    Notes: A method for the determination of a focused laser beam waist via photoionization and retarding potential analysis is discussed. The focused laser beam was employed to photoionize gas phase clusters generated from a supersonic cluster source. It intersects the cluster beam at right angle in the first stage of the time-of-flight (TOF) acceleration lens assembly. With a simple modification on a laser-based TOF mass spectrometer, retarding fields were applied right in front of the microchannel plate ion detector to map out the kinetic energy distribution of the cluster ions. The size of the laser beam along the molecular beam axis in the ionization region can then be directly related to this energy distribution. In addition, this method manifests a sensitive probe (within 10 μm) to the laser beam position.

    Notes: Photodepletion spectra (415–763 nm, 10 A(ring) effective resolution) were obtained for LiNa3 and Li2Na2 and compared to previous Na4 and Li4 measurements as well as to model calculations ranging from classical electrostatic to ab initio molecular orbital. Findings include (i) the assignment of planar rhombic singlet ground states for both molecules; (ii) in Li2Na2, the possibleobservation of predominantly one of three energetically close-lying planar rhombic or near rhombic substitution isomers; and (iii) in the sodium/lithium tetramer series, evidence for increasing localization of itinerant valence electrons with increasing cluster lithium content.

    Notes: Photodepletion spectroscopy was used to obtain electronic absorption data for Na5, Na6, and Na7 in a wavelength range from 420–770 nm. Measurements were rationalized using several models, ranging from classical electrostatic to ab initio large-scale configuration-interaction calculations. As in previous studies of Na4, Na8, and Na20, classical Mie–Drude theory applied to appropriately dimensioned metal droplets describes the gross but not a number of fine-structure features of the spectra observed. In contrast, ab initio large-scale configuration-interaction predictions of vertical electronic excitations from specific ground-state geometries can more fully account for the measurements. Theory–experiment comparison suggests that Na6 is predominantly formed under our molecular-beam conditions in a planar or near-planar structure. In contrast, the spectrum obtained for Na7 is consistent with a three-dimensional topology.

    Notes: Collinear molecular beam photodepletion was used to obtain particle specific electronic absorption information for Na3, Na4, and Na8 in a wavelength range from 370–835 nm. We critically discuss the experimental method used and the deconvolution procedure applied to the resulting data to yield absolute absorption cross sections. The spectra contain much information on the cluster-size–dependent transition from molecular to bulk-like optical response and are interpreted in terms of various computational approaches ranging from classical electrostatic to ab initio large scale configuration interaction.

    Notes: Sequential reactions of SiD0–3+ and Si2D0–6+ with 10−7–10−6 Torr of disilane are described. The reactions proceed, with few exceptions, by addition of SiD2 with simultaneous loss of SiD4. The growing cluster cations decrease in reactivity with increasing size. For all reaction sequences, a terminal cluster size is reached that contains fewer than nine silicon atoms. Based on our results, we conclude that the reaction of small subsilane or disilane cations with disilane does not lead to gas phase particle formation in disilane plasmas.

    Notes: A soluble form of cytotoxic T-lymphocyte-associated antigen-4 (sCTLA-4) was recently found and shown to possess a downregulatory function as a membrane-bound CTLA-4 molecule. The purpose of the study was to investigate the expression of sCTLA-4 molecule in patients with systemic lupus erythematosus (SLE). One hundred patients with SLE and 40 age- and sex-matched healthy individuals were enrolled in the study. The results showed that patients with SLE have significantly higher levels of sCTLA-4 in sera than healthy controls (21.6 ± 12.3 ng/ml versus 5.9 ± 5.4 ng/ml, P 〈 0.001). Increased expression of sCTLA-4 mRNA in peripheral blood mononuclear cells (PBMCs) was also found in SLE patients. However, we could not find a statistically significant correlation between the serum levels of sCTLA-4 and lupus disease activities. The reported CTLA-4 gene polymorphism in promoter region at position −318 did not affect the levels of sCTLA-4. To the best of our knowledge, this is the first report showing that patients with SLE have increased sCTLA-4 expression. However, the mechanism and role of increased sCTLA-4 in the pathogenesis of SLE remains elucidated.

    Notes: CD4+CD25+ T cells have been shown to play a regulatory or suppressive role in the immune response and are possibly relevant to the pathogenesis of autoimmune diseases. In the present study, we attempted to investigate the levels of CD4+CD25+ T cells in the peripheral blood (PB) and synovial fluid (SF) of patients with rheumatoid arthritis (RA) and the effects of CD4+CD25+ T cells on the in vitro cytokine production by stimulated SF mononuclear cells (SFMC). The results showed that RA patients had similar frequencies of CD4+CD25+ T cells in PB, expressed as a percentages of the lymphocyte population, as did healthy subjects (mean ± SD: 10.52 ± 5.87% versus 11.11 ± 4.58%., respectively). But in contrast to PB, the SF of RA patients contained significantly higher levels of CD4+CD25+ T cells (17.77 ± 7.92% versus 10.52 ± 5.87%, respectively. P 〈 0.001). When cocultured in vitro with SFMC, CD4+CD25+ T cells purified from either PB or SF were found to exert a considerable suppressive effect on the production of cytokines including TNF-α, IFN-γ and interleukin-10 (IL-10). The percentages of inhibition of each cytokines ranged from 41.8 to 98.4% (mean, 80.0%) for TNF-α, 42.8 to 98.9% (mean, 83.2%) for IFN-γ and 59.3 to 96.6% (mean, 80.0%) for IL-10. Because both pro-inflammatory and anti-inflammatory cytokines were suppressed by CD4+CD25+ T cells, whether CD4+CD25+ T cells might play a beneficial role in the suppression of sustained inflammation in rheumatoid synovium remains to be elucidated.
