    Notes: Summary A series of experiments was conducted in the urethane anaesthetised rat to determine the organisation of some hippocampal pathways in this species, using stimulating and recording microelectrodes to elicit and record population spikes. It was found that the mossy fibres, alvear fibres and perforant path were clearly arranged in a lamellar fashion. Lamellar organisation could not be demonstrated for the afferents in the stratum radiatum which include the Schaffer collaterals. It was concluded that hippocampal organisation in this species essentially resembles that in the rabbit and cat.

    Notes: Summary The effect of a single oral dose of 500 mg naproxen on the synthesis in vivo of thromboxane A2 and prostacyclin was studied in healthy volunteers. The synthesis of the prostanoids was assessed by measuring the urinary excretion of the metabolites 2,3-dinor-TxB2 and 2,3-dinor-6-keto-PGF1α, respectively, using stable isotope dilution assays based on gas chromatography — mass spectrometry. Naproxen caused significant inhibition of the excretion of both metabolites for about two days. The reduction of the thromboxane metabolite was more pronounced (75% inhibition) than that of the prostacyclin metabolite (about 50% inhibition). The data support the idea that naproxen causes reversible inhibition of cyclooxygenase.

    Notes: The electronically chemiluminescent reaction Mn+O3→MnO*+O2 was investigated using a beam-gas configuration. Light from the MnO A 6∑+–X 6∑+ transition was collected by a charge coupled device (CCD) array detector with resolutions of 0.5 and 0.1 nm. The spectrum at lower resolution (500–655 nm) encompassed the Δv=−3 to +2 sequences, while that at higher resolution (555.5–583.5 nm) encompassed only the Δv=0 sequence. These two spectra were separately fitted with a nonlinear least-squares program to obtain vibrational and rotational distributions of the nascent MnO*. The limited vibrational-state coverage of the higher-resolution spectrum made it unrealiable for determining the vibrational state distribution, and it was useful only for characterizing the rotational distribution when v′=0. The best-fit vibrational excitation is somewhat less than for the Prior model, but the rotational excitation is considerably greater. A consideration of the electronic structure of reactants and products indicates that principal changes occurring in the chemiluminescent reaction are σ–electron donation from the sdz2 hybridized Mn orbital to the O3 lowest unoccupied molecular orbital (LUMO) (2b1) and π–electron backdonation from the O–O 4b2 orbital to the Mn 3dπ orbital. Correlation of the orbitals involved indicates that direct access is allowed to the MnO A 6∑+(10σ*18σ1) state. This mechanism favors Mn approach perpendicular to the O3 plane and suggests that the product's rotational excitation may originate in O2–OMn repulsion arising from removal of electron density from the slightly bonding 4b2 orbital of O3. However, some rotational excitation could also be attributed to conservation of angular momentum arising from a sizable reactive impact parameter. The lack of significant vibrational excitation is a consequence of the short-range nature of the partial charge transfer in this reaction channel. © 2000 American Institute of Physics.

    Notes: Sedative-hypnotic medications are commonly abused prescription drugs, which are commonly prescribed by psychiatrists. We aimed at studying benzodiazepine and related drug prescriptions among patients attending psychiatric out-patient clinics of a hospital in East Lancashire. A total of 6.2 per cent of patients who attended the department in one representative week were prescribed benzodiazepine/related drugs. A majority of these had anxiety disorders, and anxiety was the major indication for prescription of these drugs. Diazepam, temazepam and related drugs like zopiclone/zolpidem were the most commonly prescribed. Most subjects had been on the drug for one to 12 months. The prescriptions were not in keeping with the BNF guidelines in most cases. The findings do not support the hypothesis of an unduly high prevalence of sedative prescriptions in this setting. The results highlight a need for complete documentation of various aspects of benzodiazepine prescriptions.

    Notes: Members of the plakin family of cytolinker proteins integrate filaments into cellula networks and anchor these networks to the plasma membrane. Their importance is supported by the existence of cell and tissue fragility disorders caused by mutations in certain family members. In this study, the human gene encoding desmoplakin (DSP) was characterized and its structure compared with the related family members: plectin, bullous pemphigoid antigen 1 (BPAG1), envoplakin (EVPL) and periplakin (PPL). Sequence analysis of genomic clones was carried out in combination with a PCR-based strategy to define intron-exon border. DSP was mapped using the GB4 radiation hybrid mapping panel to the interval between markers D6S296 and AFM043Xf2, correponding to cytogenetic band 6p24. In addition, the murine gene (DSP) was mapped to mouse chromosome 13 by interspecific backcross mapping. DSP encompasses 45 kb organized into 24 exons and 23 introns, and the pattern of intron-exon borders bears a striking resemblance to other member of the plakin family. Notable features include the fact that a single large exon encodes the entire C-termius of each gene. In contrast, the N-termini comprise numerous smaller exons with conservation of many genses will facilitate their further evaluation as targets of genetic disorders and provide insights into the evolutionary relationships among molecules in this emerging gene family.
