    Description / Table of Contents: Zusammenfassung Faktor XIII aus menschlichen Thrombozyten wurde isoliert und in langen Nadeln kristallisiert. Er wurde mit dem Faktor XIII aus menschlichem Plasma verglichen; in ihren physiko-chemischen Eigenschaften unterscheiden sich die beiden fibrinstabilisierenden Faktoren ganz wesentlich: im Molekulargewicht, in den Untereinheiten sowie in der Zusammensetzung der Kohlenhydrate und der Aminosäuren. Aus ihrem immunochemischen Verhalten kann eine partielle antigene Identität abgeleitet werden. Kaninchen-Immun-Serum gegen Plasma-Faktor XIII hemmt die fibrinstabilisierende Aktivität sowohl des Plasmas als auch der Plättchen. Das gleiche gilt auch für das Immun-Serum gegen Thrombozyten-Faktor XIII. Aus dieser gegenseitigen Hemmung kann man schließen, daß die aktiven Zentren in den beiden fibrinstabilisierenden Faktoren, zum Teil wenigstens, identisch sind. Läßt man Plasma- und Thrombozyten-Faktor XIII gleichzeitig gegen eines ihrer Antiseren diffundieren, so haben beide eine Präzipitatlinie gemeinsam. Eine zusätzliche zweite Linie, die der Plasma-Faktor XIII mit seinem Antiserum gibt, fehlt beim Thrombozyten-Faktor. Im Thrombozyten-Faktor XIII existiert nur einer von den zwei im Plasma-Faktor XIII vorkommenden Antigenbezirken.
    Notes: Summary Factor XIII from human platelets which has been isolated and obtained in pure crystalline form is compared with factor XIII from human plasma. In their physicochemical properties the two fibrin stabilizing factors are completely different: they differ in molecular weight, in subunits as well as in their carbohydrate and amino acid composition. From their immunochemical behaviour a partial antigenic identity can be derived. Rabbit immune serum to plasma factor XIII inhibits the fibrin stabilizing activity of plasma as well as of platelets. The same is true for the immune serum to platelet factor XIII. From this mutual inhibition one must conclude that the active sites in both fibrin stabilizing factors are at least in part identical. With immunodiffusion there is a line of complete identity between plasma and platelet factor XIII when diffused against either immune serum. Another separate precipitin arc which is formed by plasma factor XIII opposite its antiserum is not given by platelet factor XIII. Of the two different antigenic sites found in plasma factor XIII only one exists in platelet factor XIII.

    Description / Table of Contents: Zusammenfassung Die immunologischen Beziehungen zwischen Mutter, Foetus und Placenta werden diskutiert und die Antigene der menschlichen Placenta, soweit sie bereits physikalisch-chemisch und immunologisch genauer charakterisiert sind, werden beschrieben. Der Nachweis und die Bestimmung von Placentaproteinen wird in der Diagnostik zur Erkennung und Überwachung der Schwangerschaft, sowie zum Nachweis von Tumorerkrankungen verwendet. Antikörper gegen Placentaantigene haben eine abortive Wirkung; eine Immunisierung gegen spezifische Proteine der Placenta kann für eine immunologische Konzeptionsverhütung und für eine Immuntherapie von Chorioncarcinomen von Bedeutung werden.
    Notes: Summary The immunologic relations between mother, foetus and placenta are discussed and those antigens of the human placenta already characterized by their physico-chemical and immunochemical properties described. In diagnostics the detection and determination of placental proteins is used in pregnancy tests, for the evaluation of placental functions and the detection of malignant diseases. Antibodies to placental antigens have an abortive effect; immunization against placental specific proteins may have implications as an immunologic contraceptive method and for the immunotherapy of choriocarcinoma.

