    Notes: Abstract: We investigated the distribution of protein kinase C (PKC)isoforms in the subcellular fractions (P1, 1,000-g pellet; P2,10,000-g pellet; P3, 100,000-g pellet; S, 100,000-g supernatant) of rat forebrain after ischemia or reperfusion by immunoblotting. PKC-δ and -ε isoforms were predominant in the P2 (synaptosome-rich) fraction, whereas PKC-α, -β, -γ, -ε, and -ζ isoforms were rich in the S (cytosolic) fraction. With time of ischemia (5-30 min), PKC-α, -β, and -γ translocated to the P2 and P3 fractions, whereas reperfusion for 60 min after 30 min of ischemia reduced PKC-β activity greatly and PKC-α and -γ activities to a lesser extent. There was no redistribution of PKC-δ, -ε, and -ζ after ischemia or reperfusion. A calpain inhibitor, acetylleucylleucylnorleucinal, inhibited the down-regulation of PKC-β, through intravenous injection. The PKC translocation to the P2 fraction was accompanied by their dephosphorylation, transition of PKC-α from dimer to trimer, and the decrease in activity. These data show that PKC-α, -β, and -γ isoforms translocate chiefly to the synaptosome in ischemic brain in association with the dephosphorylation, multimeric change, and inactivation, followed by the proteolysis of PKC-β by calpain after postischemic reperfusion.

    Notes: Abstract: A membrane cytoskeletal protein, fodrin, is a substrate for a Ca2+-dependent protease, calpain. It remains unknown whether μ-calpain or m-calpain is involved in the proteolysis of either α- or β-fodrin and in what subcellular localization during ischemia and reperfusion of the brain. To address these issues, we examined the distribution of fodrin and calpain and the activities of calpain and calpastatin (endogenous calpain inhibitor) in the same subcellular fractions. Rat forebrain was subjected to ischemia by a combination of occlusion of both carotid arteries and systemic hypotension, whereas reperfusion was induced by releasing the occlusion. Immunoblotting, activity measurement, and casein zymography did not detect the presence of μ-calpain or a significant change of m-calpain level after ischemia or reperfusion. However, casein zymography revealed a unique Ca2+-dependent protease that was eluted with both 0.18 and 0.40 M NaCl from a DEAE-cellulose column. α- and β-fodrins and m-calpain were found to be rich in the synaptosomal, nuclear, and cytosolic subfractions by immunoblotting analysis. Reperfusion (60 min) following ischemia (30 min) induced selective proteolysis of α-fodrin, which was inhibited by a calpain inhibitor, acetylleucylleucylnorleucinal (400 µM, 1 ml, i.v.). The μ-calpain-specific fragment of β-fodrin was not generated during ischemia-reperfusion, supporting the possibility of the involvement of m-calpain rather than μ-calpain in the α-fodrin proteolysis.

    Notes: Abstract: The distribution of brain-type ankyrin (ankyrinB, 212 kDa) and erythrocyte-type ankyrin (ankyrinR, 239 kDa) was investigated in the subcellular fractions of rat forebrain (P1, 1,000 g pellet; P2, 15,000 g pellet; P3, 100,000 g pellet; S, 100,000 g supernatant) by immunoblotting using specific antibodies. The P2 fraction contained ∼40% of the 212- and 163-kDa isoforms of ankyrinB and the 239-kDa isoform of ankyrinR. Further subfractionation of the P2 by Percoll gradient centrifugation followed by separation of myelin showed association of the three ankyrin isoforms with the synaptosome-rich fraction but not with the myelin-rich fraction. The plasma membrane-rich P3 fraction contained a concentration of ankyrin isoforms similar to that in the P2 fraction. In vitro proteolysis of ankyrin in the P2 fraction with calpain showed that the 212-kDa ankyrinB was more susceptible to calpain than was ankyrinR. In the two-vessel occlusion model, ischemia for 30 min generated the 160-kDa fragment of ankyrinR, and reperfusion for 60 min after 30 min of ischemia remarkably increased the 160-kDa fragment. The reperfusion also significantly decreased the 212-kDa isoform of ankyrinB. Both ischemia-reperfusion and in vitro proteolysis with calpain generated the 160-kDa fragment of ankyrinR, suggesting the involvement of calpain.

    Notes: We prepared hydrogen and oxygen alloys of a-Si by the method of rf sputtering and found similarities in their photoluminescence (PL) features. Depression of PL intensity at high temperature is reduced by alloying and results in a larger PL intensity at room temperature. Nonradiative recombination processes in the alloys are discussed in terms of the temperature dependence of the PL intensity. It is concluded that band gap fluctuations induced by alloying reduce transitions to nonradiative defect centers in these systems. © 2002 American Institute of Physics.

