    Description / Table of Contents: Formation of Quinolizidine Derivatives by Condensation of meso- and racem. 1.3-Di-[2-piperidyl]-2-propanone with FormaldehydeCondensation of meso- or racemic 1.3-di-[2-piperidyl]-2-propanone (1a, 1b, resp.) with formaldehyde in a buffered aqueous solution at pH 7 and 25° gives, in addition to two condensation products C15H26N2O2 with spirane structures, an isomeric “Quinolizidine-derivative A” (3A; m. p. 154°). A stereoisomeric “Quinolizidine-derivative B” (3B; m. p. 152°) is obtained by the condensation of 1b in acetic acid at 90°. - The compounds 3A and 3B lose one mole of formaldehyde in dilute acetic acid solution in the presence of dimedone to yield a ketone C14H24N2O (4A; m. p. 96°) and regenerates the Quinolizidine-derivative A with formaldehyde (scheme 1). - The constitution of 4 follows from the replacement of its oxygen by hydrogen atoms whereby it is converted through the stereoisomeric alcohols 5 and chlorides 6 to one of the four possible stereoisomeric racemates of 3-[2-piperidyl]-quinolizidine (7); this is identical to the “Racemate C” of known constitution but unknown configuration, mentioned in the literature (scheme 2). Both quinolizidine derivatives therefore have the constitution 3; the hemi-ketal groups are especially indicated by the absence of carbonyl bonds in the IR-spectrum and by formation of methyl ethers sensitive to acid hydrolysis. - The isomerism of 3A and 3B arises since the configuration of the basic skeleton (4) differs in 3B from that in 3A and in the ketone 4A. The latter has apparently the more stable configuration, for it is formed also by loss of formaldehyde from 3B in dilute acetic acid. The expected but non-isolatable β-amino ketone 4B is easily isomerised with stereochemical inversion. - The transformation 4B → 4A does not occur when 3B is reduced with lithium aluminium hydride or treated with phenyl isothiocyanate. In the latter case loss of one mole of formaldehyde gives a product, derived from 4B but differing from the stereoisomeric product correspondingly obtained from either 3A or 4A. - Reduction of 3A or 3B with lithium aluminium hydride - or Clemmensen method in case of 3A - leads to a pair of epimeric 2-hydroxy-3-[1-methyl-2-piperidyl]-quinolizidines (9). The epimeric alcohols derived from 3A give the 3-[1-methyl-2-piperidyl]-quinolizidine (11A) when the hydroxyl groups are replaced by hydrogen. The same product is alternatively obtained by methylation of “Racemate C” and therefore has the same configuration about its three asymmetric centres as does “Racemate C”, corresponding to that of 4A and 3A (scheme 3). The epimeric alcohols 9 derived from 3B yield analogously another stereoisomer of 3-[1-methyl-piperidyl-2]-quinolizidine (11B). - The most probable configurations and conformations of the compounds are discussed.
    Notes: Bei der Kondensation des meso- und racem. 1.3-Di-[2-piperidyl]-2-propanons (1a bzw. 1b) mit Formaldehyd in wäßriger Pufferlösung vom pH 7 bei 25° bildet sich neben zwei Kondensationsprodukten C15H26N2O2 mit Spiran-Struktur eine isomere, bei 154 schmelzende Verbindung, die einen Chinolizidin-Ring enthält („Chinolizidin-Derivat A“, 3A). Ein isomeres, bei 152° schmelzendes „Chinolizidin-Derivat B“ (3B) bildet sich, wenn die Kondensation von 1b in essigsaurer Lösung bei 90° durchgeführt wird. Die Verbindungen 3A und 3B spalten in essigsaurer Lösung bei Gegenwart von Dimedon 1 Mol. Formaldehyd ab unter Bildung eines bei 96° schmelzenden Ketons C14H24N2O (4A; Schmp. 96), aus dem sich mit Formaldehyd das Chinolizidin-Derivat A zurückbildet (Schema 1). Die Konfiguration an den Asymmetriezentren des Ketons 4A liegt demnach auch in 3A vor. Die Konstitution des Ketons 4 folgt daraus, daß beim Ersatz des Sauerstoff-Atoms durch Wasserstoff über stereoisomere Alkohole (5) und Chloride (6) eines der vier möglichen stereoisomeren Racemate des 3-[2-Piperidyl]-chinolizidins (7) entsteht, welches identisch ist mit dem bereits beschriebenen „Racemat C“ bekannter Konstitution aber noch unbekannter Konfiguration (Schema 2). Beiden Chinolizidin-Derivaten kommt die Konstitution 3 zu; die Halbketal-Gruppierung ist durch das Fehlen der Carbonyl-Bande im IR-Spektrum und durch die Bildung je eines mit Säure leicht verseifbaren Methyläthers nachgewiesen. - Die Isomerie der Chinolizidin-Derivate 3A und 3B beruht darauf, daß das beiden zu Grunde liegende Gerüst von 4 in seiner Konfiguration bei 3B verschieden ist von dem in 3A und im Keton 4A vorliegenden Gerüst. Die Konfiguration 4A ist offenbar die stabilere und bildet sich auch unter sterischer Umlagerung aus dem beim Abbau von 3B in essigsaurer Lösung zu erwartenden, aber nicht isolierbaren Keton 4B, das, wie 4A, ein an den β-Kohlenstoff-Atomen leicht isomerisierbares β-Amino-keton ist. - Die Umlagerung 4B → 4A bleibt aus, wenn 3B mit Lithiumalanat reduziert oder mit Phenylisothiocyanat umgesetzt wird. Das in letzterem Fall unter Abspaltung von 1 Mol. Formaldehyd entstehende, sterisch offenbar von 4B abgeleitete Reaktionsprodukt ist verschieden von der isomeren, analog sowohl aus 3A als auch aus 4A erhaltenen Verbindung. - Die Reduktion von 3A und 3B mit Lithiumalanat - bei 3A auch nach Clemmensen - führt ohne Verlust eines C-Atoms zu je einem epimeren Paar von 2-Hydroxy-3-[1-methyl-2-piperidyl]-chinolizidinen (9). Die aus 3A entstehenden Epimeren geben beim Ersatz der Hydroxy-Gruppe über das Chlorid durch Wasserstoff 3-[1-Methyl-2-piperidyl]-chinolizidin (11A), das auch durch Methylierung des „Racemats C“ erhalten wird, und daher in der Konfiguration seiner drei Asymmetriezentren dem „Racemat C“ und damit 4A sowie 3A entspricht (Schema 3). Die aus 3B entstehenden epimeren Alkohole 9 geben analog das andere stereoisomere 3-[1-Methyl-2-piperidyl]-chinolizidin (11B). - Die wahrscheinlichsten Konfigurationen und Konformationen der Verbindungen werden besprochen.
