    Notes: Apelin peptides have recently been identified to be the endogenous ligands for the G protein-coupled receptor APJ. However, little is known about the physiological roles of this ligand-receptor pairing. In the present study we investigated the pharmacology of several apelin analogues at the human recombinant APJ receptor using radioligand binding and functional assays. This has led to the identification of key residues in the apelin peptide required for functional potency and binding affinity through structure–activity studies. In particular, we have identified that replacement of leucine in position 5, or arginine in position 2 and 4 of the C-terminal apelin peptide, apelin-13, resulted in significant changes in pharmacology. We also investigated the detailed localization of pre-proapelin and APJ receptor mRNA in a wide range of human, rat and mouse tissues using quantitative RT–PCR, and carried out a detailed immunohistochemical study of the distribution of the APJ receptor in rat brain and spinal cord. Interestingly, the APJ receptor was not only co-localized in white matter with GFAP in the spinal cord, but was also clearly localized on neurones in the brain, suggesting that this receptor and its peptide may be involved in a wide range of biological process yet to be determined.

    Notes: In this paper, an important but frequently overlooked aspect of production batch-size planning is addressed: namely, the consideration of unit load and material handling. Thus, an additional cost factor to account for the transportation of unit loads is incorporated in the total cost function which is typically broken down into reorder cost, inventory holding cost, and backorder cost. A cost minimization model is developed that includes constraints related to volume and weight restrictions on unit loads and in-process storage space. A dynamic programming procedure is utilized to find the minimum cost solution. An example problem is also given.

