    Description / Table of Contents: Summary The author presents a report on eight female patients with various hematological diseases (primary and secondary thrombopenia, thrombasthenia) in whom it was possible to control severe meno- and metrorrhagia by ovulation inhibitors (the combination drugs Anovlar or Lyndiol). The treatment may be continued for a long period of time without detrimental effects on the patient. The effect of the drug is readily reversible.
    Notes: Zusammenfassung Es wird über 8 Patientinnen mit verschiedenen hämatologischen Erkrankungen (primäre und sekundäre Thrombopenic, Thrombasthenie) berichtet, bei denen schwere Meno- und Metrorrhagien durch Ovulationshemmer (Kombinationspräparate Anovlar oder Lyndiol) unterdrückt werden konnten. Die Behandlung kann ohne Schaden für die Patienten über einen langen Zeitraum fortgesetzt werden. Die Wirkung der Präparate ist prompt reversibel.

    Description / Table of Contents: Summary The effect of epinephrine after blocking ofβ-receptors (Propranolol) was studied on three healthy males by measurement of the blood pressure and some cardiac intervals. This was carried out before and after intravenous application of atropine. 1. After drug induced blockade of adrenergicβ-receptors epinephrine caused an increase of mean blood pressure, a decrease of heart rate and a prolongation of the isometric contraction time. 2. This bradycardia and prolongation of isometric contraction time could be prevented by atropine. 3. The shortening of the pulse wave velocity remained unchanged after atropine, the increase of mean bloodpressure was more marked. 4. It is assumed, that the effects eliminated by atropine are evoked by a reflex response through the efferent vagus nerves.
    Notes: Zusammenfassung An drei kreislaufgesunden Männern wurde die Wirkung einer Adrenalininfusion nach Ausschaltung derβ-Receptoren durch Propranolol auf Blutdruck und Mechanokardiogramm vor und nach intravenöser Gabe von Atropin geprüft. 1. Nach medikamentöser Blockade adrenergischerβ-Receptoren führt Adrenalin zu einem Anstieg des mittleren Blutdrucks einem Absinken der Herzfrequenz und einer Verlängerung der Anspannungs-und Druckanstiegszeit. 2. Die durch Adrenalin nachβ-Blockade ausgelöste Bradykardie sowie die Verlängerung von Anspannungs-und Druckanstiegszeit wird durch Atropinvorbehandlung verhindert. 3. Die Verkürzung der zentralen Pulswellenlaufzeit bleibt durch Atropin unbeeinflußt, der Blutdruckanstieg ist ausgeprägter. 4. Es wird angenommen, daß die durch Atropinvorbehandlung ausgeschalteten Effekte reflektorisch über efferente cholinerge Bahnen zustande kommen.

    Description / Table of Contents: Summary The infusion of propranolol, aβ-blocking agent, results in a decrease in the concentration of free fatty acids, a significant lowering of the turnover rate of free acids, and a significant decrease of the fractional turnover rate of free fatty acids in plasma. The esterification rate of free fatty acids to plasma triglycerides is significantly reduced. It is suggested that the action of propranolol on the metabolism of free fatty acids is based on the blockage of the action of endogenous catecholamines. The observed actions of propranolol on the metabolism of free fatty acids are considered to result from the inhibition of lipolysis in the adipose tissue.
    Notes: Zusammenfassung Durch die Infusion von Propranolol kommt es beim Menschen zu einer Senkung der Konzentration der freien Fettsäuren, zu einer signifikanten Erniedrigung der Umsatzrate der freien Fettsäuren und einer signifikanten Erniedrigung der „Fractional turnover rate“ der freien Fettsäuren im Plasma. Die Bildung der endogenen Plasmatriglyceride aus freien Fettsäuren wird signifikant erniedrigt. Es ist anzunehmen, daß die Wirkung von Propranolol auf den Stoffwechsel der freien Fettsäuren auf einer Blockierung der endogenen Katecholaminwirkung beruht. Alle Wirkungen von Propranolol auf den Fettsäurestoffwechsel lassen sich kausal auf eine Hemmung der Lipolyse im Fettgewebe zurückführen.

    Description / Table of Contents: Summary On seven individuals the eosinophils in the peripheral blood could be markedly lowered by an intramuscular injection of 0,3 mg epinephrine. The average decrease of eosinophils after four hours was 55%. This effect could be inhibited, if on the same experimental conditions 10 mg Propranolol was given 10 min before epinephrine. Consequently this decrease of eosinophils after epinephrine is probably mediated by adrenergicβ-receptors located in the hypothalamic area or the pituitary gland.
    Notes: Zusammenfassung An sieben Versuchspersonen wurde durch die intramuskuläre Injektion von 0,3 mg Adrenalin ein Eosinophilensturz von durchschnittlich 55% hervorgerufen. Dieser Adrenalineffekt konnte verhindert werden, wenn bei völlig gleicher Versuchsanordnung 10 min vor Adrenalin 10 mg Propranolol intravenös injiziert wurden. Der durch Adrenalin ausgelöste Eosinophilensturz dürfte demnach über adrenergeβ-Receptoren im Bereiche des Hypothalamas oder der Hypophyse vermittelt werden.

