    Notizen: Summary The relationship between the formation of microbodies and catalase synthesis in the hepatic cells of male rats was examined with conventional electron microscopy and with the peroxidase staining technic for demonstrating catalase. Daily intraperitoneal injections of ethyl-α-p-chlorophenoxyisobutyrate (CPIB) for 5 days caused a profound increase in microbody numbers without markedly affecting the appearance of the matrix material and all microbodies retained peroxidase activity. A single injection 5 days before sacrifice of 3-amino-1,2,4-triazole (AT), an inhibitor of catalase activity but not catalase synthesis, did not affect their numbers, appearance of matrix material or peroxidase staining. Twice daily injection for 5 days of allylisopropylacetamide (AIA), an inhibitor of catalase synthesis, also did not affect microbody numbers but lowered the electron-density of the microbody matrix and abolished peroxidase staining. After combined administration of these drugs, the number of hepatic microbodies increased but they did not contain peroxidase activity. The results suggest strongly that microbody proliferation is dependent not on catalase synthesis but on synthesis of non-enzymatic protein.

    Notizen: Summary To test the possibility that stimulation of secretion leads Na,K-ATPase to be recruited from cytoplasmic pools and inserted into basal-lateral plasma membranes, we surveyed the subcellular distributions of Na, K-ATPase in resting and stimulated fragments of rat exorbital lacrimal gland. After a two-dimensional separation procedure based on differential sedimentation and density gradient centrifugation, we defined sixdensity windows, which differ from one another in their contents of biochemical markers. The membranes equilibrating inwindow I could be identified as a sample of basal-lateral membranes; in resting preparations these membranes contained Na,K-ATPase enriched 16.6-fold with respect to the initial homogenates.Windows II throughVI contained various cytoplasmic membrane populations; these accounted for roughly 80% of the total recovered Na,K-ATPase activity. Thirty-minute stimulation with 10 μm carbachol caused a 1.4-fold increase (P〈0.05) in the total Na,K-ATPase content ofwindow I; this increase could be largely accounted for by a 1.7-fold decrease in the total Na,K-ATPase content ofdensity window V. Acid phosphatase activity also redistributed following stimulation, but it was recruited from a different source, and it was inserted into membranes equilibrating inwindows II andIII as well as into the membranes ofwindow I.

    Notizen: Summary Three types of subsurface cisterns were observed in Purkinje cells of the cerebellum of the Syrian hamster. The type-1 cisterns are subsynaptic, related to axosomatic synapses, and are separated from the postsynaptic cell membranes with distances of 400–800 Å. These are probably modified rough surfaced endoplasmic reticulum. The type-2 cisterns are closely apposed to the surface membranes of Purkinje cells, and have very little intracisternal space except at the dilated lateral edges. The type-3 cisterns are similar in structure to the type-2 cisterns but in addition are closely associated with mitochondria. The type-2 and type-3 cisterns appear between one and two weeks after birth and are still present in adults, having almost the same frequency of occurrence. Thin cell processes opposite the type-2 and type-3 cisterns are considered to be glial cell processes. The morphological details of these types of subsurface cisterns are described here, and their possible functional significance is briefly discussed.

    Notizen: Summary Living dissociated cells of hydra were exposed to fluorescein- and ferritin-conjugated concanavalin A (con A) and observed by light and electron microscopy. Fluorescence microscopy indicated that the isolated cells bound con A differentially; epidermal battery cells showed the greatest binding, whereas small cells belonging to the interstitial cell class displayed the lowest levels of binding. Mature nematocytes had strong localized con A binding at the opercular region. Electron microscopy permitted accurate identification of interstitial cells, early nematoblasts, and nerve cells. The use of ferritin-labeled con A allowed quantitative assessment of lectin binding on these cells. There were significantly fewer con A-binding sites on interstitial cells as compared to nematoblasts and nerve cells, and the amount of con A binding appeared to increase with the maturation of nematocysts from nematoblasts. The findings are discussed in relation to a likely role of cell surface glycoconjugates in the development of positional signals and intercellular junctions that govern final positioning of nematocytes and nerves in hydra.

