    Notes: Summary Two groups of 60 day old male Wistar Rats were subjected to right-sided rubro-spinal tractotomy at the fourth cervical and thirteenth thoracic vertebral levels respectively. Four animals in each group were sacrificed at each of the time intervals 1, 3, 4, 7, 14, 21 and 55 days. Counts of chromatolytic neurons from both groups of animals at time intervals ranging from 1–21 days established a time course for the chromatolytic response. Morphological observations showed a more severe central chromatolysis in the cervical group commencing at Day 3, and a much less severe central chromatolysis in the thoracic group starting at Day 4. By Day 21 the majority of the neurons of the cervical group were atrophic and by 55 days all were atrophic. The reaction following thoracic lesions was less intense and the maximum number of reacting neurons was observed at Day 7. Following this stage the neurons gradually returned to a normal morphological state which was complete by Day 55. Measurements of neuronal diameters from semi-thin Epon sections in the cervical group yielded statistically significant alterations in the perikaryal diameters of experimental neurons, demonstrating swelling at Days 1, 3 and 4 followed by shrinkage at Days 7, 14 and 21. The findings of this study suggest that the severity of the chromatolytic reaction in intrinsic neurons is intimately related to the distance of the lesion from the neuronal soma, while the time of onset of chromatolysis varies with lesion distance but is not directly proportional to it.

    Notes: The effect of small temperature differentials (16 vs. 20°C) on the pathogenicity of deoxynivalenol producing single isolates of Fusarium culmorum and F. graminearum and on the fusarium head blight (FHB) response of eight wheat cultivars was examined. Fusarium culmorum inoculation caused greater visual disease symptoms at 20°C than at 16°C, both overall and on an individual cultivar basis (overall AUDPC = 13·5 and 9·6, respectively) (P 〈 0·05). In contrast, F. graminearum inoculation caused greater overall visual disease symptoms at 16°C than at 20°C, both overall and at the individual cultivar level (overall AUDPC = 12·8 and 10·9, respectively) (P 〈 0·05). Results showed both F. culmorum and F. graminearum inoculations caused a greater loss in yield at 20°C (54·3 and 46·9% relative 1000-grain weight, respectively) compared with 16°C (73·3 and 66·9% relative 1000-grain weight, respectively) (P 〈 0·05). Fusarium culmorum-inoculated heads contained similar amounts of fungal DNA at both 16 and 20°C (1·9 and 1·7 ng mg−1 of plant material, respectively) (not significant), while for F. graminearum inoculation, plants contained higher amounts of fungal DNA at 20°C (2·0 and 1·0 ng mg−1 of plant material, respectively) (P 〈 0·05). Overall, there was a significant negative correlation between AUDPC and percentage relative 1000-grain weight at both 16 and 20°C (r =−0·693 and −0·794, respectively, P 〈 0·01).

    Notes: The pathogenic and genetic diversity of Colletotrichum lindemuthianum isolates collected from a total of 10 Central and South American, European and African countries were characterized using common bean differential cultivar pathogenicity tests and amplified fragment length polymorphism (AFLP) analysis. On the basis of pathogenicity tests, 74 isolates were attributed to 30 different pathogenic races using the CIAT-defined binary race-classification system. Twenty-one races were restricted geographically, being exclusive to different countries. Race 9 was the most widespread, being detected in four different countries. Cluster analysis of consensus AFLP data generated using three selective AFLP primer combinations grouped 86 isolates (including the 74 subjected to pathogenicity tests) into three clusters (with one isolate as an outlier). This analysis showed that the majority of South and Central American isolates were divided among two clusters, and that the limited number of European and African isolates used in this study were genetically most similar to Central American isolates. Overall, the results of this research showed some associations between the genetic diversity of the isolates and their country of origin, but not between genetic diversity and race classification of isolates.

    Notes: New evidence is presented to support the contention that the amoeba/cyst colonies isolated from surface-sterilized Spongospora subterranea f.sp. subterranea-infected potato tubers and spore balls have a saprophytic phase but are contaminants and not S. subterranea. Amoebae isolated from infected tissues and spore balls formed colonies associated with bacteria on 1% water agar at 18°C and encysted after 5–7 days. These cysts were morphologically distinct from the resting spores of S. subterranea and were formed singly or in a layer, unlike the spore ball (cystosorus) of S. subterranea. Amoebae, cysts and mixtures of amoebae and cysts in primary, secondary and tertiary subcultures failed to infect tomato roots. PCR amplification of DNA from amoebae, cysts and spore balls using the S. subterranea-specific primer pair SsF/R generated a 434-bp product from S. subterranea spore balls only and not from amoebae or cysts. When an amoeba/cyst-specific primer pair AmF/R was designed and used for PCR amplification, a single 411-bp product was generated from DNA of amoebae and cysts, but not from DNA of S. subterranea spore balls. These results are discussed in relation to earlier reports claiming the successful isolation of S. subterranea and other plasmodiophorids in vitro.

    Notes: Summary A modified version of the miniature toys test, originally designed by Sheridan (1975), has been standardized on a sample of 425 children in the age range 18 months to 4½: years. The distinctive features are the assessment of symbolic play and the understanding of prepositions when used in verbal instructions. The acquisition of expressive verbal labels for miniature household objects and transport, and comprehension of questions relating to them have been explored. Responses in some of these areas are not always clear-cut or are ambiguous. The test will be more suited to the needs of the developmental paediatrician for assessment of children with developmental delay, than for doctors of primary care and health visitors in screening apparently healthy children.

    Notes: Abstract 7-Hydroxycoumarin (7-HC) was chemically conjugated by diazo coupling to carrier proteins such as bovine serum albumin (BSA), thyroglobulin and ovalbumin. These conjugates were characterised by sodium dodecyl sulphate/polyacrylamide gel electrophoresis (SDS-PAGE) and high-performance liquid chromatography (HPLC). Rabbits were immunised using the 7-HC-BSA conjugate. The highest antibody titre achieved was 1:10000, as determined by competitive enzyme-linked immunosorbent assay (ELISA). The resulting antibodies were purified by ammonium sulphate precipitation, followed by protein A affinity chromatography. Their purity was assessed by SDS-PAGE and HPLC. These antibodies have been used in the development of a competitive ELISA, an amperometric biosensor and an electrochemical immunoassay. Both the ELISA and amperometric biosensor have been successfully applied to the analysis of 7-HC and its glucuronide conjugate in human urine samples. Each of these antibody-based methods provides a novel approach to the analysis of the main metabolites of coumarin.
