    Notes: Growth kinetics of Aspergillus niger strain A.n./61 on a solid medium containing beet pulp were examined.Solid state fermentation was carried out in laboratory microfermenter (aerated packed columns). During the experiments the CO2 evolution, protein biosynthesis, as well as cellulose and sucrose utilization were determined.It was found that CO2 evolution evidently increased in the logarithmic phase, together with the protein biosynthesis. Between 30-32 h of culture CO2 evolution and protein content in product were the greatest. Subsequently, CO2 evolution decreased.During 28 h of culture, cellulose was slightly utilized, the strain using mainly sucrose. Intense utilization of cellulose was paralleled by a drop in CO2 evolution.

    Notes: The aim of the investigations was to obtain  -  from the parent strain Aspergillus niger A.n. 33  -  selectants with an increased ability of cellulolytic enzymes biosynthesis. Own selection methods allowed to receive two selectants A.n. 33/2 and A.n. 33/20 characterized by enhanced activities of saccharifying cellulase (respectively 0.11 and 0.14 FPU/cm3), endo-beta-1,4-glucanase (15.4 and 21.8 U/cm3) and cellobiase (0.6 and 1.4 IU/cm3) as compared with the parent strain (FPA  -  0.09 IU, CMC  -  8.2 U and CB  -  0.1 IU/cm3). Moreover, the selectants differed in shape and size of conidial heads, in shape and colour of conidia, as well as in structure and shape of hyphae. Enzyme preparations obtained after ultrafiltration of liquid cultures were characterized by following activites: FPA-4-16 IU, CMC  -  900-1800 U, CB  -  60-120 IU and xylanase  -  250-280 IU/cm3.

    Notes: A fungal strain suitable for protein enrichment of starchy raw materials was selected by evaluation of the growth rate, results of protein biosynthesis in solid state fermentation (SSF) and assessment of fungal biomass. The strain Aspergillus oryzae A.or. 11 selected for further studies was characterized by the radial growth rate Kr of about 300 μm/h and by the specific growth rate μ of about 0.100 h-1. Fungal biomass contained about 32% of crude protein in dry matter. The digestibility of this protein in vitro was close to 75%. Protein analysis for amino acids showed that the content of exogenous amino acids approached that in protein of the FAO standard. As a result of 34  -  h culture of this strain in solid medium, net protein increased by about 5.0 g/100 g of starting medium d.m. at the cost of decomposition of about 20 g carbohydrates.

    Notes: Study was made of the effect of medium humidity, source and dose of nitrogen, dose of phosphorus, dose of inoculum and aeration on protein biosynthesis by strain Aspergillus oryzae A. or. 11 in solid state fermentation. It was found that for a maximal protein yield the medium ought to contain about: 60% of water, 36% of starchy raw materials d. m. (e.g. 28.8% of coarse rye meal and 7.2% of beet pulp), 0.85% of nitrogen sources (1.6% of (NH4)2SO4 and 1.1% of urea), and 0.35% of phosphorus source (1.3% of KH2PO4). pH of medium should be near 6.5. The dose of inoculum should not be lower than 108 spores/100 g medium.The duration of culture ought to be 20-22 h at 30-35°C, with aeration at least 40 dm3 of air/h × 1000 g medium. Under these conditions the protein yield is about 6.0-6.3 g/100 g of starting medium d.m. at the cost of utilization of about 25 g total carbohydrates.

    Notes: The effect of the culture method in solid medium and the influence of starch-containing raw materials on the yield of fungal protein biosynthesis were studied. Three procedures (laboratory bioreactor method, tray method and pile method) gave satisfactory results. Protein yields amounted to 5.0-5.9 g/100 g of starting medium d.m. upon utilization of 20.9-24.1 g carbohydrates. The procedure involving the use of a fermenter with a mixer afforded protein yields by about 50% lower as compared with the three above-mentioned procedures, and therefore it requires technical improvement.As a result of fungal culture using various starch-containing raw materials, the protein content in post-culture products increased by 46-88%, as compared with starting medium. The contents of 13 amino acids (including some exogenous ones which increased by 52-82%) in post-culture products substantially rose (by 34-63%). The post-culture products exhibited proteolytic activity (1260-3500 HU/g of d.m.).The kind of the source of starch evidently influenced protein biosynthesis. Media based on potatoes afforded the greatest increases in protein (5.8-5.9 g/100 g of starting medium d.m.), and those containing coarse rye meal and milling by-products  -  the smallest ones (4.5-4.6 g/100 g of starting medium d.m.).

    Notes: The object of the presented studies was to determine the conditions of protein biosynthesis from lignocellulosic raw material using filamentous fungi.It was found that on beet pulp + 50% wheat straw, strain A. niger (A.n./61) shows the highest effectivity of protein biosynthesis. Using this strain, after 48-hour incubation the following results were obtained: increase in protein 4.43 g/100 g·DM, daily protein increase coeffizient 2.22 g/100 g DM x 24 h, protein yield (based on the decrease in dry matter) 22.3% and cellulose utilization 37%.Upon use of a new strain CEL 2 and milled wheat straw as the substrate, the best results obtained after 74-hour incubation were as follows: increase in protein 1.82 g/100 g DM, daily protein increase coefficient 0.61 g/100 g DM x 24 h, protein yield 15,8% and cellulose utilization 22.0%.For rape straw and strain Sporotrichum pulverulentum, after 192-hour cultivation the following results were obtained: increase in protein 5.4 g/100 g DM, daily protein increase coefficient 0.68 g/100 g DM x 24 h, protein yield 14% and cellulose utilization 50.2%.

    Notes: The influence of long-term storage on the stability of the cellulolytic enzyme and protein biosynthesis ability, was determined.Microorganisms belonging to genera Aspergillus, Fusarium, Penicillium, Sporotrichum, Allescheria and Mucor were used. The strains were stored on wort agar slants, on SAUNDER'Smedium and on a medium according to MANDELS-WEBER, under paraffin oil or without it. After 3 years storage no influence of the storage method on the biosynthesis of cellulolytic enzymes (strain A. niger 33/2 and 33/20) was found. A systematic increase in the celluloslytic activity was exhibited by strains A.n. 33/2 and A.n. 33/20 on wort agar (40 and 98%, respectively) and on SAUNDER'Smedium (79 and 66%, respectively), as well as by the new strain CEL 2 on Saunder's medium without paraffin oil.
