    Notes: Summary About 50% of the circulating platelets can be taken from healthy donors by means of the cell separation technique. In order to investigate the effect of cell separation on thrombopoesis bone marrow samples were taken from 12 voluntary donors up to 5 times. The megacaryocytes were analyzed before and after cell separation by morphometric measures and determinations of platelet count and platelet function (adhesion, aggregation, spreading, retraction) were carried out. Removing of large amounts of platelets form the circulation by stimulation of thrombopoesis resulted in an increase of the peripherical platelet count already 48 h after cell separation. Of the platelet functions tested only aggregation induced by collagen was found to be increased after cell separation as compared to that before the procedure.

    Description / Table of Contents: Zusammenfassung Die Plasmaseparation wurde mit Hilfe von Zellulose-Diacetat-Hohlfasermodulen durchgeführt. Die Porengröße der Membranen betrug 0.2 µm und hatte eine obere Ausschlußgrenze von ungefähr 3 Millionen Dalton. Fünf Liter Filtrat konnten durchschnittlich in 80 min separiert werden. Der Proteingehalt des Filtrates lag bei etwa 60% des Proteingehaltes im Blut. Die Realtionen der Immunglobuline unterschieden sich im Plasma und im Filtrat nur gering. Durch eine Plasmaseparation gelang es, die Konzentration an Immunglobulinen auf etwa 40% der initialen Konzentration zu senken.
    Notes: Summary Plasma separation was carried out using a new cellulose diacetate hollow-fibre module. The pore size of the membranes used was 0.2 µm and the cut-off at approximately 3 million dalton. Five litres of filtrate was separated in 80 min. The protein content of the filtrate was 60% of that in the blood plasma. When plasma and filtrate were compared, there was little difference in the proportions of the immunoglobulins. At the end of treatment, the immunoglobulin content of the plasma had been reduced to 40% of its initial concentration.

    Notes: Summary The carbohydrate composition of a purified plasma membrane fraction derived from the peripheral blood lymphocytes of a patient with a pro-lymphocytic leukaemia has been investigated and compared to the expression of lectin receptors at the membrane surface of the intact cell. The presence of galactose, mannose, fucose, glucose, sialic acid, N-acetylgalactosamine, N-acetylglucosamine and sialic acid substituted O-glycosidic linkedβ-D-galactopyranosyl (1 → 3) N-acetyl-D-galactosamine was confirmed on membrane glycoconjugates by gas chromatography. A lectin induced agglutinin assay using intact leukaemia cells demonstrated the accessibility of different lectin receptors at the native membrane surface, while the peanut agglutinin and the Helix pomatia receptor was only accessible after neuraminidase treatment.

    Notes: Summary The pharmacokinetics of ranitidine was investigated in 11 patients with acute or end stage renal failure during haemofiltration. Each patient received 50 mg ranitidine i.v. The mean distribution and elimination half lives were 0.13 and 2.57 h, respectively. The total body clearance (CL) and volume of distribution (Vz) were 298 ml·min−1 (5.19 ml·min−1·kg−1) and 1.081·kg−1, respectively. About 17.1% of the administered dose was removed by haemofiltration (in approximately 201 filtrate). Five of the patients still had some urine output and they excreted 0.1 to 11.8% of the dose in urine in 24 h. The haemofiltration clearance was 66.9 ml·min−1 at a filtrate flow rate of 86 ml·min−1, corresponding to a mean sieving coefficient of 0.78 (n=6). As plasma concentrations were still in an effective range after haemofiltration, dose supplementation is not recommended.

    Description / Table of Contents: Summary Six healthy test persons and 14 patients with renal insufficiency requiring dialysis were studied in order to determine the elimination kinetics of cefotaxime. The patients were studied while undergoing haemodialysis and during the interval between dialyses. The half-life increased 17-fold from 51.6 min to 14.6 h during the interval between dialyses. Haemodialysis reduced the half-life to 1.69 h, or one-eighth of the original period. The dialysance of cefotaxime is approximately 60 ml/min. The volume of distribution was 15% of the body weight. On the basis of the pharmacokinetic data presented, the dosis for patients with renal insufficiency requiring dialysis should be reduced to approximately 1/5 to 1/10 of the normal dose.
    Notes: Zusammenfassung Bei sechs gesunden Probanden und bei 14 patienten mit dialysepflichtiger Niereninsuffizienz wurde die Eliminationskinetik von Cefotaxim untersucht, bei den Patienten sowohl unter der Hämodialyse wie auch im dialysefreien Intervall. Die Halbwertszeit war im Dialyseintervall von 51,6 Minuten auf 14,6 Stunden um das 17fache verlängert. Durch die Hämodialyse verkürzte sich die Halbwertszeit mit 1,69 Stunden auf ein Achtel, die Dialysance des Cefotaxims liegt bei ca. 60 ml/min. Das Verteilungsvolumen berechnete sich auf 15% des Körpergewichts. Aufgrund der vorgelegten pharmakokinetischen Daten sollte bei dialysepflichtigen niereninsuffizienten Patienten eine Dosisreduktion auf etwa 1/5 bis 1/10 der Normaldosis vorgenommen werden.

The total carbohydrate content of hemolymph was 2.2±0.3 mg/ml. Ninety percent of these sugars are part of macromolecules which could be retained by ultrafiltration.
