    Notes: Summary Previous studies aimed at elucidating day/night changes of pineal gland size and pinealocyte nuclear volume have yielded contradictory results. The aim of the present investigation was to examine the above parameters over a period of 10 successive days, at 6-hour intervals under a lighting regimen of LD 12∶12 (7 a.m. to 7 p.m.). It was found that changes in pineal gland volume could not be correlated with the light/dark phases; instead, a rhythm with a two-day period was encountered. Pinealocyte nuclear volume, by contrast, showed clear circadian changes, exhibiting troughs during photophase and peaks during scotophase. In this experimental series, the karyometric results conformed to the melatonin forming rhythm in the rat pineal gland as reported in the literature.

Die zusätzliche Behandlung mit immunostimulierenden Substanzen (BCG, Albumin, Vitamin A-Säure) bzw. immunodepressiv wirkenden Substanzen (Hydrocortison, Cyclophosphamid, Methotrexat), beginnend am 6. Tage nach der Geburt und durchgeführt bis zum Lebensende der Tiere, vermochte die Carcinogenese weder bezüglich der Häufigkeit der entstehenden Sarkome noch der Induktionszeit zu beeinflussen.
    Notes: Summary After subcutaneous application of 0.5 mg 3,4-benzopyrene (BP) to Sprague-Dawley-rats on the 2nd day of life, 50% of the animals developed local fibrosarcomas after 250±70 days. Additional treatment with immunostimulating (BCG, albumin, vitamin A-acid) or immunodepressive agents (hydrocortisone, cyclophosphamide, methotrexat) which was started 6 days after birth and maintained throughout life, did not influence carcinogenesis with respect to tumor incidences and induction periods of tumors.

Lebenstag, entwickelten ∼90% der Tiere nach ∼330 Tagen Adenocarcinome des Dickdarms (72%), Plattenepithelcarcinome des Gehörgangs (17%). Adenosarkome der Niere (13%) sowie hepatocelluläre Carcinome (11%). Zusatzbehandlungen mit Immunodepressiva (Cyclophosphamid, Methotrexat, Hydrocortison), Immunostimulantien (BCG, Albumin, Vitamin A-Säure) oder Enzymstimulatoren (Luminal) änderten die Tumorausbeuten und Induktionszeiten nicht. Nach Applikation einer vegetarischen Diät waren die Ausbeuten an Nieren- und Leberkrebsen signifikant vermindert und die Induktionszeiten der Darm- und Gehörgangskrebse vergrößert.
    Notes: Summary After ten times monthly, subcutaneous, injections of 30mg/kg 1,2-dimethylhydrazine (DMH) to Sprague-Dawley-rats, malignant tumors were found in ∼90% of the animals after a mean induction period of ∼330 days. Injections were started on the 2nd day of life and maintained during 10 months. 72% of the tumors induced were formed in the colon, 17% were squamous cell carcinomas of the ear duct, adenosarcomas of the kidney and hepatocellular carcinomas were found in 13 and 11% of the animals, respectively. Additional treatment with immunodepressive (cyclophosphamide, methotrexat, hydrocortisone) and immunostimulating substances (BCG, albumin, vitamin A-acid) as well as an enzyme-stimulating agent (Luminal) did not alter incidences and induction periods of tumors. After application of a vegetarian diet, the rate of liver and kidney tumors was diminished significantly and induction periods of intestinal and ear duct tumors increased.

