    Notes: Abstract In the shoot apices of the haploid Pelargonium cultivar „Kleine Liebling“, all mitoses are haploid (n = 9); however, ca. 20% of the interphase nuclei have DNA contents greater than 2C (up to 4C), indicating a tendency to chromosome endoreduplication in this material. — In internodes in vivo, the few mitoses present are haploid (quite probably, cambium cells); in addition to haploid interphases (1C to 2C DNA contents), endoreduplicated (endopolyploid) nuclei and nuclei in the course of endoreduplication occur with a frequency of ca. 40–50% (DNA contents up to 8C). — When internodes are cultured in vitro, differentiated cells are stimulated to divide, thus forming a population of diploid and tetraploid mitoses in addition to the preexistent meristem (haploid) cell population. In the process of time, diploid and tetraploid mitoses continue to be present in the callus, whilst haploid mitoses may decrease in number and eventually disappear. All mitoses analyzed had euploid chromosome numbers (9, 18 and 36) and their DNA contents were correspondingly 2C, 4C and 8C. Since no extensive chromosome counts were made, aneuploidy in the cultured material cannot be excluded; but, if occurring, it should be rather rare. — Under the experimental conditions used, prolonged culture in vitro leads to the production of nuclei with DNA contents (16C and 32C) greater than those occurring in vivo (8C), due to one and two additional DNA replications respectively beyond the limits attained in vivo. Even in these cultures, however, a population of the meristematic haploid cell line (DNA values 1C to 2C) is still present. — The present results are discussed in their relations with previous works on nuclear conditions in vivo and in vitro and on regeneration processes in cultured tissues in plants.

    Notes: Abstract Tritiated ribosomal RNA (rRNA) was prepared from hypocotyls of Phaseolus coccineus grown in liquid culture in the dark and in presence of 5-3H-uridine. A mixture of the 18S and 25S 3H-rRNA fractions was used for hybridization with DNA in the polytene chromosome cells of the embryo suspensor of P. coccineus. It was shown that the ribosomal cistrons (rDNA) are located in the nucleolus organizing system (satellite, nucleolar constriction and organizer) of the satellited chromosome pairs I (S1) and V (S2), in the proximal heterochromatic segment of the long arm of chromosomes S1 and in the terminal heterochromatic segment of chromosome pair II. The micronucleoli which are produced by the satellite and nucleolus organizer of the chromosome pair S1 contain rDNA; on the contrary, no rRNA-DNA hybridization is found in the DNA containing granules which are produced by the satellite and nucleolus organizer of chromosome pair S2. The DNA which is amplified during production of DNA puffs at some chromosomal regions apparently does not code for ribosomal RNA (no detectable rRNA-DNA hybridization).

    Notes: Abstract Homologous tritiated 25S, 18S and 5.8S rRNAs were used separately for in situ hybridization to the polytene chromosomes of the embryo suspensor cells of Phaseolus coccineus. Hybridization occurred at the same chromosomal sites which were labeled in previous in situ hybridization experiments with 25+18S rRNAs in the same material (Avanzi et al., 1972), namely: nucleolus organizing system (satellite, nucleolar constriction and organizer) of chromosome pairs I (S1) and V (S2), proximal heterochromatic segment of the long arm of chromosome pair I, and terminal heterochromatic segment of chromosome pair II. Competition hybridization experiments confirmed for P. coccineus the high sequence homology between 25S and 18S rRNA already known for other plants. Homologous 125I-5S rRNA was found to hybridize to three sites in the polytene chromosomes of P. cocdneus: the proximal heterochromatic segment in the long arm of chromosome pair I (which also bears the sequences complementary to 25S, 18S and 5.8S RNAs), most of the proximal heterochromatic segment plus a small portion of adjoining euchromatin in the long arm of chromosome pair VI and the large intercalary heterochromatic segment in the same chromosome pair. Simultaneous labeling of the two 5S RNA sites in chromosome VI was quite rare (3%), the rule being labelling of one site to the exclusion of the other, with a labeling frequency of 43.7% and 53.3% for sites no. 1 and no. 2 respectively. These results are interpreted as being due to differential hybridizability of chromosomal sites such as described in other materials.

Le energie di soglia sono state confrontate con quelle del para-idrogeno, misurate nello stesso apparato, e i fattori di scala sperimentali sono discussi.
    Abstract: Резюме Вынужденное вращательное Рамановское рассеяние излучения CO2 лазера в орто-D2 представляет когерентный источник высокой интенсивности в инфракрасной области от 11 до 14 мкм. Получается максимум энергии на выходе ≃1 Дж для энергии накачки 4.0 Дж. Пороговые энергии сравниваются с данными, измеренными той же аппаратурой, в случае пара-H2. Обсуждаются экспериментальные масштабные факторы.
    Notes: Summary Stimulated rotational Raman scattering of CO2 laser radiation in ortho-D2 provides a high-power coherent source in the infrared region from 11 to 14 μm. A peak output energy of ≃1 J has been obtained for a pump energy of 4.0 J. Threshold energies have been compared with those of para-H2 measured in the same apparatus and experiment scaling factors are discussed.

    Notes: Summary In this paper the realization is described of CO2 laser with high energy (≈10J/pulse), long pulses (≈2μs) and narrow bandwidth (≈200 kHz), using suitable discharge device (Lumonics 602) and cavity configuration (modified self-filtering unstable resonator). The theoretical considerations which led to these choices are also discussed and the results obtained are shown.

    Notes: Coherent operation of an array of AlGaAs diode lasers was obtained by placing the array in an external cavity which made use of the Talbot self-imaging effect to couple the laser diodes together. A spatial filter was required to suppress oscillation of the highest order mode of the array. This filter introduced no significant loss to the cavity mode, and the mode was observed to be stable up to the maximum rated drive current for the device.

    Notes: The spectrum of CCl3F (trichlorofluoromethane, CFC 11) has been measured in the region of the ν1 fundamental (1050–1120 cm−1) by high resolution interferometric Fourier-transform spectroscopy [0.004 cm−1 bandwidth full width at half maximum, apodized] and by diode laser spectroscopy (bandwidth 0.0008–0.0020 cm−1 FWHM) at room temperature, in cold cells and in supersonic jet expansions. Fourier-transform infrared and diode laser spectra of isotopically pure C35Cl3F have been recorded at room temperature in static cells. The C35Cl3F spectra allowed an analysis of the rotational structure to be started successfully for the first time. The rotational analysis of the isotopic species C35Cl237ClF and C35Cl37Cl2F, which are asymmetric rotors, was initiated from diode laser spectra of natural CCl3F with a rotational temperature of about 20 K (5% seeded in He) resulting from expansion in a supersonic pulsed slit jet. The rotational analysis yielded effective Hamiltonian constants including accurate band centers for the three most abundant isotopomers C35Cl3F (ν1=1081.2801 cm−1), C35Cl237ClF (ν1=1080.7330 cm−1), and C35Cl37Cl2F (ν1=1080.0663 cm−1), excited state rotational constants, and for C35Cl3F, quartic centrifugal distortion constants. The results are important for and discussed in relation to simulations of atmospheric absorption, line coincidences with CO2 lasers for sub-Doppler spectroscopy, as well as IR multiphoton excitation and infrared laser chemistry. © 1995 American Institute of Physics.
