    Notes: Summary A hybridoma cell line (OV632) producing monoclonal antibody against ovarian carcinomas was developed from the spleen cells of a mouse immunized with cystic fluid from a serous cystadenocarcinoma. Immunohistological studies in frozen sections showed that 22 out of 28 nonmucinous ovarian carcinomas, which included serous, endometrioid, clear cell, and undifferentiated tumours, reacted with this antibody. Three out of 7 mucinous ovarian carcinomas were positive, whereas only 7 out of 122 extra-genital malignant lesions, predominantly adenocarcinomas, were positive. The negative cases included 38 breast carcinomas and 24 colon carcinomas, tumours which are responsible for most of metastatic disease in the ovary. On the basis of these findings, the antibody OV632 is considered appropriate for histodiagnostic purposes as an aid in the distinction between primary and secondary ovarian cancer.

    Notes: Summary Monoclonal antibodies recognizing various structures of the nephron were used to stain renal cell carcinomas and associated metastases. Generally, renal cell carcinoma is subdivided into tumors of a clear-cell type, tumors of a mixed-clear-cell/granular-cell type, and tumors in which spindle cells are present. The staining patterns observed with the monoclonal antibodies used correlated in part with the histological appearance of the carcinomas. One monoclonal antibody did not stain other carcinomas, sarcomas and melanomas tested. Notably the same monoclonal antibody stained 75% of primary tumors, whereas only 16% of metastases were stained. This finding indicates that metastases exist which are made up of less well-differentiated tumor cells than the primary tumors.

    Notes: Summary A DNA polymerase present in particles with a density greater than 1.20 g/cm3 and capable of using a synthetic RNA template has been sought in human malignancies. This report deals with a study of a great number of peripheral-blood samples from normal controls and patients with malignant and non-neoplastic haematological disorders. For screening purposes a simplified detection test was used. In 63 controls low levels of enzyme activity were found. The enzyme activities showed a biphasic distribution pattern. Three out of 53 patients with non-neoplastic, miscellaneous haematological disorders had an elevated enzyme level associated with active viral infections (hepatitis, mononucleosis infectiosa). In 128 patients suffering from haematological malignancies 18 out of 21 cases of elevated enzyme level were associated with the presence of more than 10% pathological cells in the white blood cell fraction.

    Notes: Summary Simultaneous biochemical and electron microscopical investigations on surgically removed spleens yielded evidence for the presence of reverse transcriptase containing (Retra) virus in two patients with hematological malignancies with spleen involvement. In three other patients with hematological diseases and in one control patient, the spleens were negative in both assays. The results of these combined studies support the view, that retraviruses are present in human malignancies.

    Notes: Summary Vaccinia virus was purified by ultrasonic and enzymatic treatment followed by centrifugation through a sucrose gradient. Serumblocking protein was prepared by treatment of the purified virus with potassium hydroxide followed by either high speed centrifugation or filtration. The antigenicity of alkali-treated protein was compared with that of serumblocking antigen prepared according to the method ofAppleyard. Both preparations, when administered by 4 to 6 subcutaneous injections, proved capable of producing haemadsorption-inhibiting antibodies and a certain degree of resistance to challenge with live vaccinia virus in rabbits.

    Notes: Abstract In extracts of spleen tissue from two patients with haemotological malignancies an RNA dependent DNA polymerase was found in particle with a density of 1.16, that is at the density of oncorna viruses. After treatment with noniomic detergents the enzyme activity was found in particles with a density of 1.23–1.24, similar to the density of oncorna viral cores. A simultaneous detection test with this core fraction material for 70 S RNA and RNA dependent DNA polymerase was positive for both patients. Electron microscopical inspection of the material with a density of 1.16 revealed immature C-type virus like particles, various stages of maturing particles and a number of particles resembling mature C-type oncorna viruses. In two normal spleens from patients with carcinoma of the colon and oesophagus respectively and in three spleens from patients with no history of malignancy no RNA dependent DNA polymerase was found. Material from one normal spleen was examined in the electron microscope and no virus-like particles were seen.