The effect of propranolol (an adrenergicβ-receptor blocking drug) and an intravenous infusion of epinephrine before and after propranolol was studied in five normal persons. Blood pressure, heart rate and some intervals of the cardiac cycle were measured. 2. After injection of propranolol a decrease of heart rate and a prolongation of the isometric contraction time was observed. 3. After blocking theβ-receptors by propranolol, an intravenous infusion of epinephrine was followed by a rise of the systolie and diastolic blood pressure, and an even more marked decrease of heart rate and prolongation of the isometric contraction time was noted. The velocity of the pulse wave was increased. 4. No change of Q-1 time and ejection time was observed. 5. The decrease of heart rate and the prolongation of the isometric contraction time after blocking theβ-receptors is suggested for the estimation of endogenous catecholamine effect.

Die Wirkung von Propranolol (eine adrenergischeβ-Receptoren blockierende Substanz) und einer intravenösen Adrenalininfusion vor und nach Propranolol wurde an fünf kreislaufgesunden Personen untersucht. Blutdruck, Herzfrequenz und Mechanokardiogramm wurden registriert. 2. Propranolol bewirkte bei allen Probanden eine Bradykardie und eine Verlängerung der Druckanstiegszeit. 3. Nach Blockade derβ-Receptoren führte Adrenalin zu einem Anstieg des systolischen und diastolischen Blutdrucks. Gleichzeitig nahm die Herzfrequenz weiter ab, die Druckanstiegszeit wurde länger und die zentrale Pulswellenlaufzeit kürzer. 4. Umformungs- und Austreibungszeit änderten sich nicht signifikant. 5. Die Frequenzabnahme und die Verlängerung der Druckanstiegszeit nachβ-Blockade wird zur Beurteilung der endogenen Katecholaminwirkung vorgeschlagen.

Phospholipids extracted from plasma of eight individuals without hepatic diseases and of six patients with acute hepatitis were separated by thinlayer chromatography and quantitatively determined. 2. The total amount of plasma phospholipids in patients with acute hepatitis was significantly increased. 3. The individual fractions of phospholipids in patients with hepatitis showed a different pattern: Lecithin was stronglyi ncreased, a relative decrease of sphingomyelin and an absolute decrease of phosphatidylaethanolamin was observed.

Die Plasmaphospholipide von acht lebergesunden Patienten und von sechs Patienten mit akuter Hepatitis wurden dünnschichtchromatographisch getrennt und quantitativ bestimmt. 2. Die Gesamt-Plasmaphospholipide waren bei den Hepatitisfällen signifikant erhöht. 3. Die einzelnen Phospholipidfraktionen zeigten bei den Patienten mit Hepatitis charakteristische Abweichungen von der Norm: Lecithin war gegenüber der Norm deutlich erhöht, Sphingomyelin nur relativ erniedrigt, Phosphatidyläthanolamin jedoch auch absolut vermindert.

The turnover rate of free fatty acids in plasma and the esterification rate of free fatty acids to plasma triglycerides was estimated by constant infusion of palmitic acid-9,10-H3 in 16 healthy individuals who received a simultaneous infusion of norepinephrine (0.25µg/min/kg) over a period of two hours. 2. The infusion of norepinephrine in this dosage was followed by a mean percentile increase of plasma free fatty acid concentration of 100.6% ± 49.35 and a mean increment of blood sugar level of 11.38 mg-% ± 4.18. Under the influence of norepinephrine the mean turnover rate of free fatty acids in plasma was significantly higher than in a control group of 11 healthy individuals without the administration of norepinephrine. The mean fractional turnover rate was significantly lower in the individuals receiving norepinephrine than in the control group. 3. During the infusion of epinephrine (0.25, 0.125, 0.063µg/min/kg resp.) blood sugar rose to much higher levels than during the administration of norepinephrine. During the first 30 min of the infusion there was a marked increase in the concentration of free fatty acids in plasma; after this period, however, a decrease in free fatty acid levels was observed. Since there was no steady state in free fatty acid turnover, the turnover rates of free fatty acids in plasma and the influx rates of plasma triglycerides could not be exactly calculated with the given formulas. 4. Under the influence of norepinephrine a highly significant relationship was found between the turnover rate of free fatty acids and the concentration of free fatty acids in arterial plasma. 5. As compared with the control group, the mean esterification rate of free fatty acids to plasma triglycerides was increased during the administration of norepinephrine; the difference, however, was not statistically significant. 6. In contrast to the control group, no relationship between the turnover rate of free fatty acids and influx rate of plasma triglycerides could be shown during the infusion of norepinephrine. Generally, only individuals with high plasma triglycerides show an increase of plasma triglyceride synthesis corresponding to their high free fatty acid turnover rate. An individual response in the magnitude of plasma triglyceride synthesis to endogenous norepinephrine is suggested. 7. In healthy individuals a highly significant positive correlation between the logarithm of the concentration of plasma triglycerides and synthesis of plasma triglycerides was shown during the infusion of norepinephrine.