    Description / Table of Contents: Zusammenfassung Es werden Serumproteine beschrieben, deren Bedeutung sowohl für die Schwangerschaft, als auch für das maligne Wachstum, bedingt durch die drastische Zunahme oder das erstmalige Auftreten, naheliegen. Da beiden Vorgängen immunologische Nichtreaktionen (Toleranz) gegen Fremdantigene eigen ist und in vitro Befunde für immunsuppressive Eigenschaften der beschriebenen Serumproteine vorliegen, kann an eine Funktion dieser Proteine im Rahmen der veränderten Immunität gedacht werden. Es wäre eine Rollenverteilung der sog. schwangerschaftsspezifischen Proteine, und der Proteine deren Produktion normalerweise nur in geringem Umfang im Organismus stattfindet, aber durch die Auswirkungen der Gravidität oder eines malignen Wachstums stimuliert wird, denkbar. Aufgabe der vom Neoplasma gebildeten Serumproteine könnte es sein, den efferenten Schenkel des Immunapparates zu blockieren. Sie würden antigene Determinanten der Tumorzelle besetzen und damit die spezifischen Reaktionsmechanismen der Effektor Zellen verhindern als auch den Tumor nicht angreifbar machen. Letzteres käme früheren Vorstellungen vom Glykoproteinmantel der Tumorzelle sehr nahe. Das humane Choriongonadotropin scheint in diesem Zusammenhang am besten untersucht zu sein und dieses Modell zu bestätigen. Den Serumproteinen die normalerweise in kaum nachweisbaren Mengen beim Gesunden vorkommen, käme dagegen die Funktion der Blockierung des afferenten Schenkels des Immunsystems zu, d.h. diese Proteine wären in der Lage den Erkennungsmechanismus immunkompetenter Lymphozyten außer Kraft zu setzen und die Sensibilisierung zu verhindern. Hierfür liefern sowohl das schwangerschaftsassoziierte α2 Glykoprotein im Electrophoretic Mobility Test als auch der Nachweis von α1-Fetoprotein Rezeptoren auf T-Lymphozyten, Hinweise.
    Notes: Summary Serum proteins are described the significance of which for pregnancy or for malignant growth is associated with their drastic increase or initial occurrence. In that immunological non-reaction (tolerance) against foreign antigen is inherent in both of the proceeding, and in that in vitro findings display immunosuppressive properties for the described serum proteins, a function within the scope of altered immunity can be conceived for these proteins. A dual role of the so called pregnancy-specific proteins and the pregnancy-associated proteins would be possible. The purpose of the neoplasma-derived serum proteins could be to block the efferent branch of the immune apparatus. They would occupy antigen determinants of the tumor cells and thereby inhibit the specific reaction mechanism of the effector cells as well as render the tumor invulnerable. The latter would closely resemble the earlier concept of a glycoprotein coat of the tumor cell. In this connection, the human Choriongonadotropin appears to be the best investigated and seems to confirm this model. The serum proteins, present in healthy individuals normally in almost indetectable amounts could, by contrast, acquire the function of blocking the afferent branch of the immune system. That is to say these proteins would be able to annul the recognition mechanism of immunologically competent lymphocytes, and to inhibit sensibilization. The electrophoretic mobility test of the pregnancy-associated α2-glycoprotein as well as the determination of α1-fetoprotein receptors on T-lymphocytes are evidence for this concept.

    Description / Table of Contents: Summary By using an enzyme-bridge immunoperoxidase (PAP) technique, localization of so-called pregnancy-specific β1-glycoprotein (SP1) and placental-specific tissue proteins (PP5, PP10, PP11, PP12) was investigated in 19 cases of breast cancer, 24 cases of testicular malignant tumours, 12 cases of gastric cancer and some cases of other malignant tumours. These five glycoproteins isolated from human term placentae and characterized by Bohn are localised mainly in the cytoplasm, or nucleus of syncytiotrophoblast and appear to be specific for the trophoblast. But to a certain percentage these proteins could also be detected in the cytoplasma of malignant cells: In breast cancer, SP1 was present in 52.6% of cases, PP5 in 63.2%, PP10 in 68.4%, PP11 in 55.6% and PP12 in 31.6%. In testicular malignant tumours the detection rates of these proteins varied from 20.8 to 75.0% and in gastric cancer from 41.7 to 66.7%. In total 50% of the tumours tested were SP1-positive, 58.3% PP5-positive, 55.6% PP10-positive, 38.0% PP11-positive and 31.9% PP12-positive. In addition, it was found that mononuclear histiocytes showed a strong cytoplasmic staining for PP10 and PP12 and that a few other non-cancerous cells (i.e. striated cells in gastric metaplasia, gastric polyps etc.) stained for PP5, PP10, PP11 and/or PP12. All control sections were negative in malignant cells as well as in other tissue cells. This study confirms the reports that some kinds of malignant tumours produce SP1 and indicates that the ectopic production of proteins normally produced by the trophoblast is a common finding in malignant tumours. It furthermore suggests that such proteins may be useful as markers in monitoring patients with malignant diseases. The possible mechanisms of ectopic production of these inappropriate proteins in malignant tumours are discussed.
    Notes: Zusammenfassung Unter Verwendung der ‚Enzyme-Bridge‘-Immunoperoxidase (PAP)-Technik wurde das sogenannte schwangerschaftsspezifische β1-Glykoprotein (SP1) und einige placentaspezifische Gewebsproteine (PP5, PP10, PP11, PP12) im Gewebe von malignen Tumoren lokalisiert. Diese von Bohn aus menschlichen Placenten isolierten und charakterisierten fünf Glykoproteine wurden in der Placenta hauptsächlich im Zytoplasma oder in den Zellkernen des Syncytiotrophoblast lokalisiert und scheinen für die Placenta spezifisch zu sein. Zu einem gewissen Prozentsatz konnten diese Proteine aber auch im Zytoplasma von malignen Zellen nachgewiesen werden. Untersucht wurden 19 Fälle von Brustkrebs, 24 Fälle von malignen Hodentumoren, 12 Fälle von Magen-Carcinomen und einige Fälle von anderen malignen Tumoren: Bei Brustkrebs war SP1 in 52,6%, PP5 in 63,2%, PP10 in 68,4%, PP11 in 55,6% und PP12 in 31,6% der Fälle positiv. Bei malignen Hodentumoren lag die Nachweisrate dieser Proteine zwischen 20,8% und 75,0% und bei Magen-Carcinomen zwischen 41,7% und 66,7%. Insgesamt waren von den getesteten malignen Tumoren 50% SP1-positiv, 58,3% PP5-positiv, 55,6% PP10-positiv, 38,0% PP11-positiv und 31,9% PP12-positiv. Zusätzlich wurde gefunden, daß mononukleare Histiozyten im Krebsgewebe eine starke zytoplasmatische Färbung auf PP10 und PP12 zeigten und daß einige wenige andere nichtkanzeröse Zellen (Saumzellen bei intestinaler Metaplasie, Magenpolypzellen) auf PP5, PP10, PP11 und/oder PP12 anfärbten. Alle Kontrollschnitte waren negativ, sowohl bei malignen Zellen als auch bei anderen Gewebszellen. Diese Untersuchung bestätigt die Berichte über das Vorkommen von SP1 bei einigen malignen Tumoren und zeigt, daß auch andere Proteine die normalerweise nur im Trophoblast vorkommen, von malignen Tumoren ektopisch produziert werden. Sie weist außerdem auf die Möglichkeit hin, solche Proteine als Marker bei der Überwachung von Patienten mit malignen Erkrankungen zu verwenden. Die möglichen Mechanismen der ektopischen Produktion dieser Proteine in malignen Tumoren werden diskutiert.

