension specimens and tapered double cantilever beam specimens. These investigations have confirmed the existence of cyclic fatigue effects in coarse-grained alumina and have shown the crack increment per cycle (da/dN) to have a power-law dependence on the peak stress intensity factor. A technique, based on repeated indentation, has been used to investigate the propagation of sub-surface cracks subjected to cyclic loading in both fine-grained alumina and ZTA. The results of the investigation suggest that compressive or closure loads on the crack faces are factors which affect the cyclic fatigue crack growth in ceramics. Based on those observations, an explanation is proposed for the mechanical cyclic fatigue effects in the ceramics investigated.

    Notes: Summary Screening for rice germplasm which can establish seedling from flooded soil was conducted using 256 accessions of conserved germplasm from the International Rice Germplasm Center (IRGC) and 404 accessions from the International Network for Genetic Evaluation for Rice (INGER), IRRI. IRGC germplasm represented broad genetic diversity while INGER germplasm involved desirable agronomic characters. Seeds germinated for 2 d were planted at 25 mm depth in seedling trays. The trays were then submerged to a depth of 30–50 mm. Seedling establishment was evaluated by analyzing leaf development, seedling height, and percentage establishment 15 d after planting. Eight percent and 2% of IRGC and INGER germplasm, respectively, were identified statistically as superior to the control semidwarf IR varieties. Among the superior germplasm were those from Northeast India and Bangladesh which were adapted to deepwater and early summer rainfed lowland cultures. These could be utilized as parents in breeding programs which aim to develop varieties suitable for direct seeding technology (i.e., germinated seeds are sown under the surface of flooded soil).

    Notes: Abstract To better understand the genetic diversity of the wild relatives of rice (Oryza sativa L.) in the O. officinalis species complex repetitive DNA markers were obtained from the diploid species of this complex. One cloned sequence from O. eichingeri gave intense hybridization signals with all species of the O. officinalis complex. This 242 bp clone, named pOe.49, has a copy number from 0.9 to 4.0 × 104 in diploid species of this complex. Analysis of the primary structure and database searches revealed homology of pOe.49 to a number of sequences representing part of the integrase coding domain of retroviruses and gypsy-like retrotransposons. Sequencing of specific PCR products confirmed that pOe.49 is part of a gypsy-like retrotransposon. RFLP analysis was used to study the genomic organisation of pOe.49 among 30 accessions of the O. officinalis complex using 10 restriction enzymes. Diversity analysis based on 120 polymorphic fragments obtained from the RFLP assay grouped the O. officinalis complex accessions by genome, species and eco-geographic groups. The results suggest that, with further characterization, this retrotransposon-like DNA sequence may be useful for phylogenetic analysis of species in the O. officinalis complex.
