Retinoids and Carotenoides, III. - [8,10,12,14,19,19,19,20,20,20-D10]RetinalsThe perdeuterated α-pyrone 11 can be prepared by reacting hexadeuterioacetone (1) and the ylide 5 and treating the resulting [D6]dimethylacrylic ester 6 with deuteriopolyphosphoric acid (8). The same reaction of the nondeuterated ester 12 is accompanied by a high rate of deuterium incorporation yielding 11. The latter is transformed into the aldehyde 13. Subsequently 13 is reacted with 14 to give the pyrone 15 in a Witting reaction. In the course of the ring opening of 15 with NaBH4/CH3OH/KOH five deuterium atoms are lost, and the (13Z)-vitamin A acid 16 is formed. Reacting 15 with NaBH4/CH3OD/NaOD produces the [D10]acid 17 which cannot be transformed selectively into the corresponding aldehyde 18. The resulting isomers 18, 19, and 20 can be separated by HPLC. 18 and 20 react with n-butylamine to give the Schiff bases 22 and 23. The Z/E-isomerization 22 → 23 is not catalyzed by bases. It takes place, however, thermally at 40°C. By this way, one D atom is exchanged for an H atom.

Das perdeuterierte α-Pyron 11 läßt sich aus Hexadeuterioaceton (1) durch Umsetzung mit dem Ylid 5 und anschließende Behandlung des gebildeten [D6]Dimethylacrylesters 6 mit Deuteriopolyphosphorsäure (8) gewinnen. Bei der gleichen Reaktion von undeuteriertem Ester 12 erfolgt in hohem Maße Einbau von Deuterium unter Bildung von 11. Letzteres wird in den Aldehyd 13 übergeführt und anschließend durch Wittig-Reaktion mit 14 zum Pyron 15 umgesetzt. Die Ringöffnung von 15 mit NaBH4/CH3OH/KOH ist mit dem Verlust von fünf Deuteriumatomen verbunden. Es bildet sich die (13Z)-Vitamin-A-Säure 16. Mit NaBH4/CH3OD/NaOD erhält man aus 15 die [D10]Säure 17. Selektive Überführung von 17 in den korrespondierenden Aldehyd 18 gelingt nicht. Es bilden sich die Isomeren 18, 19 und 20, die sich durch HPLC trennen lassen. 18 und 20 reagieren mit n-Butylamin zu den Schiff-Basen 22 und 23. Die Z/E-Isomerisierung 22 → 23 wird nicht von Basen katalysiert. Sie findet jedoch bei 40°C thermisch statt. Dabei wird ein D-Atom gegen ein H-Atom ausgetauscht.