Bei 16 stoffwechselgesunden Personen wurden die Umsatzrate der freien Fettsäuren im Plasma und die Einbaurate von freien Fettsäuren in Plasmatriglyceride unter gleichzeitiger Infusion von Noradrenalin (0,25γ/min/kg) bestimmt. 2. Die Infusion von Noradrenalin in der angegebenen Dosierung bewirkte einen Anstieg der Konzentration der freien Fettsäuren im Mittel um 100,6% ± 49,35 des Ausgangswertes sowie eine Erhöhung des Blutzuckerspiegels im Mittel um 11,38 mg-% ± 4,18. Die Umsatzrate der freien Fettsäuren war unter der Wirkung von Noradrenalin im Mittel signifikant höher als bei einer Kontrollgruppe von elf Normalpersonen ohne Noradrenalin. Die „Fractional turnover rate“ war unter Noradrenalin gegenüber der Kontrollgruppe signifikant erniedrigt. 3. Die Infusion von Adrenalin (0,25, 0,125 bzw. 0,063γ/min/kg) bewirkte in jeder der drei Dosierungen einen wesentlich stärkeren Anstieg der Blutzuckerkonzentration als Noradrenalin; die Konzentration der freien Fettsäuren im Plasma nahm nach Beginn der Adrenalininfusion deutlich zu, begann aber bereits während der ersten Stunde der Infusion wieder deutlich abzusinken. Da somit hinsichtlich des Fettsäureumsatzes kein „steady state“ bestand, konnten der Fettsäureumsatz und die Einbaurate von freien Fettsäuren in Plasmatriglyceride mit den angegebenen Formeln nicht exakt berechnet werden. 4. Unter der Wirkung von Noradrenalin ließ sich eine positive Korrelation zwischen Umsatzrate der freien Fettsäuren und Konzentration der freien Fettsäuren im arteriellen Plasma nachweisen. 5. Die Einbaurate von freien Fettsäuren in Plasmatriglyceride war unter der Wirkung von Noradrenalin gegenüber der Kontrollgruppe im Mittel zwar erhöht, der Unterschied ließ sich aber statistisch nicht sichern. 6. Im Gegensatz zur Kontrollgruppe ließ sich während der Infusion von Noradrenalin keine Beziehung zwischen Umsatzrate der freien Fettsäuren und Einbaurate von freien Fettsäuren in Plasmatriglyceride nachweisen. Im allgemeinen reagieren nur Personen mit einem höheren Plasmatriglyceridspiegel auf Noradrenalin mit einer der hohen Fettsäureumsatzrate entsprechenden Steigerung der Plasmatriglyceridsynthese. Es besteht demnach wahrscheinlich auch gegenüber endogenem Noradrenalin eine individuelle Empfindlichkeit. 7. Bei stoffwechselgesunden Personen ließ sich eine positive Korrelation zwischen Logarithmus des Plasmatriglyceridspiegels im Nüchternzustand und Plasmatriglyceridbildung unter Noradrenalin nachweisen. Es konnte weiters gezeigt werden, daß bei denselben Personen unter Noradrenalin ein umso höherer Anteil der umgesetzten freien Fettsäuren in Plasmatriglyceride eingebaut wird, je höher die Plasmatriglyceridkonzentration vor der Verabreichung von Noradrenalin war.

    Description / Table of Contents: Summary 1. The splitting ofl-Leucyl-β-naphthylamide-HCl by cytolyzed white blood cells is measured. The enzyme content of normal polymorphonuclear cells, cells of chronic myeloic leukemia and macrophages from the peritoneal cavity of guinea pigs is 3 times higher than that of cells from chronic lymphatic leukemia. 2. In the assay system employed the reaction runs lineary for 30 min. The pH optimum of the enzyme is 7.1, the substrate optimum 5×10−4 M, the Michaelis constant 5×10−5 M. There is no activation by MnCl2 (5×10−5 M, pH 7.1), the reaction is even inhibited in the alkaline range and at higher concentration. 3. The enzyme differs from leucine aminopeptidase.
    Notes: Zusammenfassung 1. Die Spaltung vonl-Leucyl-β-naphthylamid-HCl durch Cytolysate von weißen Blutzellen wird biochemisch gemessen. Normale Granulocyten, Zellen von chronischen Myelosen und Meerschweinchen-Makrophagen weisen einen etwa dreifach höheren Enzymgehalt als Lymphocyten von chronischen Lymphadenosen auf. 2. Das pH-Optimum beträgt 7,1, Mangan-Ionen (5×10−5 Mol/l, pH 7,1) bewirken keine Aktivierung, im alkalischen Bereich und in höherer Konzentration wird eine Hemmung beobachtet. Unter den gegebenen Versuchsbedingungen folgt die LNA-Spaltung einer Reaktion nullter Ordnung. Das Substrat-Optimum liegt bei 5×10−4 Mol/l. Die Michaelis-Konstante beträgt 5×10−5 Mol/l. 3. Das untersuchte Enzym weist dieselben Eigenschaften auf, wie sie für LNA spaltende Enzyme anderer Gewebe beschrieben wurden, unterscheidet sich jedoch von der Leucinaminopeptidase.
