    Notes: Abstract  As a member of the trithorax-group, the Trithorax-like (Trl) gene of Drosophila melanogaster contributes to the expression of homeotic genes and many other genes. Trl encodes different isoforms of the GAGA factor which is thought to act as an ”antirepressor” of transcription by remodelling chromatin structure and thereby rendering control regions accessible for transcriptional activators. A more global role of the GAGA factor in chromatin structure and function is suggested by various phenotypes of Trl mutations, such as modification of position effect variegation. To better define the molecular basis of these pleiotropic effects, we cloned cDNAs encoding the GAGA isoforms of D. melano- gaster and a distantly related species, D. virilis. We also characterized the genomic organization of both the D. melanogaster and D. virilis genes, and analysed the expression patterns of isoform-specific mRNAs. The D. virilis GAGA isoforms show high similarity to their D. melanogaster counterparts, particularly within the BTB/POZ protein-interaction and the zinc finger DNA-binding domains. Interestingly, conservation clearly extends beyond the previously defined limits of these domains. Moreover, the comparison reveals a completely conserved block of amino acid residues located between the BTB/POZ and DNA-binding domains, and a high conservation of the C-terminus specific for one of the GAGA isoforms. Thus, sequences of as yet unknown functions are defined as rewarding targets for further mutational analyses. The high conservation of the GAGA proteins of the two species is in accord with the nearly identical genomic organization and expression patterns of the corresponding genes.

    Notes: Abstract Using ultrastructural immunogold histochemistry on LR-Gold-embedded 6- and 7-day-old mouse embryos we investigated the appearance of the A- and B1-chains of the laminin molecule during mesoderm formation. With the help of antibodies against the A-chain and the E4 fragment of the B1-chain of the laminin molecule we were able to detect the subunits in vivo. Staining for the E4 fragment of the short arm of the laminin molecule from day 6 was negative. In contrast, strong staining for the A-chain of laminin was observed. Our results show, that the A-chain of laminin appears before the B1-chain in the 6-day-old mouse embryo before a basement membrane is seen between the ectodermal and entodermal cell layers. Furthermore, the staining pattern indicates, that the laminin molecule changes its orientation in the basement membrane of the ectoderm during mesoderm formation. On day 7 staining for the A-chain of laminin and for the E4 fragment was seen in a random distribution throughout the entire basement membrane, whereas in areas were the onset of mesoderm formation was taking place, the E4 fragment was restricted to the edge of the disintegrating basement membrane.

    Description / Table of Contents: Summary Rabbits given sublethal injections of 40 γ/kg E. coli lipopolysaccharide (Coli-ET) first exhibit an inhibition and later an augmentation in square dimension and intensity of the inflammatory skin reaction elicited by 0,1 γ Salm. abortus equi lipopolysaccharide (Salm.-ET) i.c. The inhibitory effect of endotoxin is observed to occur from 0 to 12 hours, augmentation from 24–48 hours after Coli-ET i.v. Only the period of enlargement and intensification of the inflammatory skin reaction after intravenous administration of endotoxin coincided with successful elicitation of generalized Sanarelli-Shwartzman phenomenon.
    Notes: Zusammenfassung Nach intravenöser Vorbehandlung von Kaninchen mit 40 γ/kg E. coli Lipopolysaccharid (Coli-ET) findet sich zunächst eine verminderte, später eine gesteigerte Intensität und Flächenausdehnung der mit 0,1 γ Salm. abort. equi Lipopolysaccharid i.c. (Salm.-ET) ausgelösten Hautentzündung. Die auch im Formalin-Hauttest registrierbare Hemmung der Hautentzündung besteht im Zeitintervall von 0–12 Std, die Steigerung im Zeitintervall von 24–48 Std. Das mit 40 γ/kg E. coli Lipopolysaccharid präparierte generalisierte Sanarelli-Shwartzman-Phänomen (SSP), dessen Auslösung mit gleicher Dosis versucht wurde, kommt nur in der Phase der gesteigerten Hautentzündung (Zeitintervalle 16, 28 und 48 Std nach Präparation) zustande. Während der Hemmphase und nach Wiedernormalisierung der Hautentzündung (Zeitintervalle 8 und 96 Std nach Präparation) gelingt die Auslösung des generalisierten SSP nicht. Damit findet das bekannte SSP-positive Zeitintervall nach Präparation im Verhalten der ET-Hautentzündung eine Parallele.

    Notes: Abstract  For the production of microstructured ceramic parts, development efforts are necessary in the field of powder synthesis as well as in the area of shaping. Several processes have been developed. Powder synthesis was performed starting from metal organic precursors and using a two stage thermal process. Shaping was performed by sol-gel methods, by pressing of a ceramic slurry or an organic precursor paste as well as by direct electrophoretic deposition of gels in a microstructured form. An overview is given on the methods and the results.

    Notes: Abstract  The developed process of tape casting with subsequent stamping is a promising way to make three-dimensional ceramic microstructures. Examples of different ceramic materials illustrate, that a good quality of the moulding with high form fit can be attained after optimisation of the slurry and the stamping process. The sintered microstructures are characterised by lateral dimensions in the micron range and high aspect ratios. The unique properties of ceramic materials in combination with this new process can be used to produce microstructured ultrasonic transducers or micro heat exchangers/reactors.

    Notes: Abstract We investigated the genetic control of heterotopic heart allograft rejection using a family of standard inbred, major histocompatibility complex (MHC)-congenic, and intra-MHC recombinant rat strains. Gene products of the various regions within the rat MHC differed markedly in their capacity to induce rejection. Isolated incompatibility at class I antigens encoded by theRT1. A andRT1. C regions failed to induce rejection within the observation period of 100 days, whereas class II antigens encoded by theRT1.B/D region provoked rapid rejection within 10 days. By comparison of the rejection times of isolated and combined incompatibilities a number of functional interactions could be demonstrated between individual MHC regions which either prolonged or shortened allograft survival. In contrast to rapid rejection of MHC-mismatched heart allografts, differences at non-MHC histocompatibility antigens were associated with graft survival beyond 100 days, although chronic rejection of variable severity was detected histologically. Disparity at non-MHC plus class I antigens, however, provoked acute heart allograft rejection.
