N-freie Versuchsansätze enthalten geringere Ketosäuremengen, zeigen jedoch innerhalb der späteren Entwicklungsabschnitte ein antagonistisches Verhalten von Pyruvat und α-Ketoglutarat. Von der 24. Std bildet sich α-Ketoglutarat unter Abnahme des ausgeschiedenen Pyruvates. Die Umbildung erfolgt stöchiometrisch. Zusätze von Succinat oder α-Ketoglutarat verzögern lediglich die Wiederaufnahme der extracellularen Brenztraubensäure, sie beeinflussen nicht die Anstiegswerte der Pyruvatbildung. Versuchsansätze mit Glycerin anstelle von Glucose zeigen, daß die Pyruvatstauung keinem Glucoseeffekt im Sinne spezieller Enzymrepressionen zugeordnet werden kann. Aus den Ergebnissen der NH4 .-Versuche geht hervor, daß Wachstumsvorgänge und Pyruvatstauung Antagonisten sind. Eine NH4 .-bedingte Pyruvatanhäufung erfolgt nur bei Anwesenheit von Succinat. Die Ketosäureausscheidung der Streptomyceten wird auf zwei ungleiche Stoffwechselstörungen zurückgeführt: die anfängliche gehemmte Pyruvatoxydation und die anschließende Konkurrenzbeziehung der Pyruvat-bzw. α-Ketoglutaratoxydation auf der Basis eines früher formulierten Kontrollprinzipes.
    Notes: Summary Submerged cultures of Str. rimosus excrete pyruvic and α-oxoglutaric acid in glucose-containing mediums in the presence of casaminoacids. After buffering with carbonates the amounts of keto acids increase about threefold. N-free cultures show smaller amounts of keto acids, but after 24 h we can see an antagonistic behaviour of both curves: pyruvic acid is converted stoichiometrically to oxoglutaric acid. Succinate and α-oxoglutarate delay the secondary reception of pyruvate without changes in primary excretion. Replacement of glucose by glycerine-containing cultures shows that pyruvate excretion is not due to a glucose-effect in the sense of enzyme-repression. NH4-ions depress, pyruvate-excretion and induce a very good growth. A NH4-dependent pyruvate peak appears only in the presence of succinate. The excretion of keto acids by Streptomycetes is caused by two unequal metabolic disturbances: the initially inhibited pyruvate-oxidation with failed pyruvate dehydrogenase, and the associated competition from pyruvate and/or α-oxoglutarate oxidation based on a previously formulated regulation principle.

    Description / Table of Contents: Zusammenfassung Overflächen- und Submerskulturen von Streptomyces rimosus zeigen nach Kältebehandlung einen raschen Vitalitätsverlust. Physiologische Untersuchungen an flüssigen und festen Nährubstraten haben ergeben, daß die Ausreifung des Mycels von einer erheblichen Alkalisierung der Medien begleitet wird; in den Flüssigkeitskulturen werden pH-Werte, von über 8,5 erreicht. Submerse Folgepassagen solcher alkalischer Aufbewahrungskulturen zeigen in Abhängigkeit von der Zeitdauer der Kältewirkung physiologische Veränderungen, die auf synthetischen Medien als gestörter Kohlenhydratstoffwechsel zu fassen sind. In Übereinstimmung mit einschlägigen Literaturangaben unterliegen die reifen Streptomycetenkulturen der Wirkung ihres selbstgebildeten Antibioticums, die entweder eine Selektion stoffwechselabweichender Organismen auslöst oder die den Stoffwechsel der Resistenten erst während der Aufbewahrungszeit prägt. In diesem Falle wäre der abweichende Stoffwechselzustand über mehrere Folgekulturen im Mycel manifestiert worden.
    Notes: Summary Surface and submerged cultures of Streptomyces rimosus show a rapid loss of viability after storage at + 4°C. Physiological tests with fluid and solid nutrient substrates indicate that as the mycelium ripens, the medium becomes strongly alkaline (fluid cultures show a pH of over 8,5). Subsequent submerged passages of such alkaline stored cultures show physiological changes dependent on the duration of the cold treatment which identified on synthetic media as disturbances of the carbohydrate metabolism. From the literature it is known that cultures of ripe Streptomycetes are affected by their own antibiotic. This either causes a selection of organisms with unusual metabolism, or affects the metabolism of the resistant strains during cold storage. In this case, the deviant metabolism should persist in the mycelium through several culture passages.

    Notes: Zusammenfassung Keimversuche hatten ergeben, daß Mischungen von Silber-, Nickel-, Cadmium- und Kobalt(II)-Salzen mit DMDT gegenüber Conidien von Fusarium decemcellulare einen konzentrationsabhängigen Toxicitätsverlust (Inversion) zeigten. Anhand UV-spektrographischer Untersuchungen wurde dargestellt, daß die Inversion nicht auf einer Veränderung komplexchemischer Bindungen zwischen Schwermetall und DMDT im Sinne der Umwandlung eines toxischen 1:1-Komplexes in einen nichttoxischen 1:2-Komplex beruht.

