    Notes: Abstract Quantitative and reproducible information concerning the development of the extraradical mycelium of arbuscular mycorrhizal fungi (AMF) is lacking due to the difficulties in extracting, identifying and estimating hyphal lengths. In this study, using a rhizobox growth system, the lengths of hyphae of AMF estimated using an image analysis system were not significantly different from data obtained by a trained observer using a modified grid-line intersect method. The assessment of lengths of hyphae on membrane filters or slides was, however, much quicker using image analysis, and allowed the complete sample to be quantified, unlike the grid-line method where a limited number of fields of view are assessed. The image analysis procedure is objective, observer-independent and less laborious than the manual method of assessment. Of the four different methods of sample preparation compared, membrane filter methods were found to be the most appropriate for quantitative sampling from three non-soil substrates. Glomus monosporum (UKC M3) produced twice as much extraradical mycelium and hyphal length per centimetre of colonised root than G. geosporum (BEG 11) on both leek and linseed in a durite sand at final harvest (63 days). Both AMF also produced more hyphal length per centimetre of colonised root on linseed than on leek. The spatial distribution of both AMF, however, was similar in durite sand and no correlation with levels of NaHCO3-extractable phosphorus was noted. In a third experiment, with G. manihotis (UKC INDO-1) colonising a tropical forage legume, Pueraria phaseoloides, in two other growth substrates, a different pattern of development of the extra-radical mycelium was observed. Because of a higher content of particulate matter, which collected on the membrane filters, the extraction technique had to be modified to give optimal performance of the image analysis system.

    Notes: Abstract  Two glasshouse experiments were done to assess the development and metabolic activity of mycorrhizas formed by isolates of arbuscular mycorrhizal fungi (AMF) from three different genera, Acaulospora, Gigaspora and Glomus on either Pueraria phaseoloides L. or Desmodium ovalifolium L. plants. The second of the two experiments included three levels of a localised phosphate source in the pots. Alkaline phosphatase (ALP), stained histochemically in the intra-radical mycelium (IRM) of AMF over sequential harvests, did not provide a direct marker for the efficiency of AMF in mobilising phosphorus (P) for plant growth and development. The ability of the extra-radical mycelium (ERM) to scavenge a localised phosphate source, determined by its extraction from buried 35-μm mesh pouches, was dependent on the species of AMF tested. This work indicates that AMF from different genera have unique patterns of mycelial development when forming mycorrhizas with tropical hosts in the presence of a localised phosphate source. AMF also appear to have different mechanisms for the control of P transfer, within the mycelium, to the host. The significance of the architecture of the ERM is discussed as well as the localisation of ALP in the IRM in determining the efficiency of AMF in terms of P accumulation in planta and subsequent growth of plants.

    Notes: Summary We studied the effects of field application rates of three fungicides on spore germination and root infection in three species of Glomus. Bavistin (carbendazim) prevented germination of spores of G. monosporum and G. mosseae whereas Calixin (tridemorph) had no effect. Tilt Turbo (propiconazole) inhibited spore germination less than Bavistin. Spores of G. geosporum germinated in the presence of all three fungicides. Foliar applications of Bavistin alone and in a triple-spray programme significantly reduced infection of all three Glomus species in wheat roots grown in conditions of low P availability (2 mg P 1−1). Two fungicides with triazole derivatives, Sportak and Tilt Turbo, also decreased infection and the former significantly reduced spore production of all three Glomus species. Calixin-treated plants, however, showed increased infection levels, resulting in greater yields and greater foliar-P concentrations than control plants. Milgo (ethirimol) was not detrimental to the vesicular-arbuscular mycorrhizal (VAM) symbiosis when applied as a foliar spray.

    Notes: Summary We studied the effects of field application rates of four herbicides (Avenge, Ceridor, Dicurane, and Harrier) on spore germination and infection of wheat roots by three species of Glomus grown under conditions of low P availability. Low concentrations of Ceridor (bifenox, mecoprop) and Harrier (mecoprop, ioxynil, clopyralid) inhibited spore germination while higher concentrations were stimulatory. Avenge (difenzoquat methyl sulphate) prevented spore germination completely, while Dicurane (chlortoluron) had no effect. The herbicide applications had no significant effect on the infection rates of any of the three fungi except Harrier on G. geosporum. The herbicide treatments did, however, affect plant growth and ear yields. Ceridor and Harrier increased ear yields, while Dicurane showed marked phytotoxic effects.