    Description / Table of Contents: Summary A number of placental and pregnancy proteins has been detected by immunochemical methods in extracts from human term placentae. Several of these proteins already have been isolated and characterized and are now investigated for their usefulness as markers in oncology. Several of these proteins so long tested appear to be useful as markers in monitoring tumour patients, i.e. in the early detection of tumour recurrence or metastases and in the control of therapy. In addition, we hope that a combination of several of these new markers with already established tumour markers will enable to better diagnose tumours. Also will the use of solitary tissue proteins aid in the differential diagnosis of cancers.
    Notes: Zusammenfassung Im Extrakt reifer menschlicher Plazenten konnte mit immunologischen Methoden eine Reihe von Plazenta- und Schwangerschaft-Proteinen nachgewiesen werden. Einige dieser Proteine sind bereits isoliert und charakterisiert worden; sie werden jetzt auf ihre Brauchbarkeit als Marker in der Tumordiagnostik untersucht. Mehrere dieser Proteine wurden bereits auf ihre klinische Verwendbarkeit getestet und als Tumormarker zum Frühnachweis von Rezidiven und Metastasen oder zur Überwachung der Therapie bei Tumorpatienten nützlich befunden. Es wird erwartet, daß eine Kombination dieser neuen Marker mit bereits etablierten Tumormarkern einen besseren Nachweis von Tumoren ermöglicht und daß sich spezifische und solitäre Gewebsproteine zusätzlich zur Differentialdiagnose von Tumorerkrankungen verwenden lassen.

    Notes: Summary Pregnancy-specific β1-glycoprotein (PSβG) is a heterogeneous product of the human syncytiotrophoblast, closely related to the CEA-NCA multigene family. In the present study, immunoscreening was carried out with anti-PSβG antibodies to isolate cDNA sequences from a placental λgt11 expression library. One 1847-bp cDNA clone comprising the major portion of the coding sequence of a putative 48-kd peptide was sequenced and characterized. Hybridization of human genomic DNA to the PSβG sequence revealed a complex pattern of restriction fragments, a finding well in agreement with the assumption that there are several independent PSβG genes. A variable PstI band was found in human DNA. Transfer blot analysis of human placental RNA identified transcripts of 2.2 kb and 1.7 kb that appear transiently with increasing levels of expression during gestation. No hybridization of PSβG cDNA to human RNA from liver, kidney, heart, thyroid, and ovary was observed. In analyses of placental RNA from mouse, goat, sheep, and cow, no correponding transcripts could be detected, and DNA hybridization under low-stringency hybridization conditions resulted in very faint cross-reacting bands, presumably indicating sequences that were scarcely related. However, PSβG-specific DNA sequencies with similar restriction patterns were found in primates. These results are compatible with the assumption of late evolutionary development of certain PSβG sequences.
