    Notes: Summary There is much evidence that diabetes and hyperglycaemia contribute to the impairment of endothelial function and induce severe changes in the proliferation, the adhesive and synthetic properties of endothelial cells. Induction of apoptosis could represent one mechanism to prevent the new accumulation of those vascular defects and to allow generation of vascular endothelium. In this study, we demonstrate that high concentrations of glucose or proinsulin induce apoptosis in human umbilical endothelial cells by three independent methods (DNA fragmentation, fluorescence activated cell sorting analysis, and morphology). The number of apoptotic cells was increased by glucose (30 mmol/l or proinsulin (100 nmol/l) from less than 10 % to about 30 %. Activation of protein kinase C (PKC) largely prevented the induction of apoptosis, whereas inhibition of PKC further increased the number of apoptotic cells. Similar changes as induced by glucose were also observed after incubation of the cells with the non-metabolisable 3-0-methylglucose. These findings indicate that hyperglycaemic conditions stimulate the induction of apoptosis in endothelial cells by a mechanism which is independent from the formation of diacylglycerol and the activation of PKC. The induction of apoptosis by the non-metabolisable glucose suggests that formation of oxygen derived radicals by autoxidative processes is involved and may lead to an activation of transcription factors such as nuclear transcription factor-ϰB (NF-ϰB) transferring the activation signal into the nucleus and leading to changes in gene expression necessary for induction of apoptosis. As it has been shown that oestrogen enhances the cholinergic muscarinic activity in the central nervous system, we studied sex differences in the responses to parasympathetic nervous stimulation in the rat heart using in vivo and in vitro preparations.2. In in situ perfused, innervated hearts, stimulation of bilateral vagus nerves (15 Hz with 1 μmol/L physostigmine) inhibited sympathetic nerve stimulation (5 Hz) induced noradrenaline release to a greater extent in female than in male rats (54 ± 5 vs 72 ± 5% of control). Similarly, vagus nerve stimulation at 1–20 Hz reduced heart rate (HR) more in females than males, and this sex difference became more marked in the presence of physostigmine. The chronotropic effect of vagal stimulation was attenuated after ovariectomy but potentiated after castration when compared with sham-operated controls. In contrast, the muscarinic agonist methacholine reduced neural NA release and HR equally well in both sexes.3. In anaesthetized rats, reduction in HR and mean arterial pressure by vagus nerve stimulation (1–20 Hz) was more pronounced in females than in males after inhibition of acetylcholinesterase with physostigmine.4. The results indicate that activation of parasympathetic nerve leads to greater presynaptic and postsynaptic effects in female than in male rat hearts, presumably due to a higher level of acetyl-choline release following nerve activation.

    Notes: Cotton fibre mutants that were fuzzless and/or lintless were crossed with each other and a normal genotype (fuzzy, linted) to produce F2 and BC1 generations. F2 segregation ratios from the cross of fuzzless-lintless × fuzzy-linted, for fuzzy-linted, fuzzless-linted and fuzzless-lintless were 45 : 15 : 4. From the cross of fuzzless-lintless × fuzzy-linted, the F2 segregation ratios were 9 : 39 : 16 whereas the BC1F1 segregation ratios from the F1 backcrossed to fuzzless-lintless were 1 : 3 : 4. These data suggest that the presence or absence of lint and fuzz are controlled by the interaction of four gene loci on non-homologous chromosomes. We designate these loci as N1, N2, Li3 and Li4, where N1 N1 confers the presence of fuzzy, N2N2 confers inhibition of fuzzy initiation and development, and duplicate gene pairs, Li3Li3 and Li4Li4, determine the presence of lint. Homozygosity for li3li3 and li4li4 might also inhibit fuzz from development. In other words, they were recessive epistatic to fuzz genes.

    Notes: To extend the measurement range to 100 GPa, a modified manganin gauge was prepared and examined. The modifications were focused on the package materials and technique. The manganin sensing elements were first deposited by magnetron sputtering on Al2O3 substrates, and then covered by a layer of Al2O3 films several μm thick by electron beam evaporation. After being placed on slabs of Al2O3 ceramic, the gauges were shocked to 40–80 GPa with a light gas gun. The stress-time profiles showed a good plateau of about 1 μs in duration. The initial calibration resulted in a linear curve with piezoresistance coefficient of 0.0198±0.0002 GPa−1. © 2002 American Institute of Physics.

    Notes: Silica-titania sol-gel planar waveguides were deposited on silica glass disks by spin coating. Fluorescence lifetimes as high as 375 μs were measured in neodymium-doped sol-gel planar waveguides and 1.78 ms for an erbium-doped film. Quenching was found to occur for the former at concentrations above 1 at. % of neodymium. © 1996 American Institute of Physics.
