    Notizen: Abstract: Through the use of a screening strategy designed to isolate novel cDNAs encoding proteins concerned with pituitary secretion in Xenopus laevis, we discovered clone X7365, which encodes a transmembrane protein with a signal peptide, two follistatin modules, a unique epidermal growth factor domain, and a short cytoplasmic region. RNA expression analyses indicated that the X7365 transcript is enriched in neuroendocrine tissues. Immunohistochemical studies demonstrated that, in addition to being expressed in the optic tectum and in astrocytes in the optic nerve, the X7365 protein is concentrated in a discrete population of hypothalamic magnocellular neurons. Axons projecting to the median eminence and the neurointermediate lobe of the pituitary were also immunopositive, suggesting that X7365 functions in the regulation of magnocellular neurosecretion.

    Notizen: Abstract: Chromogranins and secretogranins are acidic secretory proteins of unknown function that represent major constituents of neuroendocrine secretory granules. Using a differential screening strategy designed to identify genes involved in peptide hormone biosynthesis and secretion, we have isolated cDNA clones encoding the first nonmammalian homologues of secretogranin II (SgII) and secretogranin III (SgIII) from a Xenopus intermediate pituitary cDNA library. A comparative analysis of the Xenopus and mammalian proteins revealed a striking regional conservation with an overall sequence identity of 48% for SgII and 61% for SgIII. One of the highly conserved and thus potentially functional domains in SgII corresponds to the bioactive peptide secretoneurin. However, in SgII and especially in SgIII, a substantial portion of the potential dibasic cleavage sites is not conserved, arguing against the idea that these granins serve solely as peptide precursors. Moreover, SgIII contains a conserved and repeated motif (DSTK) that is reminiscent of a repeat present in the trans-Golgi network integral membrane proteins TGN38 and TGN41, a finding more consistent with an intracellular function for this protein. When Xenopus intermediate pituitary cells were stimulated in vivo, the mRNA levels of SgII and SgIII increased dramatically (15- and 35-fold, respectively) and in parallel with that of the prohormone proopiomelanocortin (30-fold increase). Our results indicate that the process of peptide hormone production and release in a neuroendocrine cell involves multiple members of the granin family.

    Notizen: Abstract: To investigate if the prevention of disulfide bond formation affects the intracellular transport, sorting, and processing of a distinct set of neuroendocrine proteins in the regulated secretory pathway, we have treated Xenopus intermediate pituitaries with the thiol-reducing agent dithiothreitol. Pulse-chase incubations in combination with immunoprecipitation analysis were used to monitor the fates of the prohormone proopiomelanocortin (POMC), prohormone convertase PC2 and its helper protein 7B2, as well as secretogranin III. Manipulation of the disulfide bonds in POMC and proPC2 blocked their transport to the trans-Golgi network and strongly inhibited their processing. Reduction of the single disulfide bond in 7B2 did not disturb its transport and cleavage, but caused its missorting to the constitutive secretory pathway. Moreover, the liaison between proPC2 and 7B2 was prevented. Dithiothreitol did not affect transport, sorting, and cleavage of secretogranin III, which lacks disulfide bonds. When the reducing agent was washed away, POMC processing, proPC2 maturation, and the association between proPC2 and 7B2 were reestablished. Collectively, our findings indicate that manipulation of disulfide bonds differentially affects the fates of neuroendocrine proteins during their transit through the secretory pathway.

    Notizen: Recently, the efficient tuning of the carrier capture efficiency of quantum well by introducing a barrier asymmetry was demonstrated [J. M. Gérard, Appl. Phys. Lett. 63, 240 (1993)]. The electron launching effect was proposed to improve the gain of multiquantum well detectors. In this article, we give experimental results on the dark current, noise, optical gain, responsivity, and detectivity of barrier asymmetric quantum well detectors. We confirm that the electron launching reduces the capture efficiency and thus improves the gain. In addition to this effect, the modification of the barrier potential profile induces a dark current reduction and a blue shift of the intersubband transition. All these effects are then taken into account for analyzing detector performances. As a result of the comparison between symmetric and asymmetric detectors, we do not see any net advantage of using asymmetric structures for improving detector performances. © 1995 American Institute of Physics.

    Notizen: The intersubband photoresponse in GaAs/AlGaAs quantum-well detectors has been measured with a free-electron laser delivering picosecond pulses. The saturation of the detector photocurrent with increasing power was observed. A model is presented that takes the absorption saturation and transport properties into account and allows the electron lifetime determination from the photocurrent saturation. The electron lifetime is found to increase with the electric field applied in the detector from 1 ps at low field up to 10 ps or more at high field (15 kV/cm). Effects of doping and well asymmetry are discussed. © 1995 American Institute of Physics.

    Notizen: A GaAs/AlAs planar cavity containing a collection of InAs quantum boxes in its core region has been grown in a single step by molecular beam epitaxy, and processed by electron-beam lithography and reactive ion etching into pillar microresonators. The optical study by photoluminescence of these localized light emitters allows a systematic and precise determination of the energies of the first confined photon modes of such microstructures, in good agreement with theoretical estimates. More generally, such probes facilitate the experimental study of the modes of complex photonic microstructures and of the spontaneous emission alteration they entail on a quasimonochromatic light emitter. © 1996 American Institute of Physics.

    Notizen: We report on the optical study of ultrathin (1–3 monolayers) InAs quantum wells in GaAs. The samples have been grown either by standard molecular beam epitaxy (MBE) or low-temperature (350 °C) modulated-fluxes MBE. The latter technique enlarges the range of pseudomorphic deposition of strained InAs layers on GaAs. Both types of samples display sharp (down to 6 meV spectral width) and intense low-temperature excitonic photoluminescence, and a high in-plane homogeneity. Higher energy optical transitions have been observed in both absorption and photoluminescence excitation experiments, performed on multiple quantum well structures.

    Notizen: We discuss optical data obtained on (InAs/GaAs)-InGaAlAs multiquantum well structures grown by molecular beam epitaxy. The combined use of photoluminescence and photoluminescence excitation to study such structures is an efficient test of the quality of the highly strained InAs/GaAs ordered alloy, which is used as the well material. The electron effective mass and the lifting of the valence-band degeneracy in InAs/GaAs short-period superlattices are obtained experimentally for the first time.

Such samples exhibit clearly improved structural and optical properties.