    Notes: Bacillus subtilis TnrA is a global regulator that responds to the availability of nitrogen sources and both activates and represses many genes during nitrogen-limited growth. In order to obtain a holistic view of the gene regulation depending on TnrA, we performed a genome-wide screening for TnrA-regulated genes associated with a TnrA box. A combination of DNA microarray hybridization and a genome-wide search for TnrA boxes allowed us to find 36 TnrA-regulated transcription units associated with a putative TnrA box. Gel retardation assaying, using probes carrying at least one putative TnrA box and the deletion derivatives of each box, indicated that 17 out of 36 transcription units were likely TnrA targets associated with the TnrA boxes, two of which (nasA and nasBCDEF) possessed a common TnrA box. The sequences of these TnrA boxes contained a consensus one, TGTNANAWWWTMTNACA. The TnrA targets detected in this study were nrgAB, pucJKLM, glnQHMP, nasDEF, oppABCDF, nasA, nasBCDEF and ywrD for positive regulation, and gltAB, pel, ywdIJK, yycCB, yttA, yxkC, ywlFG, yodF and alsT for negative regulation, nrgAB and gltAB being well-studied TnrA targets. It was unexpected that the negatively regulated TnrA targets were as many as the positively regulated targets. The physiological role of the TnrA regulon is discussed.

    Notes: Target gene candidates of the seven extracytoplasmic function (ECF) sigma factors of Bacillus subtilis have been surveyed using DNA microarray analysis of mRNA extracted from cells grown in Luria–Bertani broth, in which an ECF sigma factor gene was placed under the control of the spac promoter on multicopy plasmid pDG148 and overexpressed. The number of target candidates for each of the sigma factors varied greatly, and a total of 278 genes were selected. Interestingly, the above target gene candidates shared only one gene out of 94 target genes of the general stress sigma B that have been reported in the literature thus far. Furthermore, lacZ-fusion experiments based on the results of DNA microarray analysis indicated that each ECF sigma factor directs transcription of its own operon, with the exception of sigZ.     Notes: Abstract The K+-EDTA-ATPase and Ca2+-ATPase activities of cardiac ventricular actomyosin prepared from autopsy samples from subjects of various ages were determined. We could not detect differences in the K+-EDTA-ATPase or Ca2+-ATPase activity of actomyosin prepared from the hearts of young (0–30 yrs.), middle-aged (31–60 yrs.) and old (61-yrs.) subjects at pH 7.0 and at 30°C conditions widely used in previous studies. However, there were significant decreases in both ATPase activities of actomyosin isolated from old and middle.aged subjects, compared with young subjects at pHs 6.0, 8.0 and 9.0 (at 30°C or at 37°C (at pH 7.0). The alkaline lability of the ATPase activities also increased significantly with age. There has been much debate as to whether or not there is a relationship between age and myocardial myosin ATPase activity. In many studies regarding myocardial myosin ATPase activity of hypertrophied or failed hearts, the age of the subjects was ignored. Our results draw attention to the effect of aging on the myocardial actomyosin ATPase activity and the importance of choosing appropriate assay conditions.

    Notes: Abstract It has been found that the Pb-3222 phase sample could been synthesized in Ce-free Pb-Sr-La-Gd-Cu-O system and the nearly single-phase samples can be obtained within a limited composition. Then, among the nearly single-phase samples, a sample with a nominal composition of Pb2Sr1.4La0.7Gd1.9Cu3O z shows a diamagnetic signal with an onset around 20 K. The superconducting behavior is supposed to arise from the Pb-3222 phase.

    Notes: The effects ensuing from the interaction between membrane-bound sarcosine dehydrogenase and the surrounding lipids as well as the effects of membrane fluidity were described in this study. A 25-fold activation was observed upon the reconstitution of the enzyme in bilayers of SUVs made of DMPC. The considerable decrease in Km and increase in Vmax suggest the induction of favorable conformational changes in both the substrate-binding site and the catalytic site of the enzyme due to the lipid-protein interaction. In SUVs of negatively charged phospholipids, the enzyme retained its initial activity over 1 month. The break point in the Arrhenius plot of the activity of reconstituted enzyme was found at temperatures close to the gel-liquid crystalline transition point of the phospholipid showing that the activity is sensitive to the physical state of membrane phospholipids. Further, immobilization of the reconstituted enzyme by use of ENT prepolymer resulted in a high activity, whereas no remarkable activity was detected with the immobilized enzyme without reconstitution.