    Notes: Two field trials were conducted in the Philippines in successive years to compare the effectiveness of different pre- and post-harvest treatments on the development of anthracnose on mango fruits caused by Colletoirichum gloeosporioides. In one experiment, the pre-harvest application of benomyl (250 mg/l a.i.) significantly reduced the number of conidia sampled in tree canopies compared with control trees (no fungicide) and was associated with a significant reduction in the post-harvest development of anthracnose on fruits. A hot-benomyl dip (850 mg/l a.i. at 52 55 C for 10 min) completely eradicated anthracnose on fruit treated on the day of harvest. In a second experiment, pre-harvest applications of prochloraz (500 mg/l a.i.) either within a pre-planned spray schedule or applied strategically (when 18 h or more continual leaf wetness was recorded by a sensor placed within a tree canopy) gave the best control of anthracnose on fruits. A hot-benomyl dip was again the most effective post-harvest treatment for fruit treated on the day of harvest and on the day after. There was no significant difference, however, between hot-benomyl dips or prochloraz dips (500 mg/l a.i. for 10 min) at ambient temperatures when fruit were treated on the third day after harvest. The implications of these results for the production and treatment of Carabao mangoes in the Philippines are discussed.

    Notes: A comparison of rates of germination and appressorium formation by an isolate of Colletotrichum gloeosporioides on mango leaves, fruit surfaces and cellophane membranes showed that behaviour was broadly similar on all three substrates. Frequency of appressorium formation was slightly higher on cellophane membranes, and both hyaline and melanized appressoria were formed. Only melanized appressoria were formed on mango surfaces. In vitro experiments on membranes showed comparative differences in physiological behaviour with temperature for two Philippine isolates of C. gloeosporioides. The most stimulatory temperature for production of appressoria differed in isolates I-2 and I-4 (25 and 20°C, respectively). At 30°C more appressoria became melanized than at lower temperatures, but the frequency of formation of penetration pegs was highest at 25°C. Conidia of C. gloeosporioides germinated on cellophane membranes at relative humidities as low as 95%, but the percentage of conidia germinating and forming appressoria increased as the RH approached 100%. Approximately 18% of conidia of C. gloeosporioides I-2 held at 62 and 86% RH for 4 weeks retained viability, and some were capable of forming appressoria when placed at 100% RH. These results have implications for epidemiological models for disease control.

    Notes: Infection estimates determined by a predictive model developed by Dodd et al. (1991b) were used to time fungicide sprays to control anthracnose disease of mango in the Philippines. For an amount of disease on fruits after harvest which was acceptable to growers, this approach resulted in the application of five fewer sprays compared with a standard spray programme used by the growers in a field trial conducted in 1991–1992. The model predicted only two high anthracnose-risk periods (〉40% of conidia forming appressoria) throughout the duration of the growing period. Rainfall intensity and its time of occurrence during fruit development was found to greatly influence the amount of anthracnose and stem-end rot disease on fruits after harvest. Three relatively strong precipitations (〉20 mm) within a month before harvest resulted in relatively high anthracnose infection of fruits after harvest. At a second field trial, rainfall periods during fruit development did not exceed 4 mm and resulted in virtually disease-free fruits after harvest, including those not treated with fungicide. Again the use of the predictive model resulted in the elimination of five fungicide treatments compared with the standard programme. The amount of rainfall and the time of its occurrence should be considered when planning a disease management scheme for the control of anthracnose on mango fruit.

    Notes: The epidemiology of the anthracnose pathogen of mango, Colletotrichum gloeosporioides, was studied over two growing seasons in the Philippines. This pathogen spreads within tree canopies as water-borne conidia during rainfall, and causes symptoms on young flush leaves, flowers and fruit. Infection studies with leaves and fruit incubated at different temperatures and humidities led to the derivation of a logistic regression model of the percentage of conidia forming appressoria. This model was compared with a similarly derived model from work in Australia; it appeared that the Philippine isolates of C. gloeosporioides were adapted to the higher mean temperatures of the Philippines. Conidia were observed to germinate and form appressoria at relative humidities (RH) between 95 and 100%, even though free surface moisture was only visible at 100% RH. This model was used retrospectively to estimate infection levels in two field trials which had been established to compare pre- and post-harvest practices in the control of the disease. Using this information to plan applications of a curative fungicide might have resulted in four fewer sprays in the first trial and one less in the second, compared with the standard protective spray programme employed.
