    Notes: Ten-day-old broiler chickens were inoculated with oocysts of a characterized strain of Eimeria mitis, and tissues were fixed at 4, 8, or 24-h intervals after inoculation for histopathological examination. Tissue collections were initiated at the time of inoculation and extended up to 168 h postinoculation. The preferred site of development of E. mitis was found to be the ileum although more limited development of the parasite also took in the jejunum, cecal pouches, cloaca, and bursa of Fabricius. No distinctive and consistent intestinal lesions were macroscopically evident even in heavily parasitized chickens. The prepatent period was approximately 92 h postinoculation. The histopathological features of the E. mitis infections were characterized using conventional bright-field microscopy as well as both scanning and transmission electron microscopy. No extra-intestinal development of the parasite was observed.

    Notes: Sources of pesticides and chlorinated organic compounds potentially found in water sources are reviewed. International and UK drinking water standards for these compounds are discussed and compared with concentrations found in ground and surface waters derived from lowland agricultural and urban areas.Water soluble herbicides including triazines, phenylamides (urons) and chlorophenoxy acids are the most commonly reported types of pesticide compounds found above 0.1 μg/l in water sources. The more toxic, and less water soluble, organo-chlorine and organophosphorus pesticides are rarely reported.Chlorinated organic solvents are reported as contaminants of some groundwaters. Disinfection by-products, in particular trihalomethanes, formed during the chlorination of waters containing natural organics, are widely reported, sometimes at concentrations above current UK standards.The effects of conventional water-treatment processes on the concentrations of these organic compounds are discussed. Additional water-treatment processes, designed specifically for trace organic micropollutant removal, are identified and reviewed. Ozonation, activated-carbon adsorption and air stripping are acknowledged as appropriate advanced water-treatment process solutions. Developments in novel technologies include organics destruction via advanced oxidation processes, biological processes, membrane technology and novel adsorbents.Thames Water Plc is contributing to research in this area, investing over £5 million in pilot- and large-scale trials prior to a £200-£300 million capital investment programme in advanced water treatment in the 1990s.

    Notes: Abstract Acute monocytic leukaemia (M5a) was diagnosed in a 17-year-old Standardbred gelding with lethargy, intermittent pyrexia, oedema of the limbs, harsh lung sounds and submandibular lymphadeopathy. Haematological findings included moderately severe anaemia, thrombocytopenia and a leucocyte count within the reference interval, but characterised by neutropenia and numerous blast cells. Monocytic lineage of the cell population was suggested by examination of Wright-Leishman-stained blood and bone marrow smears. A panel of cytochemical stains disclosed diffuse cytoplasmic α-naphthyl-acetate esterase activity which could be markedly inhibited or abolished in all leukaemic cells by pretreatment with sodium fluoride. In ultrastructural preparations of buffy coat, neoplastic monoblasts had one to two nucleoli, dispersed chromatin, elongated mitochondria, scattered profiles of rough endoplasmic reticulum, bundles of microfilaments and pseudopodia. More differentiated monocytoid cells had infrequent lysosomal granules.

    Notes: Abstract Rapid intravenous adminstration of the glycopeptide antibiotic, vancomycin, may cause a hypotensive reaction which can usually be prevented by infusing vancomycin in dilute solutions. The release of histamine from circulating cells such as basophils and tissue mast cells has been implicated in hypotensive reactions since the effects can be prevented by antihistamine pretreatment. The direct effects of vancomycin on histamine release were therefore investigated in rat peritoneal mast cells and rat leukemic basophils (RBL-1 cells). Suspension cultures of mast cells or RBL-1 cells were exposed to vancomycin for 30–60 minutes at concentrations comparable to those infused clinically (2.28 or 4.56 mg/ml). Vancomycin induced a time- and dose-dependent release of histamine into the culture media from both cell types. The reference degranulating agent, Compound 48/80 (CP 48/80), was also shown to induce histamine release from mast cells and RBL-1 cells. Mast cells were significantly more sensitive to vancomycin and CP 48/80 than RBL-1 cells and, unlike RBL-1 cells, were responsive to the inhibitory effects of cromolyn sodium on histamine release. Cromolyn sodium did not inhibit vancomycin-induced histamine release in RBL-1 or mast cells. Morphologically, mast cells exposed to either vancomycin or CP 48/80 exhibited dose-releated degranulation. On the other hand, treatment-related degranulation effects of either vancomycin or CP 48/80 on RBL-1 cells could not be reliably distinguished from controls by qualitative evaluation. Based upon these findings it is concluded that mast cells may represent a more useful model to evaluate the potential of investigational agents to relase histamine and to study mechanisms of histamine release than RBL-1 cells.

