    Notizen: Disturbances in processing simple acoustic changes in a stream of stimuli have been widely reported in patients with schizophrenia, but little is know about auditory feature conjunction in these individuals. This study was designed to examine the extent to which patients with schizophrenia automatically process changes in conjunction of auditory features by using event-related brain potentials. Seventeen patients and 17 age-matched controls were presented with frequent low pitch tones at 45° to the left of center and frequent high pitch tones at 45° to the right of center while performing a continuous visual serial-choice reaction time task. The sequence of auditory stimuli included rare conjunction-deviants comprised of a different combination of features (e.g., low pitch tone at 45° right) and double-deviant tones that differed from the standard tones in both pitch and location (i.e., middle pitch at 0° azimuth). Conjunction-deviant stimuli elicited an MMN wave that was maximum at frontocentral sites. Compared with controls, the MMN to conjunction-deviant was reduced in patients and was more centrally distributed. Double-deviant sounds generated a biphasic MMN followed by a P3a wave at central sites. Both MMN and P3a were reduced in patients compared with controls. These results show that patients with schizophrenia have difficulty in automatically detecting changes in a combination of auditory features as well as orienting to what “normally” would be considered salient by healthy individuals.

    Notizen: . The phylogenetic relationships among peritrichs remain unresolved. In this study, the complete small subunit rRNA (SSrRNA) gene sequences of seven species (Epistylis galea, Campanella umbellaria, Carchesium polypinum, Zoothamnium arbuscula, Vaginicola crystallina, Ophrydium versatile, and Opercularia microdiscum) were determined. Trees were constructed using distancematrix, maximum-likelihood and maximum-parsimony methods, all of which strongly supported the monophyly of the subclass Peritrichia. Within the peritrichs, 1) E. galea grouped with Opercularia microdiscum and Campanella umbellaria but not the other Epistylis species, which indicates that the genus Epistylis might not be monophyletic; 2) the topological position of Carchesium and Campanella suggested that Carchesium should be placed in the family Zoothamniidae, or be elevated to a higher taxonomic rank, and that Campanella should be independent of the family Epistylididae, and probably be given a new rank; and 3) Opisthonecta grouped strongly with Astylozoon, which suggested that Opisthonecta species were not the ancestors of the stalked peritrichs.

    Notizen: Abstract. Microsatellite DNA has been developed into one of the most popular genetic markers. We have identified and cloned microsatellite loci in the genome of a free-living protozoan Euglena gracilis FACHB-848, using the random amplified microsatellites method (RAMS). The digoxigenin-labelled oligonucleotides (CT)10 and (GT)10 served as probes to detect complementary sequences in the randomly amplified polymorphic DNA (RAPD) fingerprints produced by means of Southern blotting. Subsequently, positive RAPD fragments were cloned. From a total of 31 RAPD primer profiles, eight microsatellite loci of E. gracilis were detected and characterized. Further, six sites (i.e. EGMS1, EGMS3, EGMS4, EGMS5, EGMS6, and EGMS7) showed polymorphisms. We found a GT or CT microsatellite every 10.5 kb in the genome of E. gracilis, and similar to animal genomes, the (GT)n motif was much more abundant than the (CT)n motif. These polymorphic microsatellite DNA will serve as advantageous molecular markers for studying the genetic diversity and molecular ecology of Euglena.

    Notizen: . The peritrichs have been recognized as a higher taxon of ciliates since 1968. However, the phylogenetic relationships among them are still unsettled, and their placement within the class Oligohymenophorea has only been supported by the analysis of the small subunit rRNA gene sequence of Opisthonecta henneguyi. DNA was isolated directly from field-sampled species for PCR, and was used to resolve relationships within the genus Epistylis and to confirm the stability of the placement of peritrichs. Small subunit rRNA gene sequences of Epistylis plicatilis, Epistylis urceolata, Epistylis chrysemydis, Epistylis hentscheli, Epistylis wenrichi, and Vorticella campanula were sequenced and analyzed using both distance-matrix and maximum-parsimony methods. In phylogenetic trees, the monophyly of both the genus Epistylis and the subclass Peritrichia was strongly supported, while V. campanula clustered with Vorticella microstoma. The topology in which E. plicatilis and E. hentscheli formed a strongly supported sister clade to E. urceolata, E. chrysemydis, and E. wenrichi was consistent with variations in the thickness of the peristomial lip. We concluded that the peristomial area, especially the peristomial lip, might be the important phylogenetic character within the genus Epistylis.