    Notizen: Summary The uveal tract of the eyes of monkeys was examined by electron microscopy using both thin sections and freeze-fracture replicas. The ultrastructural features of the lamina fusca in the monkey resembled those previously described for rabbit. The lamina fusca was composed of numerous interleaved processes of fibroblastic and pigmented cells and contained tight junctions between fibroblastic cell processes that were predominantly discontinuous, as well as numerous fenestrations through the attenuated cell processes. There was no regional compaction of cellular processes traversing the entire uvea at the level of the ora serrata as reported previously in hamster eyes.

Emphasis was placed on the organization of pigmented and fibroblastic cells near the ora serrata and more generally in the lamina fusca, since these regions were previously found to restrict ingress of tracer molecules to the choroid in hamster eyes. There were significant differences between rabbit and hamster in the disposition of the pigmented cells and fibroblastic layers, the extent and patterns of tight junctions adjoining fibroblastic cells, and the presence of vesicles or fenestrations in the attenuated fibroblastic cell processes.

    Notizen: Summary The permeability of the uveoscleral outflow pathway from the anterior ocular chamber was examined in rabbit and monkey eyes using anionic ferritin as a tracer. Ferritin, infused intracamerally, had ready access to the choroidal interstitium, and the degree of penetration was generally correlated with the time and pressure relationships during infusion. In both species, there were accumulations of tracer in intercellular spaces at the lamina fusca, but tracer was also present in the sclera. Thus, in contrast to the situation in the eyes of hamsters, the uveoscleral outflow pathway in the eyes of rabbits and monkeys includes the choroidal connective tissue and allows passage of relatively large molecular weight substances.

Myonemes of the hypostome are small in diameter, have gap and intermediate-type cell junctions within each epithelial layer and are associated with the opposite epithelial layer by transmesogleal processes and gap junctions. Nematocysts and sensory cells are aggregated in the circumoral region. The fine structure of adherent flagella arising from gastrodermal gland cells, and the transition region at the mouth between epidermis and gastrodermis are described in detail for the first time.     Notizen: Summary The hypostome and mouth of fresh-water Hydra were examined by scanning electron microscopy. The external surface of the hypostome possesses cnidocils, possibly sensory hairs, and small spiny protrusions surrounding the mouth; the internal surface has cylindrical microvilli, free flagella and adherent flagella. The adherent flagella are most numerous close to the mouth where they cause the cell surface to appear smooth when viewed at low magnifications. Free flagella and leaf-like microvilli increase in prominence towards the tentacles and enter on proper. The edge of the mouth has an abrupt boundary marking the apposition of epidermal and gastrodermal cells. A transitional groove occurs at the boundary and the cells underlying the groove are smaller than those on other regions of the hypostome. The transition groove may represent a site of cell loss in normal cell turnover. Some of the small underlying cells may represent nervous elements involved in regulating hypostome activity during the feeding reation.

    Notizen: Summary Hepatic sinusoids in the calf have been studied by light and electron microscopy. The endothelial lining is shown to be continuous and to be associated with a prominent basement membrane. The perisinusoidal space (of Disse) contains collagen fibrils and is otherwise nearly filled with hepatic cell microvilli. Perisinusoidal cells resembling fibroblasts or reticular cells in morphology and distribution are present in moderate numbers. Hepatic sinusoids in the rat were examined for purposes of comparison. In the rat the sinusoidal lining is discontinuous and there is no identifiable basement membrane. The perisinusoidal space is larger than in the calf and contains more sparsely distributed collagen fibrils. Perisinusoidal cells have not been identified. It is suggested that the difference in structure is not artifactitious but represents a species variation. This species difference seems important to record since observations on the rat and other small laboratory animals provide the basis for most of the current generalizations about the organization of the mammalian liver sinusoid. The functional significance of the two types of sinusoidal arrangement is not clear.