    Notes: Abstract Specimens of the oceanic decapod Systellaspis debilis were collected from six sites in the East Atlantic Ocean between 1970 and 1984, and were analysed for Mn, Fe, Cu, Zn and Cd. The data confirm that there are small but significant differences in mean metal concentrations from some sites which showed no obvious pattern in relation to geographic location of the samples. As a result, ranges of site means are quoted as “baseline” levels for each metal (μg g-1 dry wt): 2.3 to 2.9 μg Mn g-1, 31.2 to 77.8 μg Fe g-1, 25.9 to 83.4 μg Cu g-1, 41.9 to 92.9 μgZn g-1, 11.1 to 31.8 μg Cd g-1. The concentration of cadmium in S. debilis from all sites was raised relative to cadmium concentrations reported for coastal decapods, perhaps as a result of dietary enrichment. Metal accumulation may provide useful information for understanding the complex feeding behaviour of many oceanic animals.

    Notes: Abstract Two pathogenic mitochondrial DNA mutations, a T-to-G substitution (8993T〉G) and a T-to-C substitution (8993T〉C), at nucleotide 8993 have been reported. We describe 13 pedigrees with mitochondrial DNA mutations at nucleotide 8993; 10 pedigrees with the 8993T〉G mutation and three with the 8993T〉C mutation. Prenatal diagnosis of the nucleotide 8993 mutations is technically possible. However, there are three major concerns: (i) that there is variation in mutant loads among tissues; (ii) that the mutant load in a tissue may change over time; and (iii) that the genotype–phenotype correlation is not clearly understood. We have used the 13 pedigrees to determine specifically the extent of tissue- and age-related variation of the two mutations at nucleotide 8993 in the mitochondrial DNA. The tissue variation was investigated by analysing two or more different tissues from a total of 18 individuals. The age-related variation of the mutation was investigated by comparing the amount of both mutations in blood taken at birth and at a later age. No substantial tissue variation was found, nor was there any substantial change in the proportion of either mutation over periods of 8–23 years in the four individuals studied. In addition, we noted that two features were remarkably common in families with nucleotide 8993 mutations, namely (i) unexplained infant death (8 cases in 13 pedigrees), and (ii) de novo mutations (5 of the 10 8993T〉G pedigrees).

    Notes: Abstract This study investigates the comparative strategies of accumulation under standardised laboratory conditions of the essential metals zinc and copper, and the non-essential metal cadmium by three crustaceans of different taxa; vizPalaemon elegans Rathke (Malacostraca: Eucarida: Decapoda),Echinogammarus pirloti (Sexton & Spooner) (Malacostraca: Peracarida: Amphipoda) and the barnacleElminius modestus Darwin (Cirripedia: Thoracica). The decapodP. elegans regulates body zinc concentrations to a constant level (ca. 79 µg Zn g−1) over a wide range of dissolved metal availabilities until regulation breaks down at high Zn availabilities and net accumulation begins. The amphipodE. pirloti accumulates zinc at all dissolved zinc concentrations but at a low net rate such that the accumulation strategy approaches that of regulation. The barnacleE. modestus accumulates zinc to high body concentrations with no significant excretion of accumulated zinc. In the case of copper,P. elegans similarly regulates body copper concentrations to a constant level (ca. 129 µg Cu g−1) over a range of dissolved copper availabilities until regulation breaks down at high copper concentrations. Both the amphipodE. pirloti and the barnacleE. modestus on the other hand accumulate copper at all dissolved copper exposures with no evidence of regulation. All three crustaceans accumulate the non-essential metal cadmium at all dissolved cadmium concentrations without regulation. Heavy metal accumulation strategies therefore vary between crustacean taxa and between metals. Uptake rates for zinc and cadmium have been estimated for the three crustaceans and can be interpreted in terms of cuticle permeability and way of life of each crustacean. Examination of these uptake rates provides an insight into possible reasons behind the adoption of particular metal accumulation strategies.