    Notizen: The greylag goose (Anser anser), which lives on lowlands and cannot tolerate hypoxic conditions, presents a striking contrast to its close relative the bar-headed goose (A. indicus), which lives at high altitude and possesses high-altitude hypoxia adaptation. There are only four amino-acid residue differences at α18, α63, α119 and β125 between the haemoglobins of the two species. The crystal structure of greylag goose oxy haemoglobin was determined at 3.09 Å resolution. Its quaternary structure is slightly different from that of the bar-headed goose oxy haemoglobin, with a rotation of 2.8° in relative orientation of the two dimers. Of the four mutations, those at α119 and β125 produce contact changes in the α1β1 interface and may be responsible for the differences in intrinsic oxygen affinity between the two species; those at α18 and α63 may be responsible for the differences in quaternary structure between the two species.

    Notizen: Bar-headed goose fluoromethaemoglobin (fluoromet-Hb) complexed with inositol hexaphosphate (IHP) has been crystallized using PEG 6000 as precipitant. The crystal belongs to space group P21, with unit-cell parameters a = 59.8, b = 72.0, c = 79.8 Å, β = 102.1°, and diffracts to 2.5 Å resolution. To prove the presence of IHP, the structure was determined by the molecular-replacement method. IHP was observed at the entrance to the central cavity between the N and C termini of two β subunits.

    Notizen: Haemoglobin from the bar-headed goose (Anser indicus) has higher oxygen affinity than that from its lowland relatives such as greylag goose (A. anser). The crystal structure of bar-headed goose aquomet haemoglobin was determined at 2.3 Å resolution and compared with the structures of the goose oxy, human, horse and other avian haemoglobins and the sequences of other avian haemoglobins. Four amino-acid residues differ between greylag goose and bar-headed goose haemoglobins, among which Alaα119 and Aspβ125 in bar-headed goose haemoglobin reduces the contacts between the α1 and β1 subunits compared with Pro and Glu, respectively, and therefore may increase the oxygen affinity by loosening the α1β1 interface. Compared with human oxy haemoglobin, the relative orientation of two αβ dimers in the bar-headed goose aquomet and oxy Hbs are rotated by about 4°, indicating a unique quaternary structural difference from the typical R state. This new `RH' state is probably correlated with the higher oxygen affinity of bar-headed goose haemoglobin.

    Notizen: LCI is a type of novel antibacterial polypeptide secreted by a Bacillus subtilis strain. It consists of 47 residues with a molecular weight of 5468 Da. Using bioengineering, LCI was expressed in Escherichia coli DH5α with recombinant plasmid pBVAB16. It was crystallized using PEG 4000 as a precipitant. The crystal belongs to space group P6222 or P6422, with unit-cell parameters a = b = 29.30, c = 187.09 Å, and diffracts to 2.44 Å. A set of diffraction data to 2.8 Å was collected.

    Notizen: Owing to localized heating by the welding process and subsequent rapid cooling, theresidual stresses can arise in the weld itself and in the base metals. The prediction procedures of theresidual stresses in welding process were presented by using finite element techniques. The bilinearelastic-plastic material model based on Von Mises yield criterion was developed. The materialnon-linearity of weldment and welding fluid was dealt with using an incremental technique. Insideeach step, the Newton-Raphson iteration method was utilized. A fully coupled thermo-mechanicaltwo-dimensional analysis was performed with finite element method. The model applied in this studyadopts the technique of element birth and death to simulate the weld filler variation with time inmulti-pass welded joints. The effects of welding speed on residual stresses are discussed