    Description / Table of Contents: Résumé Les malades qui présentent des troubles vésiculaires alors que la cholecystographie et l'ultrasonographie vésiculaire sont négatives continuent à soulever des problèmes en ce qui concerne l'attitude thérapeutique. La littérature chirurgicale fait état d'attitudes divergentes car le taux élevé de récidives des symptômes accusés par les opérés dans certaines séries n'est pas sans poser de difficiles problèmes. Pour notre part, nous avons essayé de définir le traitement approprié au cours de la dernière décennie. Au début la constatation de l'imprégnation prolongée de la vésicule après l'épreuve du repas gras fut considérée comme l'indication à intervenir. Soixante-quinze pourcent des malades qui furent opérés en se basant sur ce critère n'ont présenté aucun symptôme au cours d'une décade. Ultérieurement les autres sujets furent soumis à la cholecystographie orale après injection de cholecystokinine. En les suivant pendant 6 ans, 80% des opérés ne présentaient plus aucun trouble. Les tentatives continues pour mieux définir les sujets susceptibles de bénéficier de la chirurgie nous ont conduit à employer la scintigraphie associée à l'injection de cholecystokinine. Une centaine de patients furent soumis à cette épreuve; 76 présentaient une évacuation vésiculaire anormale, 30 dont la vésicule ne s'évacuait pas furent opérés. L'examen histologique de leur vésicule montra toujours des altérations de la paroi vésiculaire. Ils n'ont accusé aucun trouble de 1 an à 3 ans et demi et 6 mois après la cholecystectomie.
    Abstract: Resumen Los pacientes con síntomas clásicos de enfermedad de la vesícula biliar y colecistograma oral (CGO) negativo y, más recientemente, ultrasonografía negativa, continÚan siendo motivo de perplejidad para el cirujano. El manejo de la enfermedad crónica acalculosa de la vesícula biliar ha sido motivo de controversia por muchos años. Su naturaleza enigmática se debe a una falta de definición de tal entidad. Se acepta que no existe patología mascroscópica aparente en ciertos pacientes con ataques bien documentados de cólico que simula el cólico biliar y que presentan hallazgos negativos en su evolución. La literatura quirÚrgica señala que existen marcadas diferencias de opinión en cuanto al papel de la cirugía en estos pacientes. La alta tasa de recurrencia de los síntomas en los pacientes operados que informan algunas series ha sido motivo de preocupación mayor. En el curso de la Última década hemos intentado definir un enfoque apropiado de este problema. Inicialmente consideramos que el hallazgo de una prolongada visualización (evacuación retardada) de la vesícula biliar en la CGO después de una comida grasa era indicación para cirugía. Este grupo inicial, seguido por una década, mostró estar asintomático en un 75%. Más tarde un grupo adicional fue sometido a colecistografía oral con colecistokinina (CGO-CCK). En un seguimiento de más de 6 años los resultados demuestran un 80% de desaparición de la sintomatología de tracto biliar en estos pacientes (categoría CGO-CCK) después de la cirugía. El continuado esfuerzo por definir aquellos pacientes que puedan beneficiarse de la cirugía ha llevado a la utilización de imagenología con radionÚclidos asociada con la inyección de colecistokinina (IRN-CCK). Cien pacientes han sido sometidos a la imagenología cuantitativa por medio de CCK. De estos pacientes, en 76 casos se consideró que existía vaciamiento inadecuado de la vesícula biliar; treinta pacientes con vaciamiento nulo en la IRN-CCK fueron sometidos a colecistectomía, y todos exhibieron algÚn grado de anormalidad histológica en la vesícula biliar. Tales pacientes han sido seguidos entre 1 y 3 1/2 años y no se ha presentado recurrencia de los síntomas en ninguno de ellos.
    Notes: Abstract Patients with classic symptoms of gallbladder disease who have negative oral cholecystograms (OCG) or ultrasonography present perplexing management problems. The surgical literature attests to the fact that there is a marked difference of opinion as to the role of surgery in these patients. The high recurrence rate of symptoms in patients following surgery reported in some series has been a source of major concern. We have attempted to define the appropriate approach to this problem for the past decade. Initially, the findings of prolonged visualization of the gallbladder on OCG in association with a fatty meal was considered an indication for surgery. This initial group of patients followed over a decade showed about 75% to be symptom free. Subsequently, additional patients were subjected to cholecystokinin oral cholecystography (CCK-OCG). With follow-up for over 6 years, the results have shown about 80% relief of biliary tract symptoms in these patients (CCK-OCG category) following surgery. Continued attempts to further define those patients who will benefit from surgery has resulted in the utilization of radionuclide imaging in association with cholecystokinin injection (CCK-RNI). One hundred patients were subjected to CCK quantitative biliary imaging. Seventy-six patients were considered to have inadequate gallbladder emptying; 30 patients with no gallbladder emptying on CCK-RNI were subjected to cholecystectomy. All patients subjected to cholecystectomy showed abnormal histological findings in the gallbladder. These 30 patients have been followed from 1 to 3 1/2 years and there has been no recurrence of symptoms in any following surgery.

    Notes: Abstract This paper investigates the solution of manufacturing scheduling problems using different distributed computer schemes. The distributed computer schemes vary with respect to task partitioning, task allocation and the number of processors used. The best combinations of these variations are presented with respect to problem scenario and size. The distributed computing models, are analyzed with respect to computational advantages gained vs. traditional computing models. The models are implemented over a distributed computer network, designed for heterogeneous application, and are transportable to other operating systems and networks of computers. The advantages of these models are discussed and several useful extensions are presented.

    Notes: Metabolic control reactions have been studied in the intact toad bladder by means of fluorescence spectrophotometric measurement of reduced pyridine nucleotide and by measurement of respiration with the platinum electrode. substrates such as pyruvate and succinate lead to prompt increases in reduction level of pyridine nucleotide with only slight acceleration of respiration. major metabolic control is exerted by adp, which depletes the intact bladder of reduced pyridine nucleotide and accelerates respiration. respiratory control ratios, as for isolated mitochondria, depend upon the substrate being metabolized. a significant fraction of added adp appears to gain entry into the intact toad bladder and is converted to atp, anaerobiosis and amobarbital lead to increased levels of reduction of pyridine nucleotide. the spectroscopic and metabolic properties of the reduced pyridine nucleotide being studied identify it with that fraction of dpnh which is bound at one of the energy conservation sites linking phosphorylation reactions with electron transfer.
