    Notes: Abstract Genetic polymorphism in C4 in the chimpanzee was studied by agarose gel electrophoresis of desialated plasma and development of patterns by immunofixation with antiserum to human C4 and by a C4-sensitive hemolytic overlay. In general, immunofixation patterns showed multiple partially overlapping bands of which only the most cathodal had strong hemolytic activity. In analogy to human C4, the latter were designated C4B, whereas those detected by immunofixation which had little hemolytic activity were designated C4A. Chimp C4A and C4B reacted with human and mouse (monoclonal) anti-C4B and human anti-Ch1 but neither reacted with monoclonal anti-C4A or human anti-Ch2, Ch3, Rg1, or Rg2. On sodium dodecyl sulfate polyacrylamide gel electrophoresis, the alpha chain of C4B showed a slightly lower apparent relative mass than that of C4A at around M r 93 000. There were three C4A variants and two C4B variants inherited in families as autosomal codominant traits, as C4A-C4B cosegregating pairs with no detectable crossing-over. These pairs were inherited with chimpanzee leukocyte antigen types C2 and BF variants without detectable crossing-over. Half-null C4 haplotypes with C4B *Q0 were observed in family studies. Nine BF, C2, C4A, C4B allelic haplotypic combinations (complotypes) were identified among presumably unrelated chimpanzees.

    Notes: Representative microorganisms of the diverse flora associated with several varieties of surface-ripened cheese were isolated. Species of streptococci, micrococci, yeasts and Gram-positive rods accounted for most of the isolates. These were incubated at 32°C in media containing 0.5 ppm lindane, p-p DDE, and p-p DDT. The pesticides were dissolved in a small amount of ethanol to facilitate incorporation into the media. Following incubation for 10 days, the concentrations of residual pesticides were determined by electron capture gas chromatography. The growth of streptococci and micrococci isolates did not apear to produce any changes in pesticide levels, though they apparently were uninhibited by the pesticides. However, the growth of other isolates dechlorinated p-p DDT. Moreover, aerobic growth of geotrichum species caused almost complete disappearance of the p-p DDT and p-p DDE peaks from the gas chromatograms. These results suggest the possibility that residues of DDT and DDE might be degraded if they are present in certain types of surface-ripened cheeses.

    Notes: The steady state photocarrier grating measurement technique has been used and is shown to be valid for the characterization of steady-state transport properties of minority carriers in undoped improved quality hydrogenated amorphous germanium (a-Ge:H). Deviations from the theoretically predicted behavior of the measurement have been observed on some a-Ge:H samples. At room temperature in a-Ge:H, the ratio of photoconductivity to dark conductivity is less than 1. The high density of thermally generated carrier is observed to affect the measurement by reducing its sensitivity. Whether it is also the cause of the observed deviation will be discussed.

    Notes: Background Nitric oxide (NO) is believed to participate in the regulation of airway clearance and non-specific cellular immunity. Recent studies have suggested that airway epithelial cells of allergic and non-allergic individuals may differ in their ability to produce this molecule.Objective The aim of this study was to detect the difference in NO production in human nasal epithelial cells between normal subjects and patients with perennial allergic rhinitis (AR), and to assess the relationship between the expression of nitric oxide synthase (NOS) isoforms and the severity of the disease.Methods Nasal epithelial cells were obtained from the inferior turbinate. The expression of mRNAs encoding constitutive endothelial NOS (eNOS) and inducible NOS (iNOS) was studied by reverse transcription-polymerase chain reaction (RT-PCR). Direct NO production in living cells was visualized and quantified by a fluorescent indicator, DAF-2 DA.Results RT-PCR analysis demonstrated that AR patients with a RAST score of 5 or 6 showed significant increases in the levels of iNOS mRNA and slight reductions in those of eNOS mRNA. Patients with a RAST score of 2–4 also revealed the same tendency however, the difference was not significant. DAF-2 DA imaging demonstrated that epithelial cells, especially the ciliated cells, produced a larger amount of NO than non-epithelial inflammatory cells. Preincubation with L-NAME resulted in an approximate 40% decrease in both groups.Conclusion These results directly indicate that nasal epithelial cells of AR patients overall produce higher levels of NO through the concomitant expression of different NOS isoforms. Continuous NO production by the epithelial cells in normal subjects further support the hypothesis that NO derived from epithelium may play dual roles in the regulation of nasal airway clearance and in the host defense. In addition, the use of DAF-2 DA provides a reliable method to visualize and quantify the direct NO production of living cells.