    Notes: Zusammenfassung Stoffwechselintensive Streptomyceten zeigen bei extremer Stickstoffbelastung eine starke Pyruvatstauung, die durch eine verminderte Decarboxylierung von glykolytischer Brenztraubensäure ausgelöst wird. Ursache dafür ist eine erhöhte Verstoffwechslung von α-Ketoglutarsäure infolge einer konkurrierenden Inanspruchnahme der Co-Faktoren. Wir nehmen an, daß die Beanspruchung und Neusynthese der Co-Faktoren erst nachträglich regulativ koordiniert werden. Dieser Pyruvatstau führt aber keineswegs zum Stoffwechselstillstand, sondern wahrscheinlich durch Hemmung im unteren Glykolysebereich, zu einer anhaltenden Metabolitstauung und damit zu einer Aktivierung des Hexosemonophosphatweges. Die Nachschubreaktion wird damit beeinträchtigt, und das angehäufte Pyruvat wird wieder abgeführt. Auf diese Weise funktioniert der HMP als regulativer Ausweichstoffwechsel. Gleichzeitig fanden wir bei α-Ketoglutarsäurebelastung eine selektive Anhäufung von Serin (?), was wir als vermutliches Reaktionsprodukt einer Aminierung zwischen α-Ketoglutarsäure und Oxalessigsäure (im TCC) diskutieren.

    Notes: Zusammenfassung Unterschiedlich konzentrierte Mischungen von Kupfersulfat und Natriumdimethyl- bzw.-diäthyldithiocarbamat zeigen an Conidien von Fusarium decemcellulare einen konzentrationsabhängigen Wechsel im toxischen Verhalten. UV-spektrographische Untersuchungen in Aqua dest.- und in Nährlösungsversuchen haben ergeben, daß der Toxicitätsverlust in keinem Fall von komplexchemischen Veränderungen im Sinne stöchiometrisch ungleichwertiger Schwermetallchelate (1:1 und 1:2) begleitet war. Aufgrund von Versuchen mit Kupfersulfat in subletalen Dosen in Gegenwart von zwei Komplexbildnern oder von Rhodamin B liegen Argumente dafür vor, daß ein toxischer Effekt gegenüber den Mikroorganismen nur dann zu erwarten ist, wenn Cu-Ionen gleich-zeitig mit dem gesättigten 1:2-Chelat auf die Zellen einwirkt. Ob über die nachweisbare Veränderung der Permeabilität das eingedrungene 1:2-Cu-Dithiocarbamat toxisch wirkt oder seine Komponenten, geht aus den Untersuchungen nicht hervor.

    Notes: Abstract Poly(hydroxybutyric acid) (PHB) was produced by a selectant of Azotobacter beijerinckii in media containing only organic nitrogen sources such as N substrates. The chosen compounds were casein peptone, yeast extract, casamino acids and urea, each combined with carbon substrates glucose or sucrose. The PHB was synthesized under growth-associated conditions. The concentrations amounted to more than 50% of cell dry mass on casein peptone/glucose as well as urea/glucose medium within 45 h fermentation time. Corresponding to these yields, productivities of about 0.8 g PHB l−1 h−1 were discovered. The highest values increased to 1.06 g PHB l−1 h−1 on casein peptone/glucose medium and 1.1 g PHB l−1 h−1 on yeast extract/glucose medium after a period of 20 h. It was found that oxygen limitation was essential for successful product formation, as demonstrated earlier. These data from basic research may support further investigations into the use of technical proteins from renewable sources as substrates for PHB production by a strain of A. beijerinckii.

    Notes: Abstract Polyhydroxybutyrate (PHB) was produced by Ralstonia eutropha DSM 11348 (formerly Alicaligenes eutrophus) in media containing 20–30 g l−1 casein peptone or casamino acids as sole sources of nitrogen. In fermentations using media based on casein peptone, permanent growth up to a cell dry mass of 65 g l−1 was observed. PHB accumulated in cells up to 60%–80% of dry weight. The lowest yields were found in media without any trace elements or with casamino acids added only. The residual cell dry masses were limited to 10–15 g l−1 and did not contain PHB. The highest productivity amounted to 1.2 g PHB l−1 h−1. The mean molecular mass of the biopolymer was determined as 750 kDa. The proportion of polyhydroxyvalerate was less than 0.2% in PHB. The bioprocess was scaled up to a 300-l plant. During a fermentation time of 39 h the cells accumulated PHB to 78% w/w. The productivity was 0.98 g PHB l−1 h1.

    Notes: Summary Addition of L-lysine to cultures ofS. noursei enhanced the production of nourseothricin. The aspartokinase of the wild-type strain was under concerted feedback inhibition by lysine plus threonine but was stimulated by lysine alone. Threonine in the medium increased the synthesis of enzyme. 10% of the mutants resistant to AEC showed a higher specific production of the antibiotic.
