    Notes: Summary 1. Undifferentiated fore limb blastemas were denuded of their epidermis and grafted to the flank musculature; each transplant consisted of four such blastemas. Outgrowth of the mesenchyme and subsequent digit formation were prevented by covering the transplants with whole flank skin with its dermis intact. 2. Notwithstanding the absence of digits, 26 out of the 36 differentiated transplants formed one or two oblong, stout cartilages resembling proximal skeletal elements of the limb. Considering that four blastemas had been grafted in random orientation, this indicates considerable intrinsic morphogenetic capacities of the blastemal mesenchyme in the absence of organizing influences of the epidermis. 3. Thus, although permanent contact with wound epidermis has previously been shown to be necessary for blastemal mesenchyme to form distal limb structures, such contact is not required for the formation of proximal skeletal elements. The implications of this finding for regional organization in the regenerating limb are discussed.

    Notes: Summary A new PI Q0 variant (PI Q0riedenburg) is described; it is caused by a complete deletion of the α1-antitrypsin (α1AT) gene. The deletion gives rise to four new restriction fragment length polymorphisms (RFLPs) detected with a genomic probe of the 5′ region of the gene. Analysis of the RFLPs indicates that the deletion starts immediately upstream of exon Ic. The deletion extends into the 3′ flanking region of the gene but does not include the α1AT-related gene (the PIL gene), which is located 12 kb downstream of the α1AT gene.

    Notes: Summary Overlapping genomic clones of the human α2-macroglobulin (α2M) gene were isolated from a cosmid library and were used to map 80 kb of the chromosomal region of this gene. Fragments carrying the two exons encoding the bait region and the exon encoding the thiolester site were partially sequenced and PCR primers were designed for the amplification of both functional domains. By direct genomic sequencing of these domains in 30 healthy individuals and in 30 patients with chronic lung disease three mutations were detected. The first was a sequence polymorphism occurring near the thiolester site of the gene, changing Val1000(GTC) to Ile1000(ATC), with allele frequencies of 0.30 (GTC) and 0.70 (ATC), respectively. No difference of α2M serum levels was observed for these two alleles. The second mutation occured within the thiolester site of one patient, changing Cys972(TGT) to Tyr972(TAT). Since activation of the internal thiolester formed between Cys972 and Gln975 in each of the subunits of the tetrameric α2M is involved in the covalent cross-linking of the activating proteinase, this mutation is predicted to interfere with α2M function. The α2M serum level was within the normal range in this patient. In one healthy individual we detected an alteration of the bait region sequence, which is usually encoded by two different exons separated by an intron of size 1.6kb. In this individual, PCR amplification of genomic DNA using the bait region primers produced the common fragment of size 1.8 kb and an additional variant fragment of size 0.23kb. This finding, and the genomic sequencing data of this individual, indicate that he carries two different alleles of the α2M gene: one with the regular structure (bait exon I-intron-bait exon II), the other with the two bait exons fused into one. Direct genomic sequencing of the two α2M functional domains is a useful tool for the detection of the genetic, and possibly the functional, heterogeneity of α2M. This, in turn, may provide some insight into the hitherto unknown physiological role(s) of α2M, by studying in vivo effects of naturally ocurring mutations of the gene.

    Notes: Abstract The dc electrical conductivity of rare-earth doped ceria has been measured as a function of temperature (300–600 K) and composition (0.05–15 mol% M2O3) on using the complex impedance technique. Five dopants have been selected, yttrium and the lanthanides Yb, Gd, Nd, and La. For all of them, the variations of the activation energy versus dopant concentration are similar and characterized by the existence of a minimum. This peculiar property can be understood if attractive interactions between immobile dopant ions and mobile oxygen vacancies are taken into account. From an analysis of the experimental results, it is concluded that this interaction extends at least to third or fourth nearest neighbors depending on the size and the electronic configuration of the dopant ion.

    Notes: Summary Bone graft substitute success depends on the formation of healing bone tissue that will bring about the incorporation of the implant, in a sequence of discrete phases. Each of these phases may be influenced by the nature of the implant used. The biological, biochemical, and physicochemical parameters involved may be measured, quantified, and checked. All those involved in the manufacture or the use of these substitutes must thoroughly understand these aspects in order to be able to control them. Tricalcium phosphate, Hydroxyapatite and the biphasic compounds are currently the most widely used materials. TCP, whether used by itself, or as one of the constituents of the biphasic calcium phosphates, will eventually disappear, leaving the bone graft substitute to be gradually osseointegrated. This osseointegration will ensure the biocompatibility of the implant, since resorption occurs without the interposition of a fibrous tissue layer.

    Notes: Abstract Shwachman-Diamond syndrome (SDS) is an autosomal recessive disorder characterised by exocrine pancreas insufficiency, metaphyseal dysostosis and bone marrow dysfunction. Recurrent severe bacterial infections and susceptibility to leukaemia are the major causes of morbidity and mortality occurring preferentially in patients with pancytopenia and features of myelodysplasia. Here we report a patient with SDS leading to recurrent bacterial infections and a deteriorating condition since early infancy. Extensive investigations disclosed severe pancytopenia, myelodysplasia and a clonal cytogenetic abnormality, inv(14)(q11q32), as risk factors of leukaemic transformation. He therefore underwent allogeneic geno-identical bone marrow transplantation which resulted in correction of all haematological and immunological abnormalities within an 18-month follow up period. Conclusion Bone marrow transplantation may be considered early as a valuable treatment option especially in high risk Schwachman-Diamond syndrome patients anticipating malignant transformation, life-threatening severe infections or further organ damage.

    Notes: Above the 1300 K transition and in the defect fluorite phase, there is diffuse X-ray scattering which is similar for both Ca- and Y-stabilized zirconias. This is largely due to ionic displacements from the average structure, principally oxygen ions displaced locally in (100) directions. It has been proposed that some of this scattering is due to fine precipitates of ZrCa4O9 or ZrY4O9. The quantitative analysis of this diffuse scattering reported here does indeed indicate some similarities to this phase; in particular the stabilizing ions form rods in the ({\bar 3}{\bar 3}2) directions similar to those in the precipitate. However, in this compound the stabilizing ion is a second neighbor to an oxygen-ion vacancy, whereas the present results indicate that these tend to be first neighbors. This is the first direct evidence for this association in these materials. This probably occurs to reduce local distortions and to provide local charge balance. It is concluded that the diffuse intensity is from locally ordered regions, not precipitates.

    Notes: The intensities of the weak superlattice reflections in the UX3 compounds, where X = Ru, Rh, Ge, and Sn, have been measured by neutron diffraction. These compounds have the ordered AuCu3 structure. Excellent fits to the observed intensities are obtained by introducing anisotropic second-order and fourth-order (anharmonic) terms to describe the probability distribution function at the X atom site. The ratio of the magnitude of the mean-square thermal vibration parallel to and perpendicular to the unique tetragonal axis of the X atom appears to depend on whether p bonding (UGe3, USn3) or d bonding (URu3, URh3) occurs with the U electrons. In UGe3 data out to Q = 13 Å-1 show the need to include a fourth-order anharmonic term. The form of this anharmonicity suggests an attractive potential between the nearest- neighbor U-X atoms.
