    Description / Table of Contents: Zusammenfassung In vitro wird die Freisetzung von ADH aus der Neurohypophyse der Ratte durch Zugabe von Angiotensin (AII) signifikant gesteigert; entsprechend ist am Hund in vivo eine Stimulation der ADH-Abgabe nach AII nachweisbar. Beides läßt auf eine enge Beziehung des Renin-Angiotensin-Aldosteron-Systems mit ADH schließen. Um auch beim Menschen nähere Aussagen hierüber zu gewinnen, wurde ein sensitiver Radioimmunoassay für ADH entwickelt [17, 18]. Die gemessenen basalen ADH-Werte entsprechen den in der Literatur mitgeteilten Daten und betragen 3,4±2,2 pg/ml. Nach AII-Infusion (3–30 ng/min·kg) kommt es zu einem signifikanten Anstieg auf das 2–5fache des Ausgangswertes. Innerhalb von 90 min kehren die Werte wieder auf den Ausgangsbetrag zurück. Zur Kontrolle eines wirksamen AII-Spiegels diente die Erhöhung des Blutdruckes um ca. 20 mm Hg. Der Plasmareninwert nahm innerhalb dieser Zeit ab, Osmolalität und Elektrolytkonzentration des Plasmas änderten sich nicht. Eine Freisetzung von ADH durch die Erhöhung des Blutdruckes per se scheint äußerst unwahrscheinlich zu sein, da bei der zu gleichem Druckanstieg führenden Gabe von Noradrenalin keine Erhöhung des ADH-Spiegels im Plasma gefunden wurde.
    Notes: Summary Release of arginine vasopressin (AVP) from rat neurohypophysis in in vitro studies is significantly augmented by the addition of angiotensin (AII), and in in vivo studies in dogs renin and AII were found to stimulate secretion of AVP. Both these results suggest the existence of a direct relationship between the salt regulating renin-angiotensin-aldosterone system and the water controlling AVP system. To evaluate whether such observations apply also in man a sensitive double antibody radioimmunoassay for AVP was developed [17, 18]. Basal plasma levels of AVP in recumbent humans without salt and fluid restriction at room temperature were 3.4±2.2 pg/ml, and 30 min after the onset of an AII infusion at a concentration of 3–30ng/min·kg, a significant increase of AVP was found. Maximum measurements were 2–5 times basal levels which returned to normal within 90 min. During the AII infusion one also noted a 20 mm Hg rise in blood pressure, accompanied by a significant decrease in plasma renin activity. During the same period serum osmolality and serum sodium concentration did not change. Elevation of blood pressure by norepinephrine was not followed by any detectable change of plasma AVP levels, thus excluding a nonspecific blood pressure effect.

    Notes: Abstract From Sedum morganianum, which is a plant species known to have constitutive crassulacean acid metabolism (CAM), phosphoenolpyruvate (PEP) carboxylase (E.C.4.1.1.31) has been extracted and purified by (NH4)2SC4 precipitation, ion exchange chromatography and gel electrophoresis. A specific antibody to this purified enzyme was obtained by immunization of a rabbit. This antibody was used to compare the antigen–antibody reaction of PEP-carboxylases prepared from other Sedum species including constitutive, facultative and non-CAM plants. The experiments revealed partial immunological indentity of PEP-carboxylases obtained from the different sources.

    Notes: Summary Myosin and actin were localized in the adrenal gland, using antibodies against these proteins which were isolated from chicken gizzard. Myosin and actin were preferentially located in vascular walls including endothelial cells and in the capsule. In rat and guinea-pig adrenal cortex, the amount of contractile elements in vascular walls corresponded well to the density of adrenergic nerves as revealed with the glyoxylic acid method.

    Notes: Summary Antibodies against actin and myosin from smooth muscle, which may react with contractile elements from both muscular and muscle-like cells, were applied to fresh frozen sections of adult guinea-pig testis. Sections stained with an antibody against pectoralis (striated) muscle myosin or with non-immune globulin were used for controls. Peritubular cells of the lamina propria surrounding seminiferous tubulus contained large amounts of actin and myosin as judged by the intensity of immunofluorescence. Sertoli cells did not stain with the antibodies. Our results support the concept of peritubular cells being the critical force for the contractility of seminiferous tubules.

    Notes: Summary Recent immunohistochemical studies have shown that basal cells in human prostatic epithelium are not myoepithelial cells. Since in the literature the Dunning tumor, originally described as a rat prostate carcinoma derived from the dorsolateral prostate of a Copenhagen rat, was reported to have myoepithelial cells, a comparative immunohistochemical and ultrastructural study was performed in the H-, HIF- and AT3-lines of the Dunning tumor, the male accessory sex glands (ventral, dorsal, lateral prostate, coagulating gland, bulbourethral gland) and the mammary gland of both Copenhagen and Wistar rats. Mono- and polyclonal antibodies directed against intermediate filament proteins (cytokeratin, desmin, vimentin) and the contractile proteins (α-actin, muscle type specific myosin, tropomyosin) were used along with phalloidin decoration of F-actin. As in the human prostate, none of the rat prostate lobes in either strain did contain basal cells expressing cytokeratin along with α-actin, myosin and tropomyosin. Cells representing fully differentiated myoepithelial cells, however, were present as anticipated in the mammary gland, the bulbourethral gland and the H-tumor line of the Dunning tumor. This finding is difficult to reconcile with the contention of a prostatic origin of the H-Dunning tumor. Further studies are required to classify the epithelial parental tissue in order to define the true origin of the H-Dunning tumor and the tumor lines derived thereof.

    Notes: Summary Although some embryological and morphological features speak in favour of a neuronal character of rodent pinealocytes, histochemistry and ultrastructure let this issue appear controversial. Using antibodies to different neurofilaments, the neural adhesion molecule L1, synaptophysin and tubulin as neuronal markers, the pineal glands of rat and guinea-pig were studied by means of immunfluorescence. Neurofilament-immunoreactivity was present in some rat pineal nerve fibers and in the majority of guinea-pig pinealocytes, L1 decorated rat intrapineal nerve fibers, synaptophysin was almost ubiquitously distributed in the pineal of both species, while tubulin-immunofluorescence was seen in nerve fibers of rat and guinea-pig pineal and in some pinealocytes of the latter. These findings speak in favour of the neuronal character of guinea-pig pinealocytes. The lack of neurofilament- and tubulin-immunoreactivity in rat pinealocytes might be attributable to very low concentrations of these proteins or species differences as to their expression. Further studies including in situhybridisation of relevant mRNAs will be necessary to answer these questions definitely.

    Notes: Summary Actin-and myosin-like immunoreactivity is found in cells located in the theca externa of the follicle wall of the human ovary, and corresponding to previously observed myoid cells. The immunocytochemical observation provides direct structural evidence that non-vascular contractile cells are also present in the follicle wall in humans. As expected, perifollicular blood vessels showed a positive immunoreaction for actin and myosin in their smooth muscle walls.
