    Notes: Two different types of BaTiO3 seed particles, normal and twinned seeds of ∼30 μm on the average, were prepared from crushed sintered specimens. Normal seeds were obtained from the usual BaTiO3 sintered compacts, while twinned seeds containing a double twin were obtained from BaTiO3 compacts sintered with 2 mol% of SiO2. The BaTiO3 powder compacts were again prepared with 5 wt% of seed grains and sintered under various conditions. The microstructural evolution was quite different in the two cases: the growth of normal seed grains was ultimately limited but that of the twinned seeds continued extensively. The observed difference is discussed in terms of the growth mechanism and the atomic structure of interfaces.

    Notes: Objective In this study, we measured the levels of eosinophil cationic protein (ECP) and interleukin (IL)-8 in bronchoalveolar lavage (BAL) fluid from patients with acute asthma and acute bronchiolitis, to determine any similarities or dissimilarities in the profiles of these biochemical markers in the two diseases.Methods BAL fluids were obtained from children with acute asthma (n=16), infants with acute bronchiolitis caused by respiratory syncytial virus (n=18), and control subjects (n=14). Children with asthma were selected to be free of viral infection. BAL cell counts and differentials were determined, and ECP and IL-8 levels were measured by radioimmunoassay and ELISA, respectively.Results ECP levels in BAL fluids were significantly higher in the asthma group than in the bronchiolitis (P〈0.01) or control (P〈0.0001) groups. However, IL-8 levels were significantly higher in the bronchiolitis group than in the asthma (P〈0.01) or control (P〈0.001) groups. IL-8 levels in the asthma group and ECP levels in the bronchiolitis group were similar to those of the control group.Conclusion This difference in profiles of ECP and IL-8 in acute asthma and acute bronchiolitis, together with a different inflammatory cell pattern, suggests that the nature of the inflammatory process within the lower respiratory tract may be distinctive in these two diseases.

    Notes: A cDNA clone, PoMTP, encoding a putative metzincin family metalloprotease was isolated from the expressed sequence tags of a basidiomycete Pleurotus ostreatus. The 5′-end sequence of PoMTP was determined by the 5′-RACE method. Full-length cDNA sequence (1140 bp) of PoMTP contained a 870 bp open reading frame encoding a protein product of 290 amino acids in addition to a 99 bp of 5′-untranslated sequence and a 171 bp of 3′-untranslated sequence with a poly(A) tail. The deduced amino-acid sequences of PoMTP contained an extensive zinc-binding consensus sequence and a so-called Met-turn sequence which are typical for the metzincin family of metalloproteases, indicating that the PoMTP protein belongs to the metzincin metalloproteases. Four cysteine residues were also observed in the zinc-binding region of PoMTP amino-acid sequence, which are known to be important for the structure and the function of some subfamilies of the metzincins. Comparison of the PoMTP in sequence database showed no significant homology with functionally known metalloproteases of Armillaria mellea, Grifola frondosa, Lentinula edodes, Pleurotus ostreatus, Schizophyllum commune and Tricholoma saponaceum in mushroom. Northern blot and qunatitative RT-PCR analyses indicated the PoMTP mRNA to be abundant at primordial and fruit body stages, but scarce at the mycelial stage, suggesting that the PoMTP metalloprotease plays an important role in mushroom fruiting.

    Notes: Uracil auxotrophs of Pleurotus ostreatus were isolated using the selectable marker, resistance to 5′-fluoro-orotic acid (5′-FOA). Two of the nine uracil auxotrophs obtained were transformed to prototrophy using plasmid pTRura 3-2 that contains the orotidine monophosphate decarboxylase (ura3) gene from Trichoderma reesei. Southern blot analyses of the transformants showed that the transforming DNA had integrated into the genome of the protoplasts. Using 2×107 protoplasts, this system gave a transformation efficiency of about 30 transformants per μg of DNA. Normal fruiting bodies were induced in the transformants by crossing them with wild-type monokaryons, and the basidiospores collected from these fruiting bodies showed a biased segregation rate to prototrophy. These results indicate the integrated DNA was stably inherited.

