Notes: In recent years, electrospinning process is gradually applied in producingtissue-engineering scaffold. In this study, we chose polylacticacid(PLA) and β-tertiary calcium phosphate(β-TCP) as raw materials to fabricate PLA/β-TCP biodegradable composite scaffold by electrospinning process. The characteristics of the scaffold and effect of the scaffolds to cell proliferation and cell adhesion was studied. Compare with pure PLA scaffold, blendingβ-TCP in the spinning process of the scaffold could improve the properties of the scaffold, especially the hydrophilicity and the proliferation and adhesion of cells, this means that the material is more potential to be used as tissue engineering scaffolds

Notes: The influence of calcium (Ca) at titanium surface on co-precipitation of Ca-P and bovine serum albumin (BSA) has been investigated by scanning electron microscopy, X-ray photoelectron spectroscopy and X-ray diffraction. The titanium surface containing Ca was prepared by immersing titanium plates into a saturated calcium hydroxyl (Ca(OH)2) solution. A solution containing BSA, calcium and phosphate ions was used for the co-precipitation test. Both BSA and calciumphosphate co-precipitated onto titanium surfaces with and without Ca. For the former, however, a thicker carbonate apatite coating, which consisted of finer crystal grains, formed and more protein precipitated compared to the surface without Ca. These results indicated that Ca at the titanium surface was favorable to the co-precipitation of BSA and Ca-P. Moreover, chemical interactions probablyoccurred during the co-precipitation of BSA and Ca-P

Notes: Plasma-sprayed hydroxyapatite coating on metal substrate has been prepared, two kinds of post-treatment methods have been used: (1) Heating in air at 650°C for 30 min, (2) Heating in water vapor at 125°C, 0.15Mpa for 6 hours. XRD showed that the nanocrystals of HA coating increased after water vapor treated. The interfacial tensiles strength between HA and the substrate were 45.0±1.82MPa, 39.1±1.27MPa and 30.3±1.61MPa for as-received coatings, water vaportreated coatings and heated in air coatings respectively. After 3 months implant in dogs limbs, the push-out strength between implants and bone were 11.27±2.71 MPa, 11.63±3.11MPa, 23.92± 2.01MPa and 18.8±1.82 MPa for pure Ti implants, as-received coating implants, water vapor treated implants and heated in air implants respectively. The results showed that the water vapor post treated HA coating have better mechanical behavior in vitro and in vivo

Notes: Hydroxyapatite/collagen (HA/COL) composites were prepared using coprecipitation and in-situ synthesis methods. All these processes yielded nanosize poorly crystallized hydroxyapatite/collagen composites. The low temperature in situ formed composites showed some features of natural bone in phase composition, crystal size and crystallinity. Compare with the composite prepared by coprecipitation synthesis, the composite resulted from a low temperature insitu synthesis method showed better homogeneity and mechanical properties. It is confirmed that the low temperature in situ synthesis method was an effective way to obtain biomimetic nanoHA/COL composites with good homogeneity and mechanical properties

Notes: The osteoinduction of Calcium Phosphate (CaP) had been proved and generally been investigated by in vivo implantation. However, the mechanism of the osteoinductivity was not clear and it was difficult to judge the osteoinductivity in vitro. In this study, Mouse C2C12 cell line, a kind of myoblast precursor cell, was employed to co-culture with CaP. The induction of cell differentiation by materials was tested by MTT method, fluorescence observation, especially the mRNA expression of Osteocalcin, Type I collagen and Fibronectin by RT-PCR. It was founded that C2C12 cells could be induced to expression osteocalcin when growth on the surface of the HA/TCP ceramics. At the same time, the ceramics with different composition and sintering temperature seemed to induce difference expression level of the related genes. The results proved that phase composition was one of the most important factors in the regulation of bone-related genes. This study provided a potential model to evaluate the osteoinductivity of CaP ceramics in vitro

Notes: In this study, the bioactivity of a new kind of anodic oxidized titanium metal wasinvestigated in vitro and in vivo. After immersed in SBF solution for 7 days in vitro, apatite formed and covered almost all the surfaces of the anodic oxidized samples. In vivo animal experiment, the apatite was also tested precipitated on the interface of tissue/materials after 12 weeks post-operation, and there were no any fibrous capsule formed around the materials. The materials bonded with the bone very tightly and attached to the skin very closely, which would result in the achievement of the biological sealing for the bone-anchored percutaneous implants. These positive results might be contributed to the precipitated apatite layer formed on the surface of the bioactive oxidized titanium. Thus, Anodic oxidation treatment might be an effective way to prepare bioactive Ti both for bone replacement and percutaneous implant

Notes: Two sets of porous biphasic calcium phosphate ceramics (BCP) were prepared for dynamic SBF experiment: porous BCP with micropores on the walls of macropores( set A) and porous BCP with dense walls of macropores (set B). Apatite layer could only formed on the macropore walls with micropores. Four groups of specimens were prepared for animal experiments.Group A was porous BCP ceramics with micropores on the walls of macropores; group B was porous BCP with dense walls of macropores; group C was porous BCP ceramics with apatite layers formed by static SBF[2]on their surfaces; group D was porous BCP ceramics with apatite layers formed by dynamic SBF on their walls of macropores. The result of dynamic SBF animal experiments showed that microstructure of BCP played an important role in the bone-like apatite formation andosteoinductiion in biomaterials. Apatite formation may be the prerequisite of osteoinductive formation of new bone

Notes: The purpose of this study was to evaluate the response of osteoblasts to the calcium phosphate with different surface modification, and to evaluate the osteoinductive capabilities of these biomaterials. 60HA/40α-TCP ceramics sintered at 1250oC was applied in this study. The ceramic cylinder with F5mm×8mm and slice with F10mm×1mm were prepared respectively. One thirds of the ceramics were formed bone-like apatite (BLA), and the surface of another one thirds was modified with collagen. Osteoblasts (1×106/ml) were co-cultured with the three kinds of thin slices for 12h, 24h and 48h. SEM observation was applied to evaluate if the surface modification and BLA formation could affect the attachment and proliferation of osteoblast in vitro. The three kinds of cylinder samples were implanted in dog muscle to evaluate their differences in osteoinduction. Cells grew inmulti-layers and well attached to the surface and proliferated well in the group of collagen and HA/TCP. In untreated and BLA precipitated group, cells did not attach to the surface well. Osteoinduction was good in BLA precipitated group and the amount of bone formed was higher; in untreated group and collagen-treated group, no bone formation was observed in the tested period. This result indicated that the scaffold used in cell-materials composites in vitro and that in osteoinductive material based tissue engineering in vivo was not same

Notes: It is well known that the cellular responses are related with both physical and chemicalcharacteristics of substrate, including surface topography. In the present study, the effect of surfacetopography of hydroxyapatite (HA) on rat bone marrow cell (rBMCs) response was investigated. HAdisc-shaped pellets with various topography were manufactured by single-axis pressing methods. TherBMCs responses on materials including cell morphology and proliferation were evaluated by SEMand MTT methods respectively, and the differentiation potential was assessed by total protein contentand alkaline phosphatase (ALP) activity testing. The results showed that the cell porliferation washigher on HA surfaces with macropore structure, while ALP activity was lower. No significantdifference in the cellular responses on the pore distribution and orietation was observed. However, thepore structure had a potential to guide cell orientation by gathering the cells inside the pores ratherthan on the ridges. Since ALP served as an indicator of early osteoblast differentiation, in this study itshigher expression on HA surface with micropores suggested that surface microtopograhy exhibited animportant effect on early osteoblast differentiation process
