    Notes: The total number of bacteria and culturable bacteria in Adélie penguin (Pygoscelis adeliae) guano was determined during 42 days of decomposition in a location adjacent to the rookery in Admiralty Bay, King George Island, Antarctica. Of the culturable bacteria, 72 randomly selected colonies were described using 49 morpho-physiological tests, 27 of which were subsequently considered significant in characterizing and differentiating the isolates. On the basis of the nucleotide sequence of a fragment of the 16S rRNA gene in each of 72 pure isolates, three major phylogenetic groups were identified, namely the Moraxellaceae/Pseudomonadaceae (29 isolates), the Flavobacteriaceae (14), and the Micrococcaceae (29). Grouping of the isolates on the basis of morpho-physiological tests (whether 49 or 27 parameters) showed similar results to those based on 16S rRNA gene sequences. Clusters were characterized by considerable intra-cluster variation in both 16S rRNA gene sequences and morpho-physiological responses. High diversity in abundance and morphometry of total bacterial communities during penguin guano decomposition was supported by image analysis of epifluorescence micrographs. The results indicate that the bacterial community in penguin guano is not only one of the richest in Antarctica, but is extremely diverse, both phylogenetically and morpho-physiologically.

    Notes: Abstract During austral summer 1988/89, total bacterial Acridine Orange Direct Counts (AODC) in seawater, mean 6.0×106 l−1, were three to ten times lower than generally reported for the Bransfield Strait to north Weddell Sea area. In contrast, numbers of viable bacteria (Colony Forming Units, CFU), mean 10.6×103 l−1 were two to three times higher than reported. Bacterial abundance here shows large seasonal and spatial changes. On the basis of bacterial, diatom, detritus, and amino acid data from this study, two main regions were defined: ‘Cold winter water’ in the west with high salinity and low CFU, AODC, and other parameters. In the east, lower salinity and higher values for all parameters were found in warmer meltwater at the surface. CFU and AODC values in ice were respectively six and 85 times higher than in surrounding seawater. Taxonomic studies indicate considerable diversity in genera and nutritional requirements of isolated bacteria. Sea-ice and water column bacterial communities differed. Many isolated strains, however, were found in both habitats. Sea-ice seems to be important in regulating surrounding bacterioplankton.

    Notes: This report strongly suggests that two compartments in Tetrahymena thermophila contain peptidase activity: the cytoplasm and the outer cell surface.Determinations of amino acid concentrations in the extracellular medium upon incubation of cells with peptides suggest that the surface-bound peptidase activity hydrolyses di- and tri-phenylalanine equally fast on a molar basis.Growth experiments designed to characterize the in vivo peptidase specificities showed that both T. thermophila and T. pyriformis can use L-leucyl-L-leucine, but not L-leucyl-D-leucine as a leucine donor. These results are independent of whether the cells form food vacuoles or not.