    Notes: In a well established murine model relevant to human disease, graft-versus-host disease results from recognition of recipient minor histocompatibility antigens by donor bone marrow-derived T lymphocytes. Previous studies suggest that factor XIIIa-positive dermal dondrocytes may be involved in the pathogenesis of disorders involving antigen presentation to T cells and dermal fibrosis. This study was undertaken to determine (i) whether normal murine skin contains factor XIIIa-positive dermal dendrocytes, and (ii) whether such cells participate in the pathophysiology of acute graft-versus-host disease. Graft-versus-host disease was produced using B10.BR CD8+ donor T cells administered to CBA recipients. Skin samples were collected weekly for a 5-week period and evaluated by immunohistochemistry and electron microscopy. Our data indicate that normal murine dermis contains factor XIIIa-positive cells localized primarily around deep dermal microvessels. Ultrastructural analyses reveal these cells to have long processes, pinocytotic vesicles, fibronexuses, and intimate associations with mast cells. During graft-versus-host disease, factor XIIIa-positive dendrocytes appeared within the superficial dermis. By ultrastructure, the dendrocytes were hypertrophic and highly branched, and demonstrated an intimate relationship with neighboring cells. In conclusion, factor XIIIa-positive dendrocytes comprise a normal component of the murine dermis and undergo alterations in experimental acute graft-versus-host disease consistent with participation in disease pathophysiology.

    Notes: A fatty fish, Atlantic mackerel, and a lean fish, Atlantic cod, were processed and stored under conditions to simulate those of a commercial operation. The fatty acid composition of their lipids was evaluated. All fillets were examined with their skins on. A paired fillet technique was used to reduce the considerable fish-to-fish variability observed; the advantage of this technique was demonstrated. Mackerel fillets were stored on ice, fresh fillets and fillets aged on ice for 8 days were stored at −20C for up to 28 weeks and mackerel mince prepared from fresh and 7-day aged fish were stored frozen at −20C for 1 month. Fresh and aged (8 days) cod fillets were examined initially and after 32 weeks at −20C. No measurable changes were observed in C20:5 and C22:6 fatty acids in either species under these storage conditions.

    Notes: Abstract  The balance between proliferation and cell death is the major determinant of tumour growth. We analysed the proliferative and apoptotic indices (PI and AI, respectively) of 33 children with retinoblastoma. PI and AI were assessed by immunohistochemistry for Ki-67 antigen and TUNEL staining, respectively. The mean PI was 21.0±21.1%, and higher PI was associated with more advanced tumour stage (P〈0.0001) and poor clinical outcome (P〈0.05). Patients in whom amplified N-myc oncogene was found (n=6) determined by the multiplex polymerase chain reaction tended to have a higher PI (37.6±27.2%) than those without amplified N-myc (n=27; PI=17.3±18.1). A PI value of over 40% was clearly associated with an unfavourable prognosis. The AI, however, did not correlate with any of the other variables analysed. The findings suggest that proliferation, but not apoptosis, is of critical significance in retinoblastoma biology. PI, as determined by the Ki-67 antigen labelling index, seems to be a relevant histopathological parameter that can predict the clinical outcome of retinoblastoma.

    Notes: Laser welding process is widely used in the industrial field due to its numerous advantages:a small heat affected zone(HAZ), deep penetration, high welding speed, ease of automation,single-pass thick section capability, enhanced design flexibility, and small distortion after welding. Inthis paper, the laser weldability of Austenite stainless steel and INCONEL600 at dissimilar metalwelds using a continuous wave Nd:YAG laser are experimentally investigated. INCONEL600 isbeing used in a steam generator tubing of pressurized water reactor(PWR) exposed to some corrosion.Therefore stress corrosion cracking can occur on this material. A research work is conducted todetermine effects of welding parameters, on eliminating or reducing the extent welding zoneformation at dissimilar metal welds and to optimize those parameters that have the most influenceparameters such as focus length, laser power and welding speed were tested

    Notes: Extracellular signal-regulated kinases (ERKs) are phosphorylated on threonine and tyrosine residues at 183 and 185, respectively, and then translocated from cytosol to nucleus. ERK2 is retained in the nucleus for several hours by nerve growth factor (NGF), and this sustained retention of ERK2 in the nucleus has effect on the fate of biological response toward differentiation by neurite outgrowth in PC12 cells. The overexpression of Green Fluorescent Protein (GFP)-ERK2and mutated GFP-ERK2 constructs without anchoring protein MEK1 were distributed throughout the resting and the activated cells. When GFP-ERK2 coexpressed along with MEK1, cytosolic localization of GFP-ERK2 is retained by MEK1 in the resting PC12 cells. This cytosolic retention was due to the binding of ERK2 to the MEK1. Upon stimulation by growth factors, the association between GFP-ERK2 and MEK1 was detached from each other, and then GFP-ERK2 wastranslocated into the nucleus. However, inactive form of the MKP-3 cytosolic phosphatase forced ERK cytosolic retention in PC12 cells were either left untreated or stimulated by NGF. When the transfected PC12 cells were treated for 72hrs with NGF, GFP-ERK2 was distributed the cytosol. Regarding its subcellular localization, the roles of residues 179-185 located in the activation loop of ERK2 were examined. The substitution of residues in the activation loop to alanine showed different localization on the nuclear translocation of ERK2 in PC12 cells
