    Notes: Abstract: Bradykinin, which activates polymodal nociceptors, increased cyclic GMP (cGMP) in a capsaiciiji-sensitive population of cultured sensory neurones from rat dorsal root ganglia (DRG) by stimulating guanylate cyclase, but had no effect on cyclic AMP (cAMP). In nonneunbnal cells from DRG, bradykinin increased cAMP, but not cpMP. The bra-dykinin-induced increase in cGMP in thej neurones was completely blocked by removal of extracellular Ca2+, or by incubation of the cells with the calcium channel blockers nifedipine and verapamil. Pretreatment of the| neurones with either dibutyryl cGMP or sodium nitroprusside (which elevates cGMP) inhibited bradykinin-induced formation of inositol phosphates. It is possible that cGMP could be involved in the regulation of polyphosphoinositide turnover in DRG neurones.

    Notes: Abstract: Capsaicin, which induces fluxes of sodilim, calcium, and potassium ions in a subset of both neonatal and adult rat dorsal root ganglion neurones, increased cyclic GMP (cGMP) levels by a factor of 20 (EC50 0.07 μ) to 10-20 pmol cGMP/mg protein in these cells. Cyclic AMP (CAMP) levels were unaffected. Nonneuronal cells derived from rat ganglia, and both neurones and nonneuronal cells from chick were unresponsive to capsaicin. Capsaicin-induced cGMP elevation in rat dorsal root ganglion (DRG) neurones was unaffected by pertussis toxin, lowered by compounds that block voltage-sensitive calcium channels, and was abolished by the removal of extracellular calcium. Calcium, guanidine, and rubidium fluxes were unaffected by treatment of DRG cells with sodium nitroprusside or dibutyryl cGMP. The cGMP response to capsaicin is thus a function of capsaicin-evoked calcium uptake through voltage-sensitive calcium channels. Elevated cGMP levels do not, however, contribute to capsaicin-evoked ion fluxes or to their desensitisation.

    Notes: Summary Dry matter productivity under saline conditions was compared in 5 desiccation-tolerant “resurrection” grasses and one desiccation sensitive species, all in the genus Sporobolus. S. stapfianus was the most salt tolerant, requiring 215 mole NaCl m-3 to reduce shoot dry matter increments to 50% of increments in plants not treated with salt. (This was comparable to published values for the salt tolerant grass Diplachne fusca.) S. lampranthus was salt sensitive, requiring 35 mol m-3 for 50% control yields. S. festivus, S. aff. Fimbriatus, and the deisccation sensitive S.pyramidalis was moderately tolerant (150–170 mol m-3). The moderate salt resistance of S. aff. fimbriatus was attributed mainly to exclusion of NaCl by roots. Salt export through leaf surfaces was a minor factor. Half of the leaf mesophyll cells survived 50 min immersion in 200 mol NaCl m-3. Plants of S. aff. fimbriatus and S. pyramidalis tolerated a broad range of soil pH. Plants of 4 desiccation tolerant Sporobolus species survived air-dryness following 3 weeks pretreatment with salinities up to 200 mol m-3

    Notes: Summary A new monoclonal antibody (mab) against neurofilaments is described (mab 1215) and its reactions compared with previously characterized mabs (BF10; RT97). Mab 1215 recognizes an epitope on the heavy neurofilament polypeptide (NF-H). In Alzheimer's disease, mab 1215 recognizes only a subpopulation of neurofibrillary tangles and stains a proportion of tangles in the hippocampus but none of those in the olfactory bulb. However, mabs RT97 and BF10 stain the majority of tangles in both brain areas. Of the three antibodies, only mab BF10 recognizes, specifically, axons of granular cells in the dentate gyrus of the hippocampus. Mab 1215 recognizes more dendrites in the pyramidal layer than either mab BF10 or mab RT97. Our observations indicate that neurofilaments are not identical in all axons and that, contrary to previous reports, NF-H is present in dendrites. The dendritic form of NF-H appears to be different from NF-H in axons and this could be due to differences in the state of phosphorylation of NF-H. We suggest that the finding that distinct subpopulations of tangles exist indicates that tangles are not static lesions. Further investigations into this possibility may illuminate the pathophysiology of Alzheimer's disease.

    Notes: Early postnatal mouse dorsal root ganglion neurons were found to express several glycosylphosphatidylinositol-anchored (GPI) molecules from the immunoglobulin superfamily (neural cell adhesion molecule 120 kD isoform, F3, Thy1) whose expression is developmentally regulated. A hybrid cell line (ND26), made by fusing postmitotic rat dorsal root ganglion (DRG) neurons with the mouse neuroblastoma N18Tg2, could be induced to differentiate by manipulating the composition of the culture medium and expressed similar GPI molecules to DRG neurons. We used this model system to investigate the metabolism of GPI-anchored molecules. We found that neural cell adhesion molecule 120 Kd isoform expression decreased upon differentiation, whereas the level of F3 and Thy1 increased, suggesting a role in neurite outgrowth processes.The ratio of molecules cleavable by exogenous phosphatidylinositol phospholipase C (PI-PLC) was similar for all the GPI-anchored molecules, which could mean that cell-specific modifications of the basic anchoring structure determine the level of potentially releasable molecules. Measurements of spontaneous release indicated that this reflected the overall level of expression of these molecules by the ND26 cell line.Finally, we observed an effect of dibutyryl cAMP on the level of expression of F3 and Thy1 but not of N-CAM. However, we could not detect any significant effect of nerve growth factor (NGF) either on the level of expression or on the amount of spontaneously released molecules.
