    Notes: Carbon-coated gadolinum and holmium carbide nanocrystallites have been generated using a modification of the Huffman-Krätschmer carbon arc process. Bulk amounts of these particles were isolated from the other by-products using a magnetic field gradient. Transmission electron microscopy revealed the presence of 10–50 nm diameter crystallites coated with numerous graphitic layers. The nanocrystallite phases were identified as Gd2C3 and Ho2C3, respectively, by x-ray and electron diffraction. Magnetization measurements were performed using a superconducting quantum interference device magnetometer between ±5 T at temperatures ranging from 4 to 200 K. The magnetization curves were shown to scale as a function of H/T. The RE3+ sites in RE2C3 have C3 site symmetry. For Gd2C3 the universal curve was fit with a Brillouin function consistent with the Gd3+ free-ion ground-state values of J=7/2 and g=2. The 5I8 Ho3+ free-ion ground state is split, presumably due to a C3 symmetry crystal field. Consequently, for Ho2C3 the Ho3+ free-ion parameters could not be used to fit the experimental data. Empirical fits to the Brillouin function yield a reduced moment of 7.5μB, compared to the free-ion value of 10.6μB. A similarly reduced moment was observed in holmium-containing endohedral fullerenes.

    Notes: : The binding properties of 2-pentyl pyridine (2-pp) were investigated for soybean protein isolates (SPI) and the beta-conglycinin and glycinin soy protein fractions. The glycinin fraction had the highest binding affinities for 2-pp, followed by beta-conglycinin fraction, and then SPI. More 2-pp was bound by SPI and beta-conglycinin or glycinin fractions under alkaline conditions than under neutral conditions, which exhibited more binding than acidic conditions. More 2-pp was also bound at high temperature (74 °C) than at 25 °C, but greater binding affinity of 2-pp was observed at 4 °C than at 25 °C. With increased NaCl concentrations, the binding affinity of 2-pp decreased. Exposure to UV light increased binding of 2-pp to all types of soy protein.

    Notes: : Using stable isotopic 13C-2,4-decadienal and 15N-ammonia, both compounds were confirmed as precursors in the formation of 2-pentyl pyridine (2-pp) in processing of soybean protein isolates (SPI). The yields of 2-pp were higher when adding 2,4-decadienal and ammonia together than the yields when adding 2,4-decadienal only for soybean cultivars Stress land, LOX null, Edison, and KS4694. Arginine, lysine, asparagine, and glutamine increased 2-pp when added together with 2,4-decadienal, whereas aspartic acid, glutamic acid, glycine, and histidine had no effects. In both buffer and defatted soybean slurry, the highest level of 2-pp was found at pH 9, followed by pH 7, then pH 4.5. The results indicate that the synthesis of 2-pp may be a spontaneous reaction.

    Notes: Levels of 2-pentyl pyridine (2-pp) in protein fractions from Stressland soybeans increased to 0.868 ppm (±0.027) at pH 7 after removing insoluble components from defatted flours. If similar protein fractions were held at pH 4.5 or pH 9, the 2-pp concentration was 0.182 ppm (± 0.003) or 0.199 ppm (± 0.003), respectively. Soybean cultivars KS4694, Edison, and a type null for lipoxygenase 1, 2, and 3 showed similar changes. The increase at pH 7 occurred in less than 1 h. If the defatted flour solution was not subjected to alkaline extraction to remove insoluble components, the increase of 2-pp at neutral pH did not occur. Thus, the pH conditions should be carefully controlled to produce soybean protein isolates with desired flavor properties.

    Notes: Abstract Retroviral vectors were used to introduce the wild-type p53 gene into human bladder cancer cell lines BIU-87 and EJ, which express endogenous wt-p53 gene and have a mutation in H-ras gene. The expression of the exogenous wt-p53 gene in cells suppresses the growth of the bladder cancer cells in standard culture and in soft agar and blocks the cell cycle progression in G1. The BIU-87 and EJ cells developed tumors with average volumes of 6.53 cm3 and 6.61 cm3 in nude mice in 9 weeks after inoculation, while the cells transduced with wt-p53 gene failed to form tumors. The expression of H-ras gene in bladder cancer cells was reduced at mRNA level. These results suggest that the overexpression of the wt-p53 gene suppresses the expression of mutant H-ras gene and inhibits the tumor cell growth in vivo and in vitro.

    Notes: Summary As part of continuing multidisciplinary studies on the neuropathogenesis of subacute sclerosing panencephalitis (SSPE), in situ hybridisation, immunocytochemistry and electron microscopy were used to detect measles virus nucleic acid, protein and nucleocapsids in brain perivascular infiltrates of three cases. Perivascular cuffing cells which contained measles virus nucleic acid and antigens were found in all cases. Infected cuffs occurred predominantly in areas of general parenchymal cell infection and in many of these a high proportion of the infiltrating cells were infected. Other cuffs in these areas were either uninfected or contained only a few infected cells. Occasional infected cells were also seen in cuffs in non-infected areas. In contrast, no specific immunocytochemical reactions or in situ hybridisation for measles virus was observed in brain tissue from a patient with herpes encephalitis. By electron microscopy viral nucleocapsid, consistent with measles virus, was found within the cytoplasm of plasma cells in the inflammatory cuffs in SSPE brain tissue. Possible explanations for our results are that infiltrates become infected on arrival in the CNS or alternatively, that the infected infiltrates reflect a generalised infection of the reticuloendothelial system. The frequent presence of uninfected cuffs favours the former explanation.

    Notes: Summary The peptidyl alpha-amidation of biologically active peptides (a number of which are found in the endocrine pancreas) requires several co-factors, including ascorbic acid. In the present study, tissue contents and developmental changes of ascorbic acid in the rat endocrine panreas were measured using a highly sensitive HPLC system. High concentrations were found in neonatal rats, with the highest value, 42.5 nmol/mg protein, in 2-day-old rats. The concentration decreased gradually with age to 19.4 nmol/mg in 5-week-old rats. The exocrine pancreas had a lower concentration, but a peak was also observed in 2-day-old rats. In freshly isolated islet cells, an intracellular concentration of 7.5 mmol/l was estimated in 5–7-day-old rats. Secretory granules isolated from 4–6-day-old rat islets contained 10.6 nmol/mg protein. Culturing islets or cells in ascorbic acid free medium resulted in a marked decrease in their contents. Ascorbic acid in secretory granules from such islets decreased at a relatively lower rate. Addition of ascorbic acid to cultured cells or islets reduced the loss markedly. Increasing the glucose concentration in islet culture medium in the presence of 100 μmol/l ascorbic acid increased the islet ascorbic acid concentration.
