    Description / Table of Contents: Résumé Une série d'expériences a été effectuée avec des rats diabétiques par l'alloxane et des rats témoins afin d'étudier la croissance de granulomes de pellets de coton et la vitesse d'incorporation du35S-sulfate dans des mucopolysaccharides sulfatés du tissu de granulation. — Il a pu être démontré que chez les animaux diabétiques par l'alloxane, il y avait non seulement une formation nettement diminuée de tissu de granulation, mais aussi un blocage prononcé de la synthèse des mueopolysaccharides sulfatés dans le tissu de granulation. — On suppose que ces résultats sont causés par le manque d'insuline dans le diabète alloxanique et qu'ils sont en relation avec la guérison retardée des blessures chez les sujets ayant un diabète sucré mal équilibré.
    Abstract: Zusammenfassung Es wurde im Tierexperiment an gesunden und alloxandiabetischen Ratten das Wachstum und der Sulfomucopolysaccharidstoffwechsel des Granulationsgewebes von Cotton-pellet-Granulomen überprüft. — Die Untersuchungen haben ergeben, daß bei alloxandiabetischen Tieren eine deutliche Verminderung der Granulationsgewebsbildung sowie eine ausgeprägte Hemmung des Mesenchymstoffwechsels in neugebildeten Granulationsgewebe im Vergleich zu normalen Kontrolltieren bestehen. — Die Befunde werden auf den bei Alloxandiabetes nachweisbaren Insulinmangel zurückgeführt und lassen den Schluß zu, daß die verminderte Wundheilung bei Patienten mit dekompensiertem Diabetes mellitus auf einer Störung des Mesenchymstoffwechsel beruht.
    Notes: Summary A series of experiments with alloxan-diabetic and control rats was done in order to investigate growth of cotton-pellet granulomas and incorporation rate of35S-sulphate into sulphated mucopolysaccharides of the granulation tissue. — It could be demonstrated that in the alloxan-diabetic animals there was not only markedly less formation of granulation tissue, but a pronounced blocking of synthesis of sulphated mucopolysaccharides in the granulation tissue as well. — These findings are assumed to be caused by the lack of insulin in alloxan diabetes, and to be related to the delayed wound-healing in patients with badly controlled diabetes mellitus.

    Notes: Abstract Whole-cell patch clamp recordings were done on giant protoplasts of Escherichia coli. The pressure sensitivity of the protoplasts was studied. Two different unit conductance mechanosensitive channels, 1100 ± 25 pS and 350 ± 14 pS in 400 mm symmetric KCl solution, were observed upon either applying positive pressure to the interior of the cells or down shocking the cells osmotically. The 1100 pS conductance channel discriminated poorly among the monovalent ions tested and it was permeable to Ca2+ and glutamate−. Both of the two channels were sensitive to the osmotic gradient across the membrane; the unit conductances of the channels remained constant while the mean current of the cell was increased by increasing the osmotic gradient. Both of the channels were voltage sensitive. Voltage-ramp results showed that the pressure sensitivity of protoplasts was voltage dependent: there were more channels active upon depolarization than hyperpolarization. The mech anosensitive channels were reversibly blocked by gadolinium ion. Also they could reversibly be inhibited by protons. Mutations in two of the potassium efflux systems, KefB and KefC, did not affect the channel activity, while a null mutation in the gene for KefA changed the channel activity significantly. This indicates a potential modulation of these channels by KefA.

    Notes: Abstract. The effect of a kefA mutation on the mechanosensitive channels in the cytoplasmic membrane of Escherichia coli was established by introducing a mutation of the kefA gene into wild-type E. coli by P1 transduction. The mutation of the kefA gene not only made the cells sensitive to K+ in the medium but also changed the mechanosensitive channel activity. The kefA mutation did not change the conductances of the two mechanosensitive channels in the cytoplasmic membrane of E. coli, but it prolonged the channel open time. Also, the kefA mutation made the cells more sensitive to pressure in comparison to wild-type cells. The high sensitivity to pressure of the kefA mutant was not modulated by betaine or by the potassium gradient across the membrane. The effect of the kefA mutation on mechanosensitive channels was not due to a membrane fluidity change. KefA might be a regulator for mechanosensitive channels.

    Notes: Background  In the conservative therapy of venous leg ulcers modern types of dressings are used most frequently. In the past 20 years ‘active wound dressings’ − cultured epidermal keratinocytes as autografts and allografts − were also introduced in the management of leg ulcers.Methods  The aim of our study was to compare the effect of cryopreserved and lyophilized cultured epidermal allografts in the treatment of venous leg ulcers. Evaluation of the therapy was documented as photodocumentation, planimetry, healing time and evaluation of pain relief over a 3-month period after application. Fifty patients with venous leg ulcers were selected. Twenty-five patients (group I) were treated with cryopreserved keratinocytes and 25 (group II) with lyophilized keratinocytes.Results  The final evaluation 3 months after the application of allografts showed 84% of healed ulcers in group I and 80% in group II. The number of healed ulcers and the healing rate both showed no statistically significant differences. The size of the ulcer was reduced by half during the first week in both groups. The size differences during the first week are statistically significant in both groups and they are comparable (P 〈 0.001). The intensity of the pain was statistically significantly reduced during the first week after application in both groups (P 〈 0.001).Conclusions  The cryopreserved and lyophilized cultured allografts are comparable in healing rate, course of healing and relief of pain, and also in planimetric changes during the healing of venous leg ulcers. Lyophilized allografts are more convenient for routine use than cryopreserved allografts as they can be stored at room temperature. These results could give rise to the more frequent use of lyophilized allografts in slow-healing venous leg ulcers.
