    Description / Table of Contents: Zusammenfassung Das immunbiologische Verhalten von Lymphocyten sensibilisierter Patienten eröffnet die Möglichkeit zu einem neuartigen „in vitro”-Allergie-Test. In Ergänzung zu früheren Mitteilungen über die Versuchsanordnung und den Ansatz sowie über erste Ergebnisse von Lymphocytenkulturen wird an dieser Stelle über eine Weiterentwicklung der Methodik berichtet. Die fluorescenzmikroskopische Untersuchung mit Acridinorange und die Beurteilung supravital gefärbter Zellen stellt nicht nur eine wesentliche Vereinfachung dar, sondern sie bietet auch den Vorteil lebensnaher histochemischer Differenzierungsmöglichkeit.
    Notes: Summary Lymphocytes from sensitized individuals acquires the capacity in vitro to differentiate into larger blast-like cells with nucleoli capable of mitotic cell division when cultured in presence of the specific antigen. It is the purpose of this report in addition to previous papers to describe a simplified fluorescence microscopy method for studying the immune response of lymphocyte cultures with supravital acridine orange fluorochroming. This fluorescence studies in transformation of antigen-stimulated cultured lymphocytes with acridine-orange makes it possible to use it for differentiating morphological and histochemical particularities.

    Description / Table of Contents: Zusammenfassung Das Ansteigen des Histaminspiegels im Organismus nach Einwirkung ionisierender Strahlen ist bekannt. Bei “in vitro” Applikation von Röntgenstrahlen im Dosisbereich von 50 r–8000 r auf Histamin in wäßriger Lösung fand sich ein signifikanter Aktivitätsverlust dieses biogenen Amins. Mit Hilfe einer sehr empfindlichen biologischen Versuchsanordnung am isolierten überlebenden Meerschweinchendarm gelang es, beträchtliche Verluste an biologischer Aktivität (Kontraktionsvermögen auf die glatte Muskulatur) als Folgereaktion auf die Bestrahlung bis zu einem Dosisbereich von 50 r–100 r nachzuweisen.
    Notes: Summary The widespread existance of histamin in living organisms is well known. The increase of its activity after exposure of these subjects to ionizing radiation has been described by many investigators. However, we found a significant decrease in the activity of histamin after “in vitro” irradiation of these substance (solved in water) with 50 r–8000 r. The investigations, which demonstrated changes in activity, were carried out with the isolated ileum, of guinea-pig. Our observations revealed the radiation-induced effect following minimal dose of 50r–100 r.
