    Notes: Abstract A model is described of acute inflammation in the pleural cavity of rats using calcium pyrophosphate as the irritant. This model would seem to simulate the pseudogout syndrome. It has been shown to be acute in onset, dominated by polymorphonuclear cells, complement independent. The advantage of the model is that volume of exudate, numbers and types of cells may be quantitated. Prostaglandins and cyclic AMP have been measured in the migrating cells. The significance of these findings has been discussed.

    Notes: Abstract The development of hind paw oedema, in response to dextran injected into the pleural cavity of rats, has been shown to differ with the age of rat studied. It was found that newborn rats were less responsive than adults, with respect to oedema formation induced by dextran. Attempts to suppress the dextran-induced foot oedema using mepyramine and cyproheptadine, were successful only in rats from the adult age group. A semiquantitative analysis was made of the inflammatory cellular exudate produced by the intrapleural injection of dextran into rats of different ages. In contrast to the reaction of adult rats, it was found that the accumulation of polymorphs in the pleura of newborn animals was delayed. The polymorph was the dominant cell type found in exudates induced either by intrapleural saline or dextran, in rats from the 6-hour age group. This contrasted strikingly with comparable exudates induced in adult rats, whose major cell type was the mononuclear cell (after saline), and the polymorph, followed by the mononuclear cell (after dextran). Fewer cells were observed in the reaction of 6-hour-old rats, compared with 7-day-old animals following intrapleural injection of the same volume of a 6-percent solution of dextran.

    Notes: Abstract A model of reverse passive Arthus (RPA) reaction in the pleural cavity of rats is described. The time course of development of exudate and migration of cells has been examined. It has been found to be complement dependent and dominated by polymorphonuclear cells. The reaction reaches a peak around 6 hours after challenge. Cyclic AMP levels have been measured both intracellularly and extracellularly and have been found to persist at high levels after the waning of the reaction.

    Notes: Abstract Both the vascular and cellular aspects of inflammation were studied in rats aged from 6 hours to 2 months. The onset of a vascular permeability reaction was found to vary according to the age of rat and type of permeability agent injected. Induction of acute inflammations (turpentine pleurisy, 48/80 and dextran-induced foot oedema) in newborn rats revealed a marked reduction of exudate/oedema formation compared with adult rats. These observations were related to a lack of histamine and 5-HT-mediated increased vascular permeability during the inflammatory reactions of newborn rats. Qualitative differences were observed between the acute inflammatory pleural cell exudates of newborn and adult rats after intra-pleural injection of dextran. In contrast to adults, polymorphs were observed to dominate the newborn reaction, and the peak accumulation of these cells was delayed. The mononuclear cells of 4-day lesions induced by sub-cutaneous implantation of glass coverslips showed a higher rate of mitosis in newborn animals, compared with adults. Ultrastructural studies of the mononuclear cells of peritoneal exudates induced by carrageenan indicated that those of newborn rats contained fewer lysosomes than adults. The Golgi apparatus of newborn mononuclear cells was observed to be poorly-developed compared with adult mononuclear cells.

    Notes: Abstract The mast cell population of rat diaphragm was estimated between birth and adulthood and found to rise with an increase in the age of rat studied. Degranulation of these cells was observed in rats from all age groups, following treatment with compound 48/80 and dextran. The association of mast cells with the blood vessel wall in adult rat diaphragm was not observed in the comparable tissues of newborn rats. These findings are discussed in relationship to the poor vascular permeability reactions exhibited by newborn and young rats.

    Notes: Abstract Intrapleural exudates have been produced in rats and guinea-pigs using calcium pyrophosphate, cell mediated immunity, reversed passive Arthus reaction and carrageenan. The intracellular concentration of various pharmacological mediators has been measured at different times during the evolution of these inflammatory responses. These have been correlated with volume of exudate and cellular migration. The significance of these findings has been discussed and the possible application to the search for new anti-inflammatory agents and their use in diagnostic procedures.

    Notes: Summary We separated osteoclasts from bone and observed the effect of several known and potential mediators of the control of bone resorption on their cytoplasmic motility. We already found that calcitonin (CT), a hormone that inhibits bone resorption, regularly causes complete inhibition of cytoplasmic motility, specific for osteoclasts, through a trypsin-sensitive membrane receptor [1]. We report here that prostaglandin I2 (PGI2) and dibutyryl cyclic AMP induce an identical change in osteoclastic behavior. We found that theophylline, which inhibits intracellular cyclic AMP degradation, and which itself had no effect on osteoclastic motility, potentiated the cytoplasmic inhibition casued by CT, PGI2, and cyclic AMP. This suggests that PGI2 and CT cause cytoplasmic quiescence by increasing the intracellular level of cyclic AMP, a view compatible with the known ability of CT to increase cyclic AMP in bone [2]. Parathyroid hormone (PTH), PGE2, and 1,25 dihydroxycholecalciferol (1,25 (OH)2D3), hormones known to stimulate osteoclasts, did not stimulate the activity of either active or quiescent isolated osteoclasts. The undoubted ability of these hormones to stimulate osteoclastic activityin vivo may therefore be mediated through a primary hormonal interaction with another cell type.