    Description / Table of Contents: Zusammenfassung Frühere Versuche an Ratten und Mäusen hatten gezeigt, daß die akute Toxicität von Dimethylnitrosamin (DMNA) und Diäthylnitrosamin (DÄNA) durch Behandlung der Tiere mit Tetraäthylthiuramdisulfid [Disulfiram, Antabus (DSF)] vermindert wird. Im chronischen Versuch an 40 Ratten wirkte DSF in einer Dosierung vom 500 mg/kg/Woche nicht carcinogen, seine Gabe beeinflußte aber die Wirkung der untersuchten Nitrosamine. DÄNA, an 38 Tieren in einer Dosierung von 20 mg/kg/Woche verabreicht, induzierte bei 90% der behandelten Ratten maligne Tumoren der Leber, in 29% wurden neben einigen prämalignen Formen überwiegend Carcinome des Oesophagus beobachtet. DMNA erhielten 31 Ratten in einer wöchentlichen Dosierung von 4 mg/kg/Woche. In dieser Gruppe zeigten sich bei 55% der Versuchstiere Tumoren in der Leber. 26 Ratten wurden mit einer Kombination von 500 mg/kg/Woche DSF und 20 mg/kg/Woche DÄNA behandelt. Nur bei 31% der Tiere dieser Gruppe ließen sich Tumoren im Bereich der Leber nachweisen, aber 81% von ihnen entwickelten neben einigen prämalignen Formen überwiegend Oesophaguscarcinome. Bei der kombinierten Applikation von 500 mg/kg DSF/Woche und 4 mg/kg DMNA/Woche fanden sich bei einer Tierzahl von 29 lediglich ein Tumor in der Leber (=3%), aber in 59% Plattenepithelcarcinome der Nasennebenhöhle; diese wurden bei alleiniger Gabe von DMNA nicht beobachtet.
    Notes: Summary In former studies in rats and mice it was shown that the acute toxicity of dimethylnitrosamine (DMNA) and diethylnitrosamine (DENA) is reduced by additional treatment with tetraethylthiuramdisulfide [(Disulfiram, Antabus® (DSF)]. In long-term experiments in 40 rats given 500 mg/kg DSF/ week the substance did not show carcinogenic effects, however, influenced the action of the nitrosamines. DENA, which was administered to 38 rats in a dosage of 20 mg/kg/week, induced liver tumors in 90% of the animals; in 29% besides some precancerous stages predominantly malignant carcinomas of the oesophagus were seen. DMNA when given to 31 rats in a dosage of 4 mg/kg/ week induced liver tumors in 55% of the animals. 26 rats were treated with a combination of 500 mg/kg DSF/week and 20 mg/kg DENA/week. In only 31% of the animals of this group liver tumors were found, however, 81% of them besides some precancerous stages showed predominantly carcinomas of the oesophagus. The combination of 500 mg/kg DSF/week and 4 mg/kg DMNA/week induced only 1 liver tumor (=3%) out of a group of 29 animals, whereas 59% of them showed squamous cell carcinomas of the paranasal sinus that were not seen when DMNA was given alone.

    Notes: Summary In anaesthetized rats, the influence of an experimental inflammation and of acetylsalicylic acid (ASA) on the discharge properties of muscle receptors with slowly conducting afferent fibres was studied using a single-fibre recording technique. Following the induction of a myositis with carrageenan, the proportion of units having background activity and the frequency of the background discharge were significantly increased. The latter change was particularly prominent in high-threshold mechanosensitive (HTM) units. There was evidence for an inflammation-induced lowering of mechanical threshold in HTM units, but the change was not statistically significant. Administration of ASA intravenously led to a decrease in the frequency of background discharge in some units while others were unaffected, although they appeared to be sensitized by the inflammation. If one assumes that at least some of the HTM receptors fulfil nociceptive functions, the results suggest that the pain and tenderness of an inflamed muscle is largely due to a sensitization and hence increased activity of nociceptive muscle receptors. The sensitization is only partially abolished by ASA.

    Notes: Abstract Imaging cerebral GABAA receptor density (GRD) with single-photon emission tomography (SPET) and iodine-123 iomazenil is highly accurate in lateralizing epileptogenic foci in patients with complex partial seizures of temporal origin. Limited knowledge exists on how iomazenil SPET compares with magnetic resonance imaging (MRI) in this regard. We present a patient with complex partial seizures in whom MRI had identified an arachnoid cyst anterior to the tip of the left temporal lobe. Contralaterally to this structural abnormality, interictal electroencephalography (EEG) performed after sleep deprivation disclosed an intermittent frontotemporal dysrhythmic focus with slow and sharp waves. On iomazenil SPET images GRD was significantly reduced in the right temporal lobe and thus contralaterally to the MRI abnormality, but ipsilaterally to the pathological EEG findings. These data suggest that iomazenil SPET may significantly contribute to the presurgical evaluation of epileptic patients even when MRI identifies potentially epileptogenic structural lesions.
